Postmortem changes in hapten-specific IgE antibody responses in mice.

This Study was carried out to examine the post-mortem stability of hapten-specific immunoglobulin E (IgE) antibody in mice by the passive cutaneous anaphylaxis test in rats. In vitro IgE antibody responses in blood were stable for 3 days at room temperature. Even in cases when blood could not be obtained from the cadaver it was possible to measure the hapten-specific IgE antibody response up to 3 days after death using the supernatant of the visceral homogenates.

Identification of human hair stained with oxidation hair dyes by gas chromatographic-mass spectrometric analysis.

This paper describes the gas chromatographic-mass spectrometric (GCMS) analysis of oxidation hair dyes from human hair. Diamines from the dyes were directly extracted from the hair in basic solution and aminophenols were extracted after neutralization. Both extracts were derivatised with trifluoroacetic anhydride and analysed by GCMS. Five components of oxidation hair dyes namely, p-phenylenediamine, toluene-2,5-diamine, o-aminophenol, m-aminophenol and p-aminophenol were clearly identified, whilst no other compounds originating from the hair dyes were detected. The presence and relative amounts of these dye components from hair extracts may assist in the discrimination of human hair especially in cases involving forensic science.

A simple, rapid and simultaneous analysis of complex volatile hydrocarbon mixtures in blood using gas chromatography/mass spectrometry with a wide-bore capillary column.

A screening method for detecting volatile hydrocarbons in blood has been developed using gas chromatography/mass spectrometry with a wide-bore capillary column and a headspace method. Toluene-d8 and indan were used as the internal standards for quantitative analysis. Hydrocarbons with retention indices from 600 to 1200 were simultaneously and quantitatively detected in relatively low concentrations (0.01 microgram/ml) in reconstructed ion chromatography. This method could prove useful in forensic cases in which urgent examination of complex hydrocarbon mixtures, e.g. petroleum components, is required.

Cause of Death↗

An experimental model of death from anaphylactic shock with compound 48/80 and postmortem changes in levels of histamine in blood.

The present study was made on an experimental animal model of a death from anaphylaxis, in which postmortem changes in levels of histamine and 1-methylhistamine, in whole blood were measured. Instead of the usual immunological method administering compound 48/80, a degranulating agent of mast cell and the effect closely resembling the immuno-reaction, resulted in reliable death in a short time. The animals that died rapidly after the injection of compound 48/80, were found to have large increases in levels of histamine and 1-methylhistamine soon after the administration. These results were similar to the results of injecting histamine exogenously. On the other hand, the animals that died after a longer time showed no increases in levels of those amines within about 24 h, but 24 hours after death histamine levels were only increased tremendously without rise in 1-MHA levels. These phenomena closely resembled those in the control animals that were treated with overdoses of Nembutal.

Genetic variation of alpha-2-HS-glycoprotein in the Kyushu district of Japan: description of three new rare variants.

The genetic polymorphism of alpha 2-HS-glycoprotein (AHSG) was studied in the Kyushu district of Japan using polyacrylamide gel isoelectric focusing, followed by immunoblotting. Three new rare variants were observed and designated AHSG*16, AHSG*17 and AHSG*18, tentatively. The frequencies of the polymorphic genes AHSG*1 and AHSG*2 were similar to those in other areas of Japan.

[Screening of drugs and chemicals by wide-bore capillary gas chromatography. II. Detection of drugs and chemicals in the blood].

This paper describes the detection limit for 23 drugs and chemicals in the blood by means of a screening method that uses a gas chromatographic system equipped with a wide-bore capillary column and a nitrogen phosphorus detector. The detection limit by this method was determined as being 1 mm of peak height at the detector's range of 100 and 8 of attenuation. Using this scale, the absolute detection limit was in the range of 1 pg for malathion and sumithion to 1 ng for meprobamate. The detection limit of drugs and chemicals in the blood was 5 ng/ml for sumithion to 8 micrograms/ml for meprobamate. Therefore, this screening method is able to detect the presence of drugs even a therapeutic-level dosages, with the exception of compounds such as haloperidol, which have extremely low therapeutic dosage levels.

[Screening of drugs and chemicals by wide-bore capillary gas chromatography with flame ionization and nitrogen phosphorus detectors].

A method is presented for forensic toxicological screening of drugs and chemicals in blood and urine by wide-bore capillary gas chromatography with flame ionization detectors (FID) and nitrogen phosphorus detectors (NPD). The presence of drugs and chemicals in blood and urine specimens was confirmed by comparing these gas chromatograms with those of typical drug-free specimens. Peak components of drug-free specimens were piperidone, p-cresol, myristic acid, palmitic acid, stearic acid, oleic acid, linoleic acid, n-butylphthalate, bis (2-ethylhexyl) phthalate, squalene, cholesterol and two alcohols (unidentified) on FID chromatograms and were piperidone, indole, nicotine, cotinine, hydroxycotinine, caffeine and several unknown urine constituents on NPD chromatograms. In practical cases, the presence of drugs and chemicals in postmortem specimens was easily ascertained by the present method.

Simultaneous quantitative analysis of methamphetamine and 4-hydroxymethamphetamine in body fluids by gas chromatography/mass spectrometry.

Pentodeuterated 4-hydroxymethamphetamine (HMAMP-d5), 1-(4-hydroxyphenyl)-2-(methyl-d3-amino)-propane-1,2-d2 of high quality was prepared and proved to be a most useful internal standard for quantitative analyses of 4-hydroxymethamphetamine (HMAMP), a main metabolite of methamphetamine (MAMP). Highly reliable results were obtained by gas chromatography/mass spectrometry. Simultaneously we determined also MAMP, amphetamine (AMP), 4-hydroxyamphetamine (HAMP), and HMAMP in body fluids, using two internal standards, HMAMP-d5 for HMAMP and HAMP and pentodeuterated methamphetamine, 1-phenyl-2-(methyl-d3-amino)-propane-1,2-d2, for MAMP and AMP.

[The course of respiration in death caused by obstructive asphyxia].

Experiments were carried out on 30 dogs to investigate the course of respiration during obstructive asphyxia. They were asphyxiated by occlusion of the exposed trachea. Intrathoracic pressure, blood pressure in the femoral artery, electrocardiogram and electroencephalogram readings were registered. In most cases, the respiration course during the dyspnea stage by occlusion of the trachea generally had an inspiratory character. In a few animals, when the trachea was occluded in the expiratory phase, there were a few positive expiratory respirations with moderate inspiratory movement and thereafter the respirations were inspiratory during the dyspnea stage. The terminal respirations, which occurred 1-2 min before the circulatory breakdown, were generally inspiratory, but in a few dogs they were expiratory.

Swimming and loss of consciousness.

Under certain circumstances, even a good swimmer may drown during swimming exercise. Two cases of drowning, a survivor and a dead, during swimming exercise in swimming-pool are described. These cases and experimental researches with dogs indicate that the initial aspiration of water may cause extremely low heart rate and low blood pressure by reflex vagal inhibition, which deprive a good swimmer of his consciousness and make him drown.

Orosomucoid (ORM) typing by print lectinofixation: a new technique for isoelectric focusing. Two common alleles in Japan.

The genetic types of orosomucoid (ORM) were analyzed by isoelectric focusing (IEF) on polyacrylamide gels and subsequent print lectinofixation with a lectin from the beetle, allo A. In this paper, the newly devised print lectinofixation for ORM typing is described. This technique is faster, easier to perform, and has been found to be a useful tool in population genetics and forensic medicine. The results of typing for two alleles, ORM 1 and ORM 2 are described for a population of Northern Japan (n = 500).

Affinity chromatography of human serum proteins using immobilized lectin from Allomyrina dichotoma.

The beta-D-galactoside-specific lectin from Allomyrina dichotoma reacts with serum proteins which contain the corresponding carbohydrate moieties. By affinity chromatography of human serum using the insolubilized lectin coupled to Sepharose, it is possible to fractionate human serum proteins in 2 groups: those which react with the lectin (alpha 1-acid glycoprotein, haptoglobin, etc.) and those which do not (albumin, Gc-globulin, etc.). IgG is the only serum protein that can be found in both groups.
