Oxygen-derived free radical generating capacity of polymorphonuclear cells in patients with ulcerative colitis.

Oxygen-derived free radical generating capacity of polymorphonuclear cells in 27 patients with ulcerative colitis, 10 with acute bacterial diarrhea and 20 healthy volunteers, was measured by the luminol-dependent chemiluminescence method by stimulation of formyl-methionyl-leucyl-phenylalanine. Oxidative free radical generating capacity of polymorphonuclear cells in patients with active ulcerative colitis was markedly enhanced as compared with control (p less than 0.01), while this enhanced free radical production by the cells was not detected at remission stage. Serial analyses revealed that oxidative free radical production by the cells in patients with ulcerative colitis was markedly enhanced with clinical deterioration of the disease, but it returned to normal level with an improvement of clinical features. These results suggest that the increased oxidative free radical production by polymorphonuclear cells could be related to the pathogenesis or aggravation of ulcerative colitis.

[Reduction of 5-FU toxicity with improved nutritional status in tumor bearing mice].

This study was performed to determine the role of nutritional status as a determinants of 5 fluorouracil (5-FU) toxicity in a tumor bearing mice. After subcutaneous inoculation, of mammary tumor (81 B) mice were assigned randomly to two nutritional regimens, (1) 8% casein: D-mice, (2) 27% casein: N-mice. Mortality was increased significantly in D-mice (100%) compared with N-mice (0%) by administration of 5-FU (20 mg/kg). Intratumoral 5-FU concentration of D-mice was significantly lower than that of N-mice, 1 hour after i.p injection of 5-FU (100 mg/kg, 300 mg/kg). Both growth rate and intratumoral protein content of transplanted tumor was equivalent in two dietary group, N-mice and D-mice. Protein depleted status was associated with a significant reduction in Interferon production of spleen cells. Inhibition of cell proliferation in bone marrow and cytokine production were recognized in hypo-nutritional status. Thus, improved nutritional status reduced the morbidity and mortality associated with 5-FU treatment in tumor bearing mice.

[A case report of multiple intracranial tuberculoma associated with miliary tuberculosis and review of the literature].

We reported a case of multiple intracranial tuberculoma associated with miliary tuberculosis and reviewed the cases reported as intracranial tuberculoma in the past 11 years. A 41-year-old diabetic man was admitted to our hospital for the treatment of miliary tuberculosis and respiratory insufficiency. On admissIon, he had no neurological deficits except mild consciousness disturbance due to respiratory failure. He developed headache and mental confusion three weeks after the beginning of antituberculous therapy with isoniazid, streptomycin, rifampicin, and ethambutol. Neurological examination revealed that he had progressive right hemiparesis and was in a confusional state. Enhanced CT showed multiple intracranial nodular lesions. During 6 weeks, he had progressive neurological manifestations in spite of his initial antituberculous treatment. He responded well, however, to the chemotherapy with combination of isoniazid, kanamycin, pyrazinamide and ethionamide that were sensitive to tuberculous bacilli separated from his sputum. He became minimally right-hemiparetic by 6 weeks after the change of antituberculous medication. Serial enhanced CT scan proved to be of great value in the diagnosis and follow-up study of intracranial tuberculoma. From 1978 to 1988, there were 72 reported cases of intracranial tuberculoma in Japan; 37 were male, 32 were female and 3 were uncertain because of no detailed document. The age of onset was distributed from 6 month to 81 years in age and 2 peaks were seen in the second decade and fifth to seventh decade. Thirty-three (48%) out of 69 cases had multiple intracranial lesions. A few reports commented that neurological complications tended to appear even if they were under antituberculous therapy.(ABSTRACT TRUNCATED AT 250 WORDS)

[Long-term follow-up study of transcatheter arterial chemotherapy in patients with advanced breast cancer].

Intra-arterial infusion chemotherapy (I.A) was applied to 32 cases with advanced breast cancer. The subclavian artery was intubated and one shot of 30 mg of adriamycin was given every 3 days, with a total dose of over 150 mg. Then mastectomy was performed after this chemotherapy. Long-term follow-up study revealed that the response rates of the primary tumor to I.A chemotherapy provided the prospects for their prognosis. The 3-and 5-year survival of good responders (% tumor regression greater than or equal to 70%) was significantly better than those of poor responders (% tumor regression less than 70%). Then, four patients with advanced breast cancer were treated with combination of I.A infusion chemotherapy of Adriamycin and photoradiation by argon laser. The mean interval time for 50% tumor regression was shorter than in the cases treated with Adriamycin alone. In vitro, the effects of the photoradiation for MCF-7 breast cancer cell line were observed only at a high concentration of Adriamycin.

[Metastatic behavior of footpad-inoculated tumors in mice and the impact of OK-432 per os administration].

In the surgical treatment of cancer patients, it is very important to investigate the immunological potential of regional lymph nodes. The purpose of this study was to clarify the defence mechanisms of regional lymph nodes against cancer cell dissemination. The materials used for the study were C3H/He mice and a syngeneic breast cancer cell line (MM-48). When MM-48 tumor, 5 X 10(6) cells per mouse, was inoculated into the footpad, the footpad gradually increased in size, and half of the mice died within 20 days. The frequencies of metastasis for ipsilateral inguinal lymph nodes, axillary lymph nodes and mesenteric lymph nodes were as follows. On day 3 after tumor inoculation, no metastasis was found in any node. On day 6, all mice had inguinal lymph node metastasis, and half of the mice had metastases to axillary lymph nodes and mesenteric lymph nodes. On day 10, all mice had metastasis to all nodes. When 100KE/kg of OK-432 was administered orally every day, the titer of natural killer activity of mesenteric lymph nodes was remarkably increased after 7 days (from 12.3% to 62.9%). When mice were administered OK-432 simultaneously with tumor inoculation and on each day thereafter, the frequency of metastasis mesenteric lymph nodes was markedly decreased after day 7. It is thus suggested that regional lymph nodes may operate effectively against tumor cell attachment to the nodes.

A volumetric comparison of the vestibular nuclei in primates.

The volumes of each of the four vestibular nuclei, superior, lateral, medial and descending, were measured in 80 brains from 2 species of Scandentia, 18 species of prosimians, and 26 species of anthropoids. Size indices were calculated by comparing species-specific points to the nucleus volume-body weight allometry in prosimians, where the average prosimian was set at 1.00. The indices range from 1.78 in Saimiri to 0.48 in Gorilla, and the distributions by families overlap partially or completely. The observed trend in size indices is independent of changes in the neocortex and the ventral pons; average indices are 1.35 in New World monkeys, 1.20 in Old World monkeys, 0.74 in apes, 0.82 in man. Among prosimians, Galago, Galagoides and Tarsius (leaping locomotion) show significantly higher indices than Nycticebus, Loris and Perodicticus (slow movement without leaping). The lateral vestibular nuclear indices in Pongidae and man are extremely low, about half of those of the average prosimians. Correlation coefficients of size indices between the vestibular nuclei and other motor nuclei, such as the cerebellar nuclei, ventral pons and striatum, are analysed. The ratio of the vestibular nuclear volumes to the total brain volumes and the distribution of percentages of each vestibular nuclear volume to the total complex are also obtained.

[The effect of an arginine-imbalanced diet on the growth of implanted MM-48 and MH-134 tumors in C3H/He mice].

The arginine-imbalanced diet used in this study was a diet of 27% casein supplemented with 8% arginine monochloride. There was no difference in the serum protein level between C3H/He mice fed a casein diet (N-mice) and mice fed on the arginine-supplemented diet (A-mice). The serum arginine level was increased in the A-mice compared with the N-mice. Significant inhibition of MM-48 tumor incidence was seen in the A-mice when the transplanted tumor was below 5 X 10(6) cells. Growth inhibitory effect against MM-48 tumor was significantly enhanced in the A-mice with i.p. injection of OK-432. Interferon production of spleen cells was increased in the A-mice compared with the N-mice. We speculate that the effect of arginine depends partially on the immune functions in the mice.

[Interferon production in lymph nodes draining breast carcinoma and its augmentation by OK-432].

In breast cancer patients on whom modified radical mastectomy is performed, relatively more of the regional lymph nodes draining the breast carcinoma remain in comparison with standard radical mastectomy. Therefore, investigation of the functions of lymph nodes draining breast carcinoma has become important. Lymphocyte subsets of 33 axillary lymph nodes from 19 breast cancer patients were analysed using flow cytometry. In axillary lymph nodes, both OKT-3(+) cells and OKT-8(+) cells were decreased in comparison with those in peripheral blood. However, the OKT 4/8 ratio was increased in axillary lymph nodes. These findings suggest that axillary lymph nodes are immunologically more functional against cancer spread than peripheral blood. OK-M1(+) cells, Leu-7(+) cells and Leu-11a(+) cells were decreased in axillary lymph nodes in comparison with peripheral blood. The ability of IFN production in axillary lymph nodes and peripheral blood was analysed using the cytopathic effect of VSV-sindbis virus. After 72 hours incubation, IFN production of axillary lymph nodes showed maximum titer. When lymph nodes were co-cultured with OK-432, IFN production of axillary lymph nodes was strongly augmented. IFN production of axillary lymph nodes draining breast carcinoma were increased in comparison with peripheral blood. Axillary lymph nodes draining breast carcinoma would thus seem to be important as cytokine-producing organs. IFN has been found to be an activator of NK cells, cytotoxic T cells and IL-2 production. Axillary lymph nodes may therefore play an important role against the spread of breast cancer.

[Prevention of cancer chemotherapeutic agent-induced toxicity in postoperative breast cancer patients with glycyrrhizin (SNMC)].

It is well known that all cell lines, normal as well as malignant, have a sensitivity to the growth inhibitory effect of IFN or IFN inducers. We speculated that the antiproliferative effects of the IFN inducer SNMC might help alleviate the uncomfortable side effects of cancer chemotherapeutic agent. Fifty-seven patients with postoperative breast cancer received MMF (mitomycin, methotrexate, futraful therapy with SNMC and 60 were given MMF alone. There was only one patient (1.7%) in the former group in whom the MMF chemotherapy was discontinued because of liver dysfunction; however, 15 patients (25%) in the latter group had to stop the therapy because of its side effects. Thus, there was a significant effect of SNMC in preventing the side effects of MMF chemotherapy.

[Quantitative variations of lymphocyte subsets in various kinds of cancer patients in the terminal stage].

Immunological studies of the peripheral blood were made in terminal breast cancer and terminal abdominal cancer patients. Two immunological parameters were studied: (1) lymphocyte subsets and (2) proliferative response to PHA. A decrease in the number of OKT-3(+) cells and an increase in that of OKT-8(+) cells were observed in abdominal cancer. It was suggested that the immunological status in abdominal cancer is more suppressive than in breast cancer. Increases in the number of OK-M1(+) cells and Leu-7(+) cells were observed in breast cancer. It is suggested that cytotoxic lymphocytes increase in number in breast cancer more than in abdominal cancer.

Volume comparisons in the cerebellar complex of primates. I. Ventral pons.

Volumes of the ventral pons (VPo) were measured in 82 brains of primates, Scandentia, and Insectivora, and the relative size of the VPo was expressed by size indices. The reference line was through the prosimian average; the mean index of prosimians was set at 1. The taxonomic groups arranged in a sequence of ascending mean VPo indices were: Insectivora (0.04), prosimians (1.00), monkeys (2.55), apes (4.48) and man (9.44). Thus, the VPo of man is more than 9 times larger than that of isoponderous average prosimians, and more than 230 times larger than that of isoponderous "basal' insectivores. The indices may reflect trends and extent of VPo evolution in primate phylogeny.

Volume comparisons in the cerebellar complex of primates. II. Cerebellar nuclei.

Volumes of medial, interposed, and lateral cerebellar nuclei (MCN, ICN, and LCN) were measured in Insectivora, Scandentia, and Primates, including man. The relative size of the nuclei was expressed in size indices. Insectivora had by far the smallest cerebellar nuclei. The simians, in general, had larger cerebellar nuclei than the prosimians, but there was considerable overlap. From Insectivora to man, the MCN was the least progressive and the LCN the most progressive. The indices are expected to reflect the relative size of the three longitudinal zones of the cerebellum (vermis/MCN, pars intermedius/ICN, hemisphere/LCN). They, together with those of the ventral pons and cerebellum (part I), are discussed in relation to the predominant locomotor pattern of a species, and with reference to evolutionary trends in primate phylogeny.

[Correlation between the growth of transplantable tumors under protein deprivation and intratumoral amino acid concentration].

Nutritional restriction in the diet has a deleterious influence on tissue and transplantable tumor growths. Correlation between the growth of 81 B or MM 48 mammary tumors and intratumoral amino acid concentration was studied in syngeneic C3H/He mice fed a diet with a low (D) or normal (N) protein content. D-mice revealed a relatively longer lag phase than N-mice, but showed almost the same growth curve. Some kinds of amino acid, alanine, valin, glycine, and so on were recognized in D-mice tumors at higher levels more than in N-mice. In protein malnutrition, urea, the final amino acid degradation product, was not detected in the transplanted tumor or muscle of D-mice. The high tissue levels of alanine, glycine and glutamine are undoubtedly related to their participation in many metabolic reactions, so that they are readily and extensively synthesized. We speculate that D-mice would maintain an adequate intratumoral amino acid level through interorgan relationships in order for the transplanted tumor to grow.

[Prevention of 5-FU induced toxicity in C3 H/HE mice with interferon or with interferon inducers (poly 1: C, OK-432, Lentinan)].

It is well known that all cell lines, normal as well as malignant, have a sensitivity to the growth-inhibitory effect of Interferon, or Interferon inducers. 5-FU is a widely used anticancer drug considered to be a cell cycle-specific agent. We speculated that the antiproliferative effect of Interferon or Interferon inducers might affect the activity of 5-FU and diminish the uncomfortable side-effects of this agent. Initially this experiment, we observed the mortality rate in mice after administration of 5-FU alone, or 5-FU plus Interferon (alpha inducers). We then carried out cytofluorometric observation of bone marrow cells, and finally studied the influence of Interferon (or inducers) on the anticancer effects of 5-FU against MM48 tumors in C3/He mice. Findings were as follow: 1) Administration of beta-type Interferon for 2 days, following the administration of a toxic dose (140 mg/kg/w) of 5-FU, revealed significant protection from mortality in C3H/He mice. This effect was also observed when Interferon inducers (Poly I: C, OK-432, Lentinan) were administered with 5-FU. Body weight loss was 10.4% at nadir in mice treated with 5-FU alone, while there was a body weight gain of 36.3% in mice treated with 5-FU plus Interferon. 2) Cytofluorometry of bone marrow cells obtained from mice femur at timed intervals after the administration of OK-432, or Lentinan indicated that these drugs inhibited the entry of cells into the S-phase, causing then to accumulated in the G0.G1-phase during the first 48 hours. 3) Histological findings of intestinal mucosa in mice showed prominent destruction after bolus injection of 350 mg/kg of 5-FU alone, while these findings were not observed in mice after the bolus injection of 350 mg/kg of 5-FU combined with Interferon. 4) The tumor volume ratio (T/C) was significantly reduced in the group treated with 5-FU alone as well as that treated with 5-FU combined with Interferon (or inducers) in C3H/He mice bearing MM48 tumors. All mice were dead about 2 weeks after administration of 5-FU alone, while all mice treated with 5-FU plus Interferon (or inducers) were alive in the same period. From these results, we speculate that Interferon, or Interferon inducers play an important role in the prevention of side effects of cell cycle-specific cytotoxic drugs like 5-FU, without decreasing their anticancer activity.

The cells of origin of cat trigeminothalamic projections: especially in the caudal medulla.

Thalamic projections from the caudal medulla of the cat were examined using the method of retrograde axonal transport of horseradish peroxidase (HRP). Injections were made unilaterally in various thalamic regions. Large injections labeled cells in the subnuclei: zonalis (Vcz), gelatinosus (Vcg), magnocellularis (Vcm), reticularis dorsalis (Vcrd) and ventralis (Vcv) medullae oblongatae. The largest number of labeled cells were in Vcz, Vcrd and Vcrv. Most of the labeled cells in Vcz and Vcrd were contralateral to the injection site, although the labeled cells in the Vcrv were bilateral. Small injections were made into the medial, lateral and dorsal regions of the nucleus ventralis posteromedialis (VPM), rostral regions of the posterior nuclei (POm and PO1), caudal POm, the nucleus centralis lateralis (CL) and the center median-parafascicular nuclear complex (CM-Pf). Most of the neurons in Vcz were found to project to the medial VPM and some to the caudal POm. A small number of cells in the Vcrd project to the medial VPM, but a large number project to the caudal POm and CM-Pf complex. The largest number of neurons projecting to the CM-Pf complex was present in Vcrv, where the labeled cells were bilateral. The types of trigeminothalamic projecting cells and the sizes of their somata were observed for different subnuclei and a considerable difference was found to exist among the subnuclei. This anatomical differentiation of the trigeminothalamic projections probably reflects a functional specialization of neuronal location since the functional properties of neurons vary according to their locations.
