One hundred fifty-eight hemophilia A, B, and von Willebrand disease (VWD) patients treated with clotting factor concentrates from a single manufacturer were tested for antibody to the human T-lymphotropic virus type III (HTLV-III). Antibody was detected in 63% and 40% of those with severe hemophilia A and B, respectively, 12% and 0% of those with mild hemophilia A and B, and two patients with recessive VWD. Forty-two antibody-positive and 20 antibody-negative patients were studied for clinical and laboratory features of infection. Eleven seropositive patients had clinical signs of infection including Pneumocystis carinii pneumonia, lymphadenopathy, splenomegaly or diarrhea; however, only one patient had developed acquired immune deficiency syndrome (AIDS), and only two had significant impairment of their performance status. Thirty-one patients remained totally asymptomatic. Eight patients had a history suggestive of acute HTLV-III infection. Thrombocytopenia was observed in 18% of seropositive patients, lymphopenia in 60%, depressed T-helper cells in 43%, reduced T-helper:T-suppressor ratios (TH:TS) in 33%, and elevated platelet-bound immunoglobulin in 53%. The antibody-negative group had normal T-helper cell levels (except one patient) and TH:TS ratios, and normal platelet immunoglobulin levels. Both groups demonstrated a significant elevation of immunoglobulin levels and a high prevalence of antinuclear factor and antismooth muscle antibodies. The mean level of IgG was significantly higher in the antibody-positive group. This study confirms the correlation between HTLV-III infection and reduced T-helper cells in hemophiliacs but demonstrates a low incidence of clinical symptomatology. There was evidence of polyclonal B-cell hyperactivity in the antibody-negative group as well as the seropositive group.
In 50 plasma samples flash frozen at -40 degrees C within 6 h and in 50 samples flash frozen within 18 h after phlebotomy, both coagulation factors II, V, VII, VIII, IX and X as well as AT III, APTT and Quick were measured. Only factor VIII in 18-h plasma showed an average decrease from 0,99 IU/ml to 0,85 IU/ml, i. e. 15% compared to 6-h plasma. None of the other coagulation factors or parameters exhibited any change beyond normal ranges. Furthermore, 150 6-h and 150 18-h plasma samples were taken from routine production and after thawing were tested for factor VIII- and factor-V activity. In a long-term study following a one year's storage at -30 degrees C of 30 samples of 6-h plasma and 30 samples of 18-h plasma, a decrease was found with factor VIII from 0,97 IU/ml to 0,93 IU/ml in 6 h-plasma and from 0,90 IU/ml to 0,86 IU/ml in 18 h-plasma. Our results demonstrate that only minor differences exist in activity of coagulation factors of 6-h and 18-h plasma. Hence follows that by observing the instructions described, freezing must take place within an 18-h period after phlebotomy which thus allows 12 more hours for production without losses in activity.

No significant differences could be observed when the haptoglobine and group-specific component phenotypes of 51 unrelated, healthy carriers of antibodies to Hepatitis B surface antigen were compared with normal controls. These findings are contradictory to the suggestion of Prokop [2].
34 lightly fibromatous, 23 heavily fibromatous, 5 lipomatous and 10 naevus cell naevi were stained with Feulgen kit in order to evaluate their ploidy status with CAS 200 image analyzer. 26/34 lightly fibromatous, 18/23 heavily fibromatous, and 5/5 lipomatous naevi were either suspicious for aneuploidy (Auer III) or clearly aneuploid (Auer IV). In contrast all 10/10 naevus cell naevi were euploid. Proliferation (S-phase) was not increased in naevi fibromatously and lipomatously changed. The mechanisms leading to aneuploidy are discussed.
The relation between 2 DNA-repair systems was investigated in 3 groups of naevi (naevus cell naevi, dysplastic naevi, fibromatous naevi) using the correlation coefficient according to Spearman. In the group of naevus cell naevi, only 1 significant correlation between MSH2 and GADD34 expression was found. In the group of dysplastic naevi, 9 significant and highly significant correlations between GADD genes and mismatch repair genes were found. In the group of fibromatous naevi, MLH1 correlated significantly with GADD45 and highly significant with GADD34 expression. Different correlations in naevi groups investigated show the different functional connections between the genes of DNA repair.

DO-7 antibody against p-53 antigen was applied for investigation of melanomas of facial skin (25 cases), oral cavity (17 cases), eye (18 cases) and naevi (25 cases). The p-53 index value (% of p-53 positive cells) was correlated with the thickness of the tumour, the presence of metastases and survival time. The difference in p-53 index between naevi and melanomas was statistically significant (p < < 0.01). A significant correlation was found between the p-53 index value and the thickness of the tumour, the presence of metastases and follow-up for patients with skin, oral and ocular melanomas. The possible diagnostic and prognostic significance of p-53 antigen in melanomas and naevi of the head and neck area is discussed.
47 cases of malignant fibrous histiocytoma were stained immunohistochemically for the presence of Ki-67 antigen and c-myc oncoprotein. Percentage of Ki-67 and c-myc positive cells correlated with each other and with grade of tumour, mitotic index, cellularity and survival time of patients, but not with the extent of necrosis. Differences of mean values of Ki-67 index between grade I and grade II tumours as well as between grade II and grade III tumours were highly statistically significant. All 3 parameters connected with c-myc expression (index, intensity and score) differed significantly between grade I and grade II tumours but not between grade II and grade III MFH. The possible differential role of Ki-67 antigen and c-myc oncoprotein for improvement of grading is discussed.
121 cases of cutaneous melanomas, 82 lymph nodes with melanoma metastases and 62 naevi were stained immunohistochemically for presence of Ki-67 antigen and c-myc oncogene. A significant correlation between expression of investigated markers, and survival time as well as presence of metastases, in cutaneous melanomas was found (p < 0.05). The role of Ki-67 antigen for differential diagnosis between naevi and melanomas is also demonstrated.
57 cases of malignant fibrous histiocytomas (MFH) were examined, immunohistochemically for the presence of p-53-tumour suppressor gene with monoclonal antibody DO-7 (Dako) and for the presence of Ki-67 antigen using monoclonal antibody MIB-1 (Dianova). The statistical relationship between the overexpression of p-53 and grading parameters (mitoses, cellularity, necrosis) and expression of Ki-67 antigen as well as survival time, was determined. The presence of p-53 antigen was found in 29.8% of grade II and grade III tumours, but not in MFH of grade I. The p-53 positive cases demonstrated a significantly higher mitotic index (p < 0.05), higher Ki-67 index (p < 0.05) and shorter survival time (p < 0.05) in comparison to p-53 negative cases. Differences concerning cellularity and extent of necrosis, between p-53 positive and p-53 negative tumours were not found (p > 0.05). The prognostic role of p-53 overexpression in MFH is discussed.
Twenty nine benign naevi and 59 melanomas of different Clark level and locations were stained immunohistochemically for the presence of p53 and 3 members of Growth Arrest. DNA Damage inducible family: GADD 34, GADD 45 and GADD 153. All naevi were p53 negative and highly GADD positive. Differences in the expression of all 3 GADD proteins between naevi and melanomas were highly significant (p = 0.0001). The expression of p53 increased and the expression of GADD proteins decreased with the Clark level of melanoma thickness. The survival time of patients correlated negatively with p53 positivity and positively with GADD 45 and 153 expression. Prognostic significance for GADD 34 was not found. Our conclusion is that GADD proteins play an important role in the malignant transformation of naevus to melanoma and GADD 45 and GADD 153 proteins can influence patient survival.
As recurrence rates in breast cancer depend significantly on degree of malignancy (6-9) and mitotic rates as part of the grading system are difficult to assess in exact manner, we widened the grading system by evaluation of the proliferative compartment to get more information about proliferation activity as an important factor for tumor-progression (15, 16). This additional analysis can be used as a control factor for the correctness of the evaluation of mitotic activity. We practice this procedure at present as a "working formulation". New antibodies are in preparation which allow to exclude the highly variable G1-Phase. Further precision and control of degree of malignancy can be achieved by photometric analysis of S-Phase, 5c exceeding rate and state of ploidy according to Auer scheme.
