[Cathepsin D activity in amniotic fluid and serum during labor and puerperium in normal cases and those complicated by EPH-gestosis].

Cathepsin D activity during labour and puerperium in normal cases and those complicated by mild or moderate EPH-Gestosis was examined. Sera from maternal and umbilical cord blood, as well as the extraplacental blood and amniotic fluid were used for determinations. The gestational age of physiologic pregnancies and pregnancies complicated by EPH-Gestosis was 40 weeks. The first stage of labour was from 4 to 6 hours. The second stage of labour was over 10 minutes. In the group of women with mild and moderate EPH-Gestosis, the maternal serum activities on the third and fifth day of puerperium were significantly lower than in nonaffected subjects. In patients with moderate EPH-Gestosis, the activities in the maternal sera on the first day of puerperium, and in the umbilical blood were decreased when compared to controls. Uniformly, activities of the studied enzyme have been found to be lower in the umbilical cord blood sera than in the sera of the parturients. In cases of mild and moderate EPH-Gestosis the enzyme activity in the amniotic fluid was much lower than in controls, whereas in the extraplacental blood it was much higher.

Cholesterol synthesis in the rat brain in course of late development as determined by 3-hydroxy-3-methylglutaryl CoA reductase activity.

The activity of 3-hydroxy-3-methylglutaryl-CoA reductase (E.C. 1.1.1.88), the key regulatory enzyme in the process of cholesterol biosynthesis was evaluated in different development stages of rat brain. The obtained results have shown an almost twofold increase of HMG-CoA reductase activity in one year old rats compared to the reference 4 months old animals. The activity of the cerebral HMG-CoA reductase in two years old animals dropped to the level observed in the reference group. The enhancement of activity of the regulatory enzyme for cholesterol biosynthesis occurs in parallel with an appearance of quantitatively minor, other than cholesterol, free sterol normally absent in the brains of young animals.

[Lavage of the whole lung in a patient with pulmonary alveolar lipoproteinosis].

The first in Poland procedure of whole lung lavage is reported. The patient had intravitally diagnosed pulmonary alveolar lipoproteinosis. Difficulties in an effective and safe performance of the procedure are discussed. Modernized methods of lung lavage in patients with severe respiratory failure due to pulmonary alveolar lipoproteinosis are described.

Persistent rotavirus infection in mice with severe combined immunodeficiency.

Rotaviruses are important pathogens of human infants and the infants of many animal species. The disease produced by these viruses can be described as an acute, self-limiting diarrheal disease, with virus replication localized to the differentiated epithelial enterocytes of the small intestine. Immunologically normal infants shed virus for approximately 5 to 12 days after the onset of infection. Recently, it has been shown that rotavirus can produce a chronic infection in severely immunocompromised children, with virus shedding and intermittent diarrhea lasting from 6 weeks to 2 years (G. A. Losonsky, J. P. Johnson, J. A. Winkelstein, and R. H. Yolken, J. Clin. Invest. 76:2362-2367, 1985; F. T. Saulsbury, J. A. Winkelstein, and R. H. Yolken, J. Pediatr. 97:61-65, 1980). These findings point to an important role for the immune system in recovery from the disease. The study described here examined the outcome of murine rotavirus infection in mice with severe combined B- and T-cell immunodeficiency (SCID) and in immunologically normal seronegative BALB/c mice. Persistent rotavirus infection was established in all mice with SCID which had been inoculated orally as pups. Low levels of virus replication and constant fecal virus shedding characterized the chronic infection. This is the first report of a persistent rotavirus infection in an animal model.

[Cathepsin D activity in the blood serum of patients in labor and in the serum of extraplacental and neonatal blood].

Cathepsin D in serum of women in labour, retroplacental blood and newborn's blood degradate hemoglobine denatured with urea and the serum protein most intensively in pH 3,5. The denatured protein is more liable to cathepsin D activity than the serum protein. Cathepsin D of the retroplacental blood serum shows the highest activity, the activity in the blood serum of women in labour is lower and it is the lowest in the newborn's blood.

[Enzymatic activity in cerebrospinal fluid in the monitoring of the brain lesions following intracranial tumors].

After intracranial tumour surgery edema, ischaemic and haemorrhagic changes are developing in the brain. It causes leakage of certain intracellular components and enzymes into extracellular and cerebrospinal fluid. This study includes 46 patients operated on for cerebral tumours. Lumbar CSF was obtained in postoperative course and the activity of AST, ALT, LDH, alpha HBDH, CK, gamma GT and oxygen concentration were estimated. In the control group were 10 patients suffering from sciatic pain and CSF was obtained during radiculography. For diagnostic and prognostic purposes of biochemical markers statistical analyse methods were employed and comparisons with clinical factors were carried out. In conclusions we suggest that enzymes AST, alpha HBDH, CK and oxygen content give valuable additional information of postoperative brain damage.
