[Interaction of the complex copper compound Cu-2 with liver monooxygenases].

It has been shown that the antitumour drug Cu-2 (copper complex compound) inhibited the activity of liver monooxygenases in male CBA mice. The in vivo experiments have revealed a considerably increased duration of sleep in mice treated with hexenal after the administration of different Cu-2 doses. In vitro, after the incubation of intact mouse liver microsomal fractions with different concentrations of Cu-2 the level of cytochrome Y-450 was decreased and a non-active form of hemoprotein--cytochrome P-420--appeared. At the same time, after the incubation of Cu-2 with liver microsomal fractions stabilized by 20% glycerol type I spectral changes (Ks 330 microM) were registered. This shows the possible metabolism of Cu-2 by cytochrome P-450. The role of the revealed interaction of Cu-2 with liver microsomes is being discussed for the chemotherapy of cancer.

[Effect of the induction and inhibition of liver monooxygenases on the toxic and therapeutic action of vincristine].

The influence of phenobarbital and ziksorin, liver monooxygenase inductors and the influence of inhibitor SKF 525-A on the toxic and therapeutic effects of vincristine were studied on CBA and C57Bl mice (with hemoblastosis La or intact). It was shown that acute toxicity and therapeutic activity of vincristine lowered on induction of the liver monooxygenases, whereas inhibition of this enzymatic system resulted in increased toxicity and lowered therapeutic activity of vincristine. The possible use of these results in treatment of cancer patients is discussed.

[Liver mono-oxygenases and the pharmacodynamics of carminomycin and rubomycin].

The effect of carminomycin on the activity of liver monohydroxygenases was estimated. It mas noted that in doses of 2, 3 and 4 mg/kg the antibiotic had no effect on the sleep duration in mice treated with hexenal. This meant that it did not change the activity of the enzymatic system. Induction and inhibition of liver monohydroxygenases did not influence the toxic effect of carminomycin used in doses of 5 and 10 mg/kg. It was also shown that the toxicity of both rubomycin and adriamycin decreased on induction and increased on inhibition of liver monohydroxygenases. Like adriamycin rubomycin lowered the activity of this enzymatic system. Administration of rubomycin in a dose of 5 mg/kg prolonged the duration of the hexenal sleep almost 1.5-2 times as compared to the control. The possible participation of liver monohydroxygenases in metabolic transformations of the antracycline antibiotics is discussed.
