Amidophosphoribosyltransferase

Cleavage capability of water buffalo follicular oocytes classified by cumulus cells and fertilized in vitro.

Water buffalo (Murrah) oocytes were collected from ovaries obtained from the slaughter house. They were classified according to the character of the cumulus cells under a stereomicroscope, and cultured in 25 mM Hepes buffered Tissue Culture Medium-199 (TCM-199) supplemented with 5% estrous water buffalo serum in an atmosphere containing 5% CO2 in air at 39 degrees C. After 20-24 hr of in vitro maturation, the oocytes were fertilized using capacitated sperm obtained from 4 different bulls. For cleavage the oocytes were cultured at 39 degrees C in TCM-199 supplemented with 1% estrous water buffalo serum and in an atmosphere containing 5% CO2 in air. The good oocytes, with compact and dense cumulus cells cleaved significantly higher (p less than 0.01, 67.3%), than those of fair. partially naked oocytes with thin cumulus layers (27.5%, 25/91) or small remnants of cumulus cells and poor naked oocytes (3/100). A substantial variation in fertilization and developmental rates (16.0% to 43.8%) was observed among 4 different bulls. Late non-surgically into 14 buffalo recipients on day 6 or 7 of their estrous cycle. One recipient was diagnosed to be pregnant by rectal palpation on day 60 and confirmed to be so on day 90 post-estrus.

Acute intussusception: referral in relation to presentation and aetiology.

A study of 42 consecutive children with acute intussusception was undertaken. The emphasis was on the time taken for surgical referral in relation to the presenting symptom. Operative findings and outcome revealed late referral (greater than 72 h) in cases presenting with diarrhoea and bleeding per rectum. A higher incidence of gangrenous gut and irreducibility of intussusception was found in the ileoileal variety of intussusception. Some interesting facts about the aetiology have also been noted. Rupture of intussuscipiens was also encountered.

Development of a simple, direct, microtitre plate enzymeimmunoassay (EIA) for progesterone determination in whole milk of buffaloes.

A method of estimating progesterone in buffalo whole milk by EIA using progesterone 6 beta-OH-hemisuccinate-horseradish peroxidase as the enzyme label and an antiserum raised against progesterone-7 alpha-carboxyethyl-thioether-BSA was developed. The microtitration plates used in the assay were first coated with affinity purified sheep IgG developed against rabbit IgG. The immune reaction was performed by incubating a mixture of 1 microliter of whole milk (diluted to 20 microliters with assay buffer), 100 microliters of enzyme label and 100 microliters of antiserum for 90 min in the dark. After washing the plates, 150 microliters of the substrate solution was added. The mixture was incubated in the dark for 40 min before the reaction was stopped and the optical density was measured at 450 nm. The calibration curve was sensitive in the range 0.8-40 pg/well, corresponding to 0.8-40 ng/ml. Milk samples from cycling buffaloes were tested for progesterone concentration by running parallel EIA and RIA. A good correlation of 0.91 was obtained and the estimated values were similar using both techniques. The method has demonstrated about 10 times greater sensitivity than RIA in buffalo milk.
