[Liposomal systems--prospective drug forms. II. The creation and characterization of liposomal systems for the oral use of narcotic analgesics].

Liposomal systems were prepared for oral usage of narcotic analgesics morphine and phentanyl. The systems were examined for homogeneity in respect to seize size and form of liposomal structures by a diffraction-laser method and electron microscopy. The liposomal drug forms were studied on biological experiment as the change in the body temperature of rats was described. It was established that in rats oral usage of liposomal systems with morphine respectively with phentanyl caused significantly better effect in comparison with active substances, not taken in liposomes.

[COmparative pharmacokinetic studies of 5-nitrox and 5-NOK in volunteers].

The authors carried out studies on 8 volunteers with normal renal function after a single dose of 5-nitrox of 200 to 250 mg (mean 3 mg/kg of body weight). The dynamics of concentrations in plasma and urine was discovered. On the basis of the obtained experimental data the therapeutic scheme of application was indicated. The comparative studies with the analogous preparation 5-nok (of the firm LEK-Lubliana), administered on the same contingent under the same conditions of the trial, showed similar results both in respect of plasma concentrations and in respect of its excretion in urine.

[Pharmacokinetic studies of 5-nitrox in kidney failure patients].

The authors carried out initial pharmacokinetic studies of 5-nitrox on patients with compensated, first and second degree of decompensated renal insufficiency in view of determining dosage and intervals for application of the preparation. They found regular changes in the profile of the curves, manifesting the concentrations of 5-nitrox in plasma and urine for each interval of time-with advancement of renal insufficiency the peak levels diminished and were seen at later intervals as the course of the curves were of plateau character in patients with second degree of renal insufficiency. The analysis of the results showed that treatment of patients with decompensated renal insufficiency was not justified due to the retention of relatively high plasma level up to the 12th hour and rather low, unefficient concentrations of urine. 5-nitrox could be used in a single dose of 200 mg at 8-hour intervals in patients with compensated first degree of decompensated renal insufficiency.

[Effect of beta-adrenergic drugs on humoral immune response factors].

The authors examined the influence of beta-adrenergic stimulators Isoprenaline and Salbutamol as well as beta-adrenergic blockers Propranolol, Practolol and Pindolol on the proliferation of cells, forming plaques, 7S-immunoglobulins, hemagglutination titres and hemolytic activity of the complement. Primary stimulation with subsequent inhibition was induced by beta-adrenergic agonists and inhibition--by beta blocker Propronanol in a base of 10 mg/kg on the proliferation of cells, forming antibodies. The results were in correlation with hemagglutination titres. Salbutamol and beta-blockers inhibited immune hemolysis. The mechanisms of the observed effect are discussed in respect to the specific and nonspecific effect of the drugs used.

[Effect of beta-adrenergic blockaders on the uterine musculature of rats].

The authors carried out a study on beta-adrenergic blockers Proprafnolol, Pindolol and Practolol, on an isolated uterus of a rat, obtained at various phases oestrus. The Beta-adrenergic blocker Pindolol manifested inhibiting effect on the spontaneous contractility of the uterus more marked after elevated level of gestagens. The pharmacological analysis gave foundation to the authors to assume that the relaxation of Pindolol was due to direct myotropic action. Propranolol in large doses and at high gestagenic level manifested antioxytocic activity. Practolol did not affect both spontaneous uterine activity as oxytoxin contraction.

[Effect of beta-adrenergic blockers on complement].

The authors, examined the effect of beta-adrenergic blockers Propranolol, Pindolol, Practonol and P11 on complement in the serum of guinea pigs after continuous administration. The follow-up was made dynamicaly: on the 10th, 20th and 30th day. There was a considerable lowering of the complement level on the 20th and 30th day in all animals treated with beta-adrenergic blockers. The experiments in vitro showed that the effect of Propranolol and and P11 on the serum of persons, guinea pigs and rabbits presented data on the complement inhibition, having the direct effect on the first component of the complementary systems.
