Burkitt's lymphoma in Cuba. I. Clinical and morphological features and EBV association.

The first 7 cases of histologically and cytochemically confirmed Burkitt's lymphoma have recently been studied in Cuba. Some of them presented with facial bone involvement and high anti-Epstein-Barr virus (EBV) antibody titers characteristic of endemic African cases. Others showed features of nonendemic cases. The distribution of patients in these two groups may be in relation to the behavior of the healthy Cuban population with respect to the EBV virus.

Ammoniacal silver staining of lymph node cells. I. its potential value for the diagnosis of Hodgkin's disease.

Lymph node biopsy specimens from 76 cases of Hodgkin's disease, 23 non-Hodgkin's lymphomas and 19 reactive affections of lymph nodes were studied. Formalin-fixed and paraffin embedded tissues were stained with hematoxylin and eosin, and post-formalin ammoniacal silver staining was used after BLACK and ANSLEY. The presence of basic protein accumulation in the cytoplasm of both Sternberg and Hodgkin cells was confirmed. To our knowledge it is the first report concerning the presence of these accumulations in the cytoplasma of atypical reticulum cells in Hodgkin's disease and in 3 of 7 cases of reactive follicular hyperplasia, in foreign body giant cells and in some lymphoblasts from 5 cases of Hodgkin's disease. The possibility of using the ammoniacal silver staining for identifying Sternberg cells is discussed. Also, it is proposed that when Sternberg cells are absent, the ammoniacal silver staining pattern of Hodgkin cells in an appropriate histological setting may be very indicative of Hodgkin's disease.

Anaphylaxis in the mouse--a model for using two-dimensional electroimmunodiffusion to study pathological changes in multiple serum proteins simultaneously.

Two-dimensional electroimmunodiffusion is rarely used in clinical chemistry, because its potentialities are not yet widely appreciated. Studying mouse anaphylaxis, we observed with this technique certain striking short- and long-term pathologic changes in several important serum antigens, which aptly demonstrate its uniqueness and potential clinical usefulness. We present some of these observations here, for illustration. We analyzed sera from CF-1 female mice that had been anaphylactically shocked with chicken conalbumin antigen and bled 45 min to 22 days afterward. In three separate experiments, with samples of serum being analyzed only once each, this technique easily and surely detected quantitative and (or) qualitative changes in several serum antigens. We discuss here the changes in 7S gamma1 immunoglobulin, C3 globulin, hemopexin, and alpha lipoprotein, and explain how some of these changes would have been difficult and others impossible to detect by currently routine tests. We urge the adoption of two-dimensional electroimmunodiffusion for frequent use in the clinical laboratory for diagnosis, prognosis, and monitoring pathological conditions.

Heterogenous expression of the EGF-receptor in human breast carcinoma.

The epidermal growth factor receptor was analyzed in the membrane fraction from a series of human breast carcinomas using a radioligand assay. The results were compared to immunostaining with monoclonal antibodies on tissue sections. All tumors with detectable receptor levels as measured by ligand binding showed a positive staining with monoclonal antibodies. In such tumors more than 50 percent of the cells were receptor positive with varying intensity. Tumors classified as receptor negative with the radioligand assay could be divided into two major groups based on their immunostaining. The largest group showed no reactivity with monoclonal antibodies. In a second smaller group, a positivity was observed in small clusters of tumor cells while the majority of cells were negative. The staining intensity in such clusters was usually low. It seems possible that this heterogenous expression of the epidermal growth factor receptor is of significance with respect to clonal selection in tumor spreading.
