D-glucose transport activities in erythrocytes and hepatocytes of dogs, cats and cattle.

1. The activities of D-glucose transport and hexokinase were investigated in erythrocytes or hepatocytes of dogs, cats and cattle. 2. The mean D-glucose transport activity in erythrocytes of dogs was 6.0 nmol/min/mg protein, half the value of hepatocytes. 3. The activities of D-glucose transport in erythrocytes and hepatocytes or hepatic hexokinase of cats were about one-third of those of dogs. 4. Cattle with low blood glucose concentrations showed considerably low activities of D-glucose transport and hexokinase, about one-third of those of dogs.

[Effect of shikonin and its derivatives, pentaacetylated shikonin (MDS-004) on granuloma formation and delayed-type allergy in experimental animals].

Of twelve reduced and acetylated derivatives of shikonin, a chemical constituent of Shikon, the accelerating activity on granuloma formation and the inhibitory activity on delayed-type allergy were investigated in order to find a compound having more characteristic effect than shikonin on wound healing in experimental animals. As a result, it was found that a reduced and pentaacetylated derivative of shikonin, MDS-004, has more excellent pharmacological activity. MDS-004 (0.1-1 mg/pellet) accelerated dose-dependently felt-pellet-induced granuloma formation when given topically together with felt-pellets in rats. It also produced strong inhibition against delayed-type allergies (ear edema) caused by oxazolone and dinitrofluorobenzene by topical application of up to 1 mg/ear to the ear skin of mice; its potency was far superior to that of shikonin. Orally administered MDS-004, unlike shikonin, inhibited carrageenan-induced hind paw edema, and exhibited tendency to heal acetic acid-induced gastric ulcer in rats. However, MDS-004, as well as commercial wound healing drugs tested and shikonin, did not show any healing action in the incised and open wound models in rats, if applied topically to the wound as 5 and 10% powders. On the other hand, MDS-004 did not produce irritative action on the ear skin at a topical dose of 1 mg/ear different from shikonin, and any behavioral changes after oral administration of 100 mg/kg in mice. These results suggest that a white powder MDS-004, different from deep purple shikonin, has accelerating action on granuloma formation without irritative action and stronger inhibitory action on delayed-type allergy by topical application than shikonin.

Thyroxine-binding globulin variant (TBG-Kumamoto): identification of a point mutation and genotype analysis of its family.

Thyroxine-binding globulin (TBG) is the major thyroid hormone transport protein. Several inherited TBG variants resulting in partial or complete TBG deficiencies have been shown to be caused by either one or two nucleotide substitutions, or one nucleotide deletion in the coding regions of the TBG gene. In this report, a Japanese female patient (proband) with hyperthyroid state, whose lower TBG levels did not return to normal under the euthyroid state after treatment was examined. Genomic DNA samples from the proband with thyroxine-binding globulin deficiency (termed TBG-Kumamoto) and her family were subjected to the polymerase chain reaction, and the generated DNA fragments were sequenced. A single nucleotide substitution in the codon for the amino acid 363 of native TBG molecule (CCT to CTT) was found, resulting in the replacement of proline by leucine. It was revealed that the proband was a heterozygote and her father was a hemizygote. The mutation was confirmed by the allele-specific amplification of genomic DNAs from the proband and her father using oligonucleotide primers of normal or mutant residues at the 3' position in the polymerase chain reaction. These results indicate that the abnormality of TBG-Kumamoto is the consequence of this mutation. Genetically, this point mutation observed in TBG-Kumamoto might be classified as a new type of TBG deficiency.

Lethal and teratogenic effects of long-term low-intensity radio frequency radiation at 428 MHz on developing chick embryo.

Exposure of developing chick embryos to 428 MHz radio frequency (RF) radiation at a power density of 5.5 mW/cm2 for more than 20 days resulted in embryolethal and/or teratogenic effects and delayed hatching. These adverse biological effects were not due to any thermal effect of the RF radiation. We have demonstrated teratogenicity in the chick embryo as a result of protracted low-dose RF irradiation.

Atrial natriuretic peptide in the dog with mitral regurgitation.

The concentration and molecular form of the plasma atrial natriuretic peptide (ANP) in dogs with mitral regurgitation was investigated. Plasma ANP concentration in dogs with mitral regurgitation was significantly increased (29.4 +/- 1.88 pmol litre-1, n = 40) compared to that in the controls (14.5 +/- 0.62 pmol litre-1, n = 20, P less than 0.01). Molecular forms of plasma ANP were determined by the gel permeation chromatogram. A single peak corresponding to alpha-ANP was detected in the plasma from the controls. However, a peak corresponding to beta-ANP and, or, gamma-ANP was detected in the plasma of the dogs with mitral regurgitation in addition to alpha-ANP. These results suggest that the process of ANP synthesis was altered and excretion of ANP from the heart was enhanced in dogs with mitral regurgitation.

Glycated hemoglobin fractions in normal and diabetic dogs measured by high performance liquid chromatography.

We established a new analytical condition to measure the canine glycated hemoglobin by high performance liquid chromatography (HPLC) using cation exchange column. The canine hemoglobin gave five peaks consisting of 2 major and 3 minor hemoglobin fractions such as HbA1a, HbA1b and HbA1c. Measurement was done in 38 clinically normal dogs and 10 diabetic dogs. Mean HbA1c values (% of total Hb) in normal and diabetic dogs were 2.60 and 6.41%, respectively. And mean HbA1 values were 3.58 and 7.41%, respectively. The mean values of the canine HbA1c and HbA1 in diabetic dogs was higher than those in normal dogs, significantly (p less than 0.01). Advantages of the HPLC method and applicability for monitoring effectiveness of insulin therapy in the canine diabetes mellitus are discussed.

A cytotoxic antibody produced by allo-skin transplantation in cats.

A high titer of cytotoxic antibody against donor lymphocytes, tentatively designated as anti-F1, was found in one cat who received 3 times of skin allograft from the same donor. The anti-F1 antibody titer was 1:16, and the antibody activity was mostly detected in the IgG fraction. A cross-absorption test using erythrocytes from F1-positive animals revealed that the F1(+) antigen recognized by anti-F1 antibody was expressed on erythrocytes as well as lymphocytes. The frequencies of F1(+) and F1(-) types in 81 healthy cats, involving all the cats used for allo-immunization in the present study, were 30.9 and 69.1%, respectively. Moreover, F1 antigen was considered to be a hereditary antigen from family studies.
