[Significance of parameters of automated reticulocyte analysis in diagnosis and evaluation of treatment outcome in B12-deficient anemia].

The results of examinations of reticulocytes in blood samples of 12 patients with the confirmed diagnosis of B12-deficit anemia and of 20 virtually healthy subjects (control group) by means of the GEN-S hematological analyzer (Beckman-Coulter) are described in the paper. Typical changes of the reticulocyte parameters are defined and their dynamics (with treatment by Vitamin B12) is studied. It was established that the additional parameters of the reticulocyte analysis, characterizing the quantity of immature reticulocytes and the fraction of immature reticulocytes, increase by 3 days before the total quantity of erythrocytes increases, which means that the above parameters can be used earlier as criteria in evaluating the treatment efficiency.

[Reference values for automated reticulocyte analysis].

Reference values for all 7 parameters of the analysis of reticulocytes by using hematological analyzer GEN-S and obtained during blood examination of 115 men and 120 women, aged 20 to 60, are presented in the paper. Reference limits can be used in practical medicine to evaluate the results obtained through an automated analysis of reticulocytes.

[Automated analysis of reticulocytes].

An evaluation of the performance of the GEN-S analyzer in examining the parameters of reticulocytes is described. The automated and manual methods of counting the relative quantity of reticulocytes were compared. It was established that the precision of the analyzer significantly surpasses the manual method results. The variation coefficient for the parameters of examination of reticulocytes is lower at a higher content of reticulocytes and the accuracy of the method is better if the content of reticulocytes in blood is higher. The parameters, characterizing the fraction of immature reticulocytes, are least accurate. The study demonstrated that the automated method of analyzing the reticulocytes by using the GEN-S analyzer can be used to ensure a more accurate monitoring of changes of the function of erythropoesis at different anemic conditions and to assess the conducted therapy.

[Laboratory diagnosis in tuberculosis].

The most reliable signs of tuberculosis infection is detection of M. tuberculosis complex in biological fluids and biopsy specimens from focus of lesions. The sensitivity of laboratory diagnostic methods is different for different biological materials, which dictates the utilization of a complex laboratory approach, including microscopy, bacteriological methods, detection of specific IgG, IgM, IgA, and polymerase chain reaction.

[Automation and organization of technological process of urinalysis].

Results of introduction into practice of a working model of industrial technology of laboratory studies and KONE Specific Supra and Miditron M devices are shown as exemplified by clinical analysis of the urine. This technology helps standardize all stages and operations, improves the efficiency of quality control of laboratory studies, rationally organizes the work at all stages of the process, creates a system for permanent improvement of the efficiency of investigations at the preanalytical, analytical, and postanalytical stages of technological process of laboratory studies. As a result of introduction of this technology into laboratory practice, violations of quality criteria of clinical urinalysis decreased from 15 to 8% at the preanalytical stage and from 6 to 3% at the analytical stage. Automation of the analysis decreased the need in reagents 3-fold and improved the productivity at the analytical stage 4-fold.

[Main principles of bone marrow puncture biopsy studies].

A scheme for morphologic analysis of bone marrow puncture biopsy specimens, order of operations, description of the specimen and its composition by hemopoiesis stems, and order and variants of description of the morphology of unidentified cells are proposed. The paper is recommended as a methodological guide for clinical laboratory diagnosis and a base for designing computer program.
