A simple chromogenic test for rapid screening of Proteus and Providencia bacteria.

A rapid test for detection of p-nitrophenylalanine ammonia-lyase is described. The test is performed by suspending a loopful of bacteria in 0.5 ml of a buffered 1 mM solution of p-nitro-DL-phenylalanine (PNPA). The enzymatic activity is revealed by the formation of a yellow colour after 2 hours of incubation at 35 degrees C. Out of 285 strains of different enterobacteria species positive results were shown only by 75 Proteus and Providence strains. Thus, the PNPA test can be useful for rapid screening of these enterobacteria.

A rapid multistrips tests for determination of the enzyme profile in enterobacteria.

A simple test for the detection of bacterial enzymes has been developed using paper strip impregnated with different chromogenic substrates. The test was applied to 145 strains belonging to various species of the family Enterobacteriaceae. Positive reactions were revealed after four hours of incubation at 35 degrees C by the formation of a yellow colour indicating released nitrophenol or nitranilide where a loopful of bacteria had been placed. The results of the multistrips test were in agreement with those obtained by previously described methods for the detection of beta-galactosidase, beta-xylosidase, beta-glucuronidase, gamma-glutamyltransferase, and phenylalanine ammonialyase. The suitability of the test for rapid determination of the enzyme profile in enterobacteria is discussed.

Possible diagnostic value of aminopeptidases detected by chromogenic substrates in Enterobacteriaceae.

The presence of aminopeptidases revealed by three chromogenic substrates (chromozym PL, chromozym TH, and carbobenzoxy-glycyl-prolyl-arginine-p-nitranilide) has been investigated by a simple method in 153 strains of different genera and species of the family Enterobacteriaceae. The results suggest that the AP-PL, AP-TH, and AP-CGPA tests can be of diagnostic value for the identification of enterobacteria.

Aminopeptidases↗

Detection of aminopeptidases in Enterobacteriaceae by three simple chromogenic tests.

The action of 907 strains belonging to the various genera and species of the family Enterobacteriaceae were assayed on three chromogenic substrates used for the detection of aminopeptidases. The observed patterns of reactions may be useful for the biochemical identification of some serovars and biovars, particularly in the genus Shigella.

[Domestic injuries in childhood. Results of a survey carried out in a Sicilian area].

A retrospective investigation was performed with an anonymous questionnaire administered to the mothers of 2208 children aged between 3 and 11 years, randomly selected from the residents of three target areas: a country town, a residential and a popular quarter of a chief town (Catania, Sicily). Of the 2208 mothers in the three areas, 1151 completed the questionnaire with an overall response rate of 52.12%. In the last 6 months a large number of children (17.29%) suffered at least one accident at home. Predominating injuries were falls (71.86%) followed by cutting or stab-wounds (13.57%). The more frequent lesions were contusions (42.13%), wounds (29.95%) and fractures (14.22%). Almost the half of children victims of injuries required hospital care (44.72%). The majority of domestic accidents occurred in the bathroom (17.10%), in the kitchen (15.10%) and in the bedroom (10.55%). Fractures and wounds were more frequent among children aged 6-11 and 3-5 years, respectively. The highest frequency of domestic accidents among children of all ages living in the popular quarter (20.89%) and in the country town (19.18%) compared with those living in the residential quarter (11.67%) (chi2= 12,768, P<0.01) could be related to their lower socio-economic condition.

Taxonomic value of a chromogenic test for the detection of aminopeptidases in the genus Shigella.

In addition to the conventional methods for the identification of Enterobacteriaceae, enzymatic tests using chromogenic substrates have been proposed [1, 2, 5, 7, 8, 9, 10]. Many chromogenic and fluorogenic substrates are now available, and some of these have been employed for the determination of enzymatic profiles of Neisseria [3] and Enterobacteriaceae [7]. In this article, we report the activity of bacterial cultures of the genus Shigella on a new chromogenic substrate: chromozym PL.
