Ab initio simulation of carbon clustering on an Ni(111) surface: a model of the poisoning of nickel-based catalysts.

In this work, we studied the poisoning of a nickel surface due to carbon. Performing ab initio simulations, within the framework of density functional theory, we computed the surface energy of the nickel (111) surface as a function of carbon coverage. On the basis of these results, we can assert that the stable state of the nickel/carbon surface is either a clean nickel surface or a fully carbon-covered nickel surface, which has a graphitic configuration. The relative stability of the two states depends on the temperature and partial pressure of the carbon gas. At fixed nominal carbon coverage, the most stable configurations are those forming carbon clusters. However, the nickel sites hosting these clusters change from hexagonal close packed/face centered cubic (hcp/fcc) sites to on-top sites when going toward larger clusters. This indicates that poisoning due to graphitic patches occurs on on-top sites.

The immune system of sea bass, Dicentrarchus labrax, reared in aquaculture.

The sea bass Dicentrarchus labrax is one the most important seawater fish species of south Europe and Mediterranean aquaculture, and studies on its immune system are important for both scientific and applied purposes. In this paper, we summarise the results obtained in studies of the immune system in this species, and present original data on cell-mediated acquired immune response.

Immunopurification of T-cells from sea bass Dicentrarchus labrax (L.).

The monoclonal antibody DLT15, specific for thymocytes and peripheral T-cells of the teleost fish Dicentrarchus labrax (sea bass), was used to purify immunoreactive cells from blood and gut-associated lymphoid tissue. The purification was performed by immuno-magnetic sorting of leucocyte fractions enriched by Percoll density gradient centrifugation, and the purity of the isolated cells was estimated by cytofluorimetric analysis. Following a single step, the percentage of DLT15-purified cells was 88 +/- 10% for gut-associated lymphoid tissue and 79 +/- 18% for blood leucocytes. DLT15-purified cells from gut-associated lymphoid tissue were employed for RNA extraction and cDNA synthesis. In RT-PCR experiments using as primers degenerate oligonucleotides corresponding to the peptide sequence MYWY and VYFCA of the trout TcR beta chain, a 203 bp product was amplified. When sequenced, the cDNA was found to show 60% nucleotide identity to the trout TcRV beta 3. By 3'-RACE the cDNA was elongated to obtain the TcR constant region, with high similarity to other fish TcR sequences. These results strongly suggest that cells recognised by DLT15 are putative T lymphocytes.

Development of a solid phase microextraction method for detection of the use of banned azo dyes in coloured textiles and leather

A new method to detect the use of banned azo dyes in the manufacture and treatment of coloured textiles and leather is described. The determination of the azo dyes was made by quantification of aromatic amines generated by reductive cleavage in a citrate buffer medium. The aromatic amines were then extracted from 1 mL of the reaction solution by means of solid phase microextraction (SPME) and determined by gas chromatography/mass spectrometry (GC/MS). We also evaluate accuracy, precision, range of linearity and limit of detection for the eighteen aromatic amines investigated, and show that the method is comparable with current established methods. A monoclonal antibody against chorion proteins of the sea bass Dicentrarchus labrax (Linnaeus, 1758): studies of chorion precursors and applicability in immunoassays.

The monoclonal antibody DLE7 was obtained against 44- to 50-kDa polypeptides solubilized from the vitelline envelope of the Mediterranean sea bass Dicentrarchus labrax. In Western blot analysis of chorion lysates it recognized cross-reactive bands at 44 kDa, 48 kDa, and 110 kDa. Previous affinity blotting with concanavalin-A showed that most of solubilized bands were glycosylated. Enzymatic deglycosylation of chorion proteins followed by Western blot analysis with DLE7 showed that the 48-kDa and 110-kDa antigens were differentially affected by endoglycosidase-F treatment. When DLE7 was employed in immunofluorescence analysis, isolated chorions and ovarian cryosections stained intensely. Positivity was also observed in liver cryosections of spawning females but not in liver of males and nonspawning females. To study the origin and delivery of chorion proteins, DLE7 was used in Western blot analysis of liver homogenates and blood serum of spawning females. Cross-reacting bands were detected in liver (90 kDa) and serum (180 kDa, 50 kDa). DLE7 was also used for the first time to set up an indirect ELISA assay to detect egg antigens in the blood of egg-producing females, raising the possibility of using DLE7 as a female-specific marker of spawning for sea bass.

Distribution of radionuclides in the environment in northern Italy after the Chernobyl accident.

Soon after the Chernobyl nuclear accident, the air-pumping stations in Pavia (northern Italy) were alerted. In a few days, a rapid increase in radionuclide concentration in air particulates was observed. Consequently, an environmental radioactivity monitoring programme was started in which several matrices such as soil, grass, vegetables and cows' milk were subjected to direct gamma-ray spectrometry. The radioactivity distribution and its variation with time is presented, discussed and compared with other available data. Detection limits, precision and accuracy are also reported, and depth profiles in soils for 137Cs are presented and correlated with soil quality parameters. A survey of environmental radioactivity in soil, in a search for residual Chernobyl fallout, was carried out and a map of the 137Cs distribution over a large area in northern Italy is presented and discussed.

[Effects of acute intravenous administration of propafenone on left ventricular function in patients with acute myocardial infarction].

UNLABELLED: In order to evaluate the effects of propafenone (an antiarrhythmic class 1 c agent) acutely administered intravenously on left ventricular function, 10 patients with acute myocardial infarction (AMI), affected by arrhythmias (greater than or equal to Lown class III), belonging to Killip class I and II, and with normal serum electrolyte levels, were studied 2-4 days after an acute episode. No patients had previously been treated with antiarrhythmic drugs, or, if treated, the agent was withdrawn since at least 4 half-lives. In no case were inotropic substances or other pharmacological treatments, capable affecting L.V. function administered. Intravenous nitroderivative (Venitrin) infusion was continued when indicated. Each patient was administered a propafenone bolus (1 mg pro Kg in 4 minutes). Two dimensional and Doppler echocardiography were performed under basal conditions i.e. before propafenone infusion, during the infusion and after 1, 2, 3 and 4 hours, deriving following parameters: L.V. end diastolic and end systolic dimensions and volumes, ejection fraction (area-length method), II motion abnormalities, and transmitral flow velocity profile (E/A ratio). Statistical data were obtained using two way analysis of variance. RESULTS: In no case was the treatment interrupted, on the basis of our protocol criteria (reactions of hypersensitivity, proarrhythmic effects, acute hemodynamic deterioration that requires immediate positive inotropic and chronotropic agents administration, worsening of previous arrhythmic class). Heart rate and systemic arterial blood pressure did not change significatively during or after infusion.(ABSTRACT TRUNCATED AT 250 WORDS)

Rare-earth elements in the NBS standard reference materials spinach, orchard leaves, pine needles and bovine liver.

A highly sensitive radiochemical neutron activation method for the determination of all rare-earth elements (REE) in NBS biological reference materials is described. The materials are irradiated, dissolved in HF/HCl solutions, mixed with scandium and REE carriers (except La, Pr, Nd, Dy, Er), and the resulting solutions evaporated to dryness. The residues are dissolved in HCl and the REE precipitated as fluorides on addition of HF/NH4F solutions. The REE fluorides were collected, dissolved in a nitric/boric acid solution and the radioactivity of the resulting solutions determined by gamma spectrometry. The concentrations of REE in the NBS SRM Spinach, Orchard Leaves, Pine Needles, and Bovine Liver were found to be in the ng/g to microgram/g range. The relative standard deviations are approximately 8%. The results agreed, within experimental errors, with literature values. The distribution patterns of REE in the NBS materials relative to chondritic meteorites resemble the patterns for geological materials.

Immunopurification of B lymphocytes from sea bass Dicentrarchus labrax (L.).

The monoclonal antibody DLIg3, specific for immunoglobulins and B cells of the teleost fish Dicentrarchus labrax (Mediterranean sea bass), was used to enrich immunoreactive cells from peripheral blood, spleen, and head kidney leukocytes. The purification was performed by immunomagnetic sorting of leukocyte fractions enriched by Percoll density gradient centrifugation, and the purity of the isolated cells was estimated by immunofluorescence and cytofluorimetric analysis. Following a single immunopurification step, the percentages of DLlg3-purified cells were 61% +/- 6% from peripheral blood leukocytes, 66% +/- 5% from splenocytes, and 77% +/- 9% from head kidney cells. DLIg3-immunopurified cells, from the head kidney of antigen-primed fish displayed an enhanced proliferation index when incubated with the immunization antigen. DLIg3-purified cells from head kidney lymphoid tissue were employed for RNA extraction and complementary DNA synthesis, and in reverse transcriptase polymerase chain reaction experiments using specific primers corresponding to the sequences of the sea bass immunoglobulin light chain, and of T-cell receptor. DLIg3-purified cells displayed enhanced expression of the immunoglobulin gene, and lower expression of T-cell receptor.

Effects of fire on the state of several elements in some soils of Sardinia.

In order to individuate the modifications induced in the soil by fires relatively to the mobility of metals and rare earth three soils of Sardinia which differ in their mineralogical and physico-chemical characteristics were sampled. The analytical results obtained on the samples drawn at different depths (0-5 and 10-30 cm) three months after a fire and on the tests indicate that only the surface layer underwent significant modifications. The dynamics of metals and the distribution of the rare earths were found to depend, besides the amount and quality of the burned material, on the different behaviour of elements towards leaching. The diffractometric analysis showed that the soil surface layer of all the samples did not exceed 400 degrees C.
