Effect of batroxobin on expression of neural cell adhesion molecule in temporal infarction rats and spatial learning and memory disorder.

The effect of Batroxobin expression of neural cell adhesion molecule (NCAM) in left temporal ischemic rats with spatial memory disorder was investigated by means of Morri's water maze and immunohistochemical methods. The results showed that the mean reaction time and distance of temporal ischemic rats for searching a goal were significantly longer than those of sham-operated rats and at the same time NCAM expression of left temporal ischemic region was significantly increased. However, the mean reaction time and distance of Batroxobin-treated rats were shorter and they used normal strategies more often and earlier than those of ischemic rats. The number of NCAM immune reactive cells of Batroxobin-treated rats was more than that of ischemic group. In conclusion, Batroxobin can improve spatial memory disorder of temporal ischemic rats and the regulation of the expression of NCAM is probably related to the neuroprotective mechanism.

[Herpesviridae and laryngeal neoplasia].

OBJECTIVE: To investigate the relationship between herpesviridae and malignant or benign laryngeal diseases. METHOD: 128 paraffin-embedded laryngeal squamous cell carcinoma and laryngeal epithelium hyperplastic lesions were detected by polymerase chain reaction (PCR) and PCR-ISH for herpesviridae. RESULT: HSV-1 was detected in 10 cases by PCR, among them 3 were laryngeal squamous cell carcinoma (LSCC), 1 was carcinoma in situ(CIS), 4 were laryngeal polyps and 2 were laryngeal keratosis. Except 1 LSCC and 1 CIS 8 of 10 cases were positive while detected by PCR-ISH. In benign diseases, signals were shown from basal layer to superficial cell; in malignant lesions, the signals were scattered in the diseases. CONCLUSION: Most of laryngeal diseases were not related to herpesviridae, but HSV-1 may acts as initiator in the development of a few cases.

[Isolation of outer hair cells from varying turns of the guinea-pig cochlea].

OBJECTIVE: To probe the method of isolating outer hair cells (OHC) from each of four turns of the guinea-pig cochlea. METHOD: From eight guinea pigs the organ of Corti from each of four turns of the cochlea were dissected, and then treated using enzyme. RESULT: A fair amount of living OHCs from each of four turns were obtained. The length of OHCs from each of four turns were 23.81, 34.50, 60.48 and 71.37 microns. CONCLUSION: The key to success in isolating OHCs from each of four turns of the cochlea is to know very well the anatomical characteristics of each of four turns of the cochlea and be operated in accordance with normal rules.

[Expression and the role of vascular endothelial growth factor mRNA in nasal polyps].

OBJECTIVE: To explore the pathogenesis of nasal polyps, the aim of this study is to detect the expression of vascular endothelial growth factor (VEGF) in nasal polyp(NP) tissue, and compare these findings with normal nasal turbinate. METHOD: We examined the expression of VEGF in human nasal polyps from patients underwent nasal polypectomy (n = 7) and nasal turbinate from patients with snoring (n = 6). VEGF mRNA expression was determined by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). RESULT: VEGF mRNA expression was significantly stronger in the eight NP than in the turbinate tissue group (P < 0.05). CONCLUSION: We conclude that VEGF expression is upregulated in NP, which suggests a potential role of VEGF in the formation and growth and recrudescence of NP.

[Studies on transference of hydrogenase genes of Rhizobium arachis].

The hydrogen-uptake genes were transferred into wild Rhizobium arachis Ra strains (Hup-, Nif+, Apr) by triparental mating using pRK2013 as help plasmid. A transconjugant R. arachis Rz34-2(Hup+, Nif+, Apr, Tcr) which expressed high activities of hydrogenase and nitrogenase under free-living and symbiotic state was screened. Peanut inoculation test with recipient R. arachis Ra34, transcojugant Rz34-2 and control strain R. arachis L8-3 (Hup+, Nif+) was carried out respectively. The results showed that, compare to treatment without inoculation, inoculation with R. arachis Ra34 and R. arachis L8-3, the dry weight of leaf inoculated with transconjuant Rz34-2 increased 6.2%, 7.6% and 6.3% respectively; the N-content of seed increased 8.8%, 10.0% and 6.0%; the output increased 18.8%, 10.5% and 10.7%. This suggested that legume plants inoculated with Rhizobium strains (Hup+) were more efficient to accumulate N and to increase its output.

[Study on the genotoxicity of dibromoacetic acid in drinking water].

Dibroacetic acid (DBA) is a byproduct of drinking water disinfected by halogenation and it belongs to halogenated acetic acids. It also can be found in ozonated drinking water. Four short-term assays were performed to detect the genotoxic activity of DBA. In the Ames test, DBA showed mutagenic activity in Salmonella typhimurium strain TA100 with or without rat liver-derived metabolic activation system (S9). In UDS test, DBA was found to induce DNA damage in rat primary hepatocyte. Mouse bone marrow micronucleus test indicated that DBA induced the damage of chromosome. In NIH3T3cell micronucleus test, DBA increased the number of micronuclei in cells. Our results suggest that DBA induce DNA damage and has genotoxic activity.

[Effects of acute infrasound exposure on vestibular and auditory functions and the ultrastructural changes of inner ear in the guinea pig].

OBJECTIVE: To define the effects of acute infrasound exposure on vestibular and auditory functions and the ultrastructural changes of inner ear in guinea pigs. METHODS: The animals involved in the study were exposed to 8 Hz infrasound at 135dB SPL for 90 minutes in a reverberant chamber. The sinusoidal pendular test (SPT), auditory brainstem response (ABR) and distortion product otoacoustic emissions (DPOAE) were respectively detected pre-exposure and at 0(within 2 hrs), 2 and 5 day after exposure. The ultrastructures of the inner ear were observed by scanning electron microscopy. RESULTS: The slow-phase velocity and the frequency of the vestibular nystagmus elicited by sinusoidal pendular test (SPT) declined slightly following infrasound exposure, but the changes were not significant (P > 0.05). No differences in the ABR thresholds, the latencies and the interval peak latencies of I, III, V waves were found between the normal and the experimental groups, and among experimental groups. The amplitudes of DPOAE at any frequency declined remarkably in all experimental groups. The ultrastructures of the inner ear were damaged to different extent. CONCLUSION: Infrasound could transiently depress the excitability of the vestibular end-organs, decrease the function of OHC in the organ of Corti and cause damage to the inner ear of guinea pigs.

[Expression and distribution of Aquaporin 1 in the nasal polyps].

OBJECTIVE: To confirm the expression and distribution of Aquaporin 1 (AQP1) in the nasal polyps and to investigate the relation between AQP1 and the nasal polyp edema. METHODS: fourteen cases of normal inferior turbinates and 26 cases of nasal polyps were used. The expression and distribution of AQP1 in nasal polyps were examined by immunohistochemical technique. RESULTS: The level of AQP1 in the epithelial cells and serous cells from nasal polyps was higher than that in inferior turbinates, and the level of AQP1 in the epithelial cells and cilium cells from inferior turbinate was higher than that in nasal polyps. CONCLUSION: There is close relationship between AQP1 and nasal polyp edema.

[Synergetic protective effects of glial cell line-derived neurotrophic factor combined with neurotrophin-3 in F-actin on hair cell after noise trauma].

OBJECTIVE: To investigate if glial cell line-derived neurotrophic factor (GDNF) combined with neurotrophin-3 (NT-3) provides synergetic protection in filamentous actin (F-actin) on hair cell (HC) from acoustic trauma. METHODS: Guinea pigs were exposed to 4 kHz narrow band noise at 115 dB SPL for 4 h. Test group (n = 12) with a mixture of GDNF (100 ng/ml) and NT-3(2.5 micrograms/ml) or control group (n = 9) with artificial perilymph (AP) was delivered to the scala tympani via a mini-osmotic pump (0.5 microliter/h) for a total of 14 days. Auditory function was assessed by measuring thresholds of auditory brainstem responses (ABRs) elicited by clicks prior to surgery, 3 days after surgery (1 day before noise exposure) and 10 days following noise exposure (before animals were sacrificed), respectively. F-actin, labeled by rhodamine-phalloidin, was examined in the guinea pig cochlea using fluorescence microscopy for quantitative assessment of hair cell damage. RESULTS: There was a statistically significant increase the survival of out hair cell(P < 0.001, P < 0.01) and inner hair cell(P < 0.01, P < 0.01) and decrease in ABR threshold (P < 0.05, P < 0.01) in both the GDNF and NT-3 treated and untreated ear of animals. CONCLUSION: Our findings indicate that GDNF combined with NT-3 may effectively protect the inner ear from noise--induced hearing loss.

[Glutamate induced modulation of free Ca2+ in isolated inner hair cells of the guinea pig cochlea].

OBJECTIVE: To investigate the modulation effect of glutamate (Glu) on intracellular free Ca2+ concentration([Ca2+]i) of inner hair cell (IHC). METHODS: Using the laser scanning confocal microscope (LSCM), the exogenous Glu-induced changes in [Ca2+]i of isolated 10 IHCs and 10 OHCs of guinea pig cochlea were observed with fluo-3, a fluorescent probe for [Ca2+]i. RESULTS: The IHCs were identified by their unique flask shape with a distinct neck and spherical base and a large spherical nucleus. In most cases, normal cell shapes could be maintained about two hours after isolation. The images of [Ca2+]i from LSCM were similar to those from inverted microscopy. Fluorescence of Fluo-3 distributed in the isolated IHCs with brighter staining in the nucleus. In the presence of low concentration of Glu (3.85 mumol/L), there was an increase of [Ca2+]i in IHCs, whereas no change in OHCs was found. Of the 10 IHCs, increases of [Ca2+]i were observed in 9 and no change in 1. Of the 10 OHCs, 7 showed no [Ca2+]i change and only 3 showed minor reduction of [Ca2+]i. An increase of the Glu concentration (21.88 mumol/L) induced a corresponding increase of [Ca2+]i in IHCs, but eventually resulted in a gradual decrease of [Ca2+]i with a distortion of the normal shape, which indicated that the IHCs were degenerated and swelling. CONCLUSION: These results suggested that exogenous Glu is capable of modulating [Ca2+]i of IHC and may be act on autoreceptor in a positive feedback manner. Excessive Glu induced the accumulation of IHC [Ca2+]i which finally resulted in the degeneration and edema of IHC and the reduction of IHC [Ca2+]i.

[The effects of streptomycin on the calcium influx in SGCs during depolarization].

OBJECTIVE: To examine the effect of streptomycin on the calcium influx in spiral ganglion cells (SGCs) during depolarization and the differences among different concentrations of streptomycin and SGCs sources with the aim of exploring the mechanism of acute streptomycin ototoxicity. METHODS: The SGCs of guinea pig cochlea were isolated using an enzyme-machine method and loaded with 10 mumol/L Fluo-3/AM for 30 min at 37 degrees C. Dissociated SGCs loaded with Fluo-3 were examined with a confocal microscope (ACAS Ultima, USA) using a 20 x objective lens and linear scan mean. The fluorescent images collected every 5-sec for a total of 300 sec were stored in a computer. The fluorescent intensity of the images was analyzed by a software cooperated with the confocal microscope, and a curve of fluorescent intensity changes against time was obtained. RESULTS: The [Ca2+]i of SGCs was steady under the perfusion with standard extracellular solution. The [Ca2+]i of SGCs increased in 12/14 cells under the perfusion with 150 mmol/L high potassium solution, but increased in only 1/10 cells induced by the high potassium calcium free solution. After treating with 1 mmol/L, 0.1 mmol/L, 0.01 mmol/L streptomycin, the [Ca2+]i of the SGCs perfused with the potassium solution was increased in 0/9, 6/14, 7/13 cells, respectively. After the SGCs were treatment with 0.02 mmol/L streptomycin, the [Ca2+]i of the SGCs perfused with the high potassium solution was increased in 5/10 SGCs from the apical turn and in 0/15 SGCs from the basal turn. CONCLUSION: Perfusion with high potassium media can result in obviously increased [Ca2+]i of the SGCs, and the increase of the [Ca2+]i in the SGCs may originate from the extracellular calcium influx. The calcium influx can be blocked by streptomycin and such effects depend on streptomycin concentrations and SGCs sources.

[Experimental study on polyaspartic acid inhibition of gentamicin-induced generation of reactive oxygen species in guinea pig cochlea].

OBJECTIVE: To observe the polyaspartic acid(PAA) inhibition of gentamicin-induced reactive oxygen species generation in cochlea of guinea pig and to investigate the protective mechanism of polyaspartic acid on gentamicin ototoxicity. METHODS: Eighty-eight guinea pigs were divided randomly into four groups (GM, PAA + GM, PAA, and Saline). Gentamicin-induced reactive oxygen species (ROS) formation in cochlear tissue was detected directly with electron paramagnetic resonance (EPR) spectrometry at 1st, 5th and 10th day after administration of the drugs. At the same time, ABRs of guinea pigs were recorded and ultrastructural changes of lysosomes in the cochlear hair cells were observed with transmission electron microscopy. RESULTS: 1. At 1st day after administration of PAA and GM, there was some increase in EPR spectrometry in group GM and PAA + GM, There was no significant difference of ABR thresholds and ultrastructural changes of lysosomes in the cochlear hair cells among four groups(P > 0.05). 2. At 5th day, there was significant increase in of EPR spectrometry in group I (37.74 +/- 4.10, P < 0.01). At 10th day after administration of PAA and GM, there was no significant difference in EPR spectrometry among four groups (P > 0.05). In GM group, ultrastructural changes of lysosomes beneath cuticular plate of cochlear hair cells were more significant at 10th day than those at 5th day, including the increased number and volume of lysosome. In group GM, the longer the gentamicin administrated, the more significant increase in ABR thresholds had been noted. CONCLUSION: PAA significantly inhibits gentamicin-induced reactive oxygen species generation in cochlea of guinea pig, which showed that PAA has protective effect on gentamicin-ototoxicity and -phospholipidosis in guinea pigs.

[Expression of the inducible isoform of nitric oxide synthase mRNA and the role in nasal polyps].

OBJECTIVE: To explore the pathogenesis of nasal polyps. The aim of this study is to detect the expression of inducible isoform of nitric oxide synthase (iNOS) in nasal polyp tissue, and to compare these findings with that in normal nasal turbinate. CONCLUSIONS: iNOS expression is upregulated in NP, which indicate that iNOS and NO may play a potential role in the formation and growth of NP.

[Two-photon fluorescence from recombinant green fluorescent protein].

The photoconversion process of recombinant green fluorescent protein (rGFP) was investigated by two-photon excitation. The results indicated that the rGFP had very strong two-photon excitation fluorescence. The changes of two-photon-induced fluorescence polarization suggest that there is a proton transfer process between two different proton states of rGFP chromophores. The conformation of rGFP chromophores could be changed upon illumination, which partly block the energy transfer processes from amino acid residues to chromophores in rGFP, and result in the decrease of two-photon-induced fluorescence intensity. The fluorescence from amino acid residues in rGFP was also observed by three-photon excitation, which resulted from the blocked amino acid residues in rGFP. These results suggested that it is necessary to optimize rGFP excitation and detection for quantitative fluorescence microscopy.

General and efficient syntheses of C(18)-4,8-sphingadienines via S(N)2'-type homoallylic coupling reactions mediated by thioether-stabilized copper reagents.

The stereoselective syntheses of C(18)-4,8-sphingadienines 3 and 4 as analogues of sphingosine 1 are described. The key step in these syntheses involved a novel S(N)2'-type homoallylic coupling reaction between the corresponding thioether-stabilized allylic copper reagents and the allylic mesylate 7. The thioether-stabilized allylic copper reagents were easily prepared and retained the configuration of their double bond during the coupling reactions, thus overcoming the problem of isomerization which was normally associated with the use of allylic organometallic reagents in such applications.

Free radical allyl transfers utilizing soluble non-cross-linked polystyrene and carbohydrate scaffold supports

Free radical allylations were studied using (1) soluble non-cross-linked polystyrene supports, (2) carbohydrate scaffolds, and (3) a combination of both synthetic motifs. Allylations on these custom designer supports provide easily purified products, free of tin residues. A D-xylose carbohydrate scaffold bearing a bromoester was used for a diastereoselective allyl tin transfer thermally at 80 degrees C and with Lewis acids. This is the first example of a diastereoselective radical reaction directed by a removable polymer-supported carbohydrate auxiliary.

Solution structure of BmP01 from the venom of scorpion Buthus martensii Karsch.

From the venom of scorpion Buthus martensii Karsch,a short peptide (BmP01, 29 amino acid residues) was isolated and characterized as previously reported (Lebren, R. R., et al. (1997) Eur. J. Biochem. 245, 457-464). It was shown to reduce 33% outward K(+) channel (hippocampal neurons) currents at 10 microM. The solution structure of BmP01 was determined by 2D (1)H NMR spectroscopy. The NOEs, coupling constants, and H-D exchange obtained from NMR spectroscopy were used in structural calculations. The conformation of BmP01 is composed of a short alpha-helix (Cys 3-Thr 12) and a two-stranded antiparallel beta-sheet (Ala 15-Asp 20 and Lys 23-Pro 28). There are three disulfide bridges (Cys 3-Cys 19, Cys 6-Cys 24 and Cys 10-Cys 26) connecting the alpha-helix and beta-sheet. Asp 20 to Lys 23 form a type II turn linking the two strands. Structural and electrostatic potential comparison between BmP01 and its analogues are also presented.

Cocaine injection and coxsackievirus B3 infection increase heart disease during murine AIDS.

Coxsackievirus initiates myocarditis especially in the immunologically deficient or immature. To test whether Coxsackievirus B3 (CVB3) induced pronounced cardiomyopathy during severe immune dysfunction of murine AIDS, female C57BL/6 mice were infected with LP-BM5 retrovirus and superinfected with CVB3. Some were also injected daily with cocaine hydrochloride in 0.9% saline solution (30 mg/kg) intraperitoneally, because cocaine also suppresses cellular immune response. Heart tissue was analyzed histopathologically. Mice experiencing concurrent retrovirus and Coxsackievirus infection had a high degree of cardiac lesions consistent with myopathy compared with findings in uninfected animals (p <.05). Cocaine injection during murine retrovirus infection greatly exacerbated the pathogenesis of Coxsackievirus infection. C57BL/6 mice, essentially resistant to Coxsackievirus-induced cardiomyopathy, became susceptible during the immune dysfunction in murine AIDS. This suggests that retrovirus infection causes conditions favoring Coxsackie-induced cardiac lesions. Interleukin (IL)-2 and IL-4 expression by splenocytes from the dually infected retrovirus and Coxsackievirus group showed no significant differences when the animals were also cocaine treated. However tumor necrosis factor TNF-alpha production was significantly decreased in dually infected retrovirus + Coxsackievirus mice treated with cocaine, compared with findings in various controls (p <.05).
