Direct-test PCR for detection of meningococcal DNA and its serogroup characterization: standardization and adaptation for use in a public health laboratory.

A direct PCR test (DT-PCR) was established to detect Neisseria meningitidis DNA in clinical samples from patients with suspected bacterial meningitis. Specific primers for the 16S rDNA of N. meningitidis were designed to amplify a 600 bp DNA fragment. One hundred and ninety-three clinical samples were analysed, corresponding to 114 samples from patients diagnosed as positive and 79 as negative for infection by N. meningitidis using conventional methods (culture, latex agglutination and counterimmunoelectrophoresis). These samples were submitted to PCR by two different clinical sample preparation approaches (with and without DNA extraction and purification) and submitted to different PCR protocols to improve the results. In agarose gel detection, the sensitivity value for DT-PCR was 88.5 % and, using dot-blot DNA detection, the sensitivity increased to 96.4 %. The detection limit for meningococcus in cerebrospinal fluid was 2x10(2) c.f.u. ml(-1). Serogroup prediction was done using a multiplex PCR protocol and the sensitivity was 83 % for agarose gel DNA detection and 96.4 % using dot-blot DNA detection.

Variational solution for modal wave-front projection functions of minimum-error norm.

Common wave-front sensors such as the Hartmann or curvature sensor provide measurements of the local gradient or Laplacian of the wave front. The expression of wave fronts in terms of a set of orthogonal basis functions thus generally leads to a linear wave-front-estimation problem in which modal cross coupling occurs. Auxiliary vector functions may be derived that effectively restore the orthogonality of the problem and enable the modes of a wave front to be independently and directly projected from slope measurements. By using variational methods, we derive the necessary and sufficient condition for these auxiliary vector functions to have minimum-error norm. For the specific case of a slope-based sensor and a basis set comprising the Zernike circular polynomials, these functions are precisely the Gavrielides functions.

Localized defects in radiation-damaged zircon

The crystal structure of a radiation-damaged natural zircon, ZrSiO(4) (alpha-decay radiation dose is ca 1.8 x 10(18) alpha-decay events g(-1)), has been determined. The anisotropic unit-cell swelling observed in the early stages of the amorphization process (0.17% along the a axis and 0.62% along the c axis compared with the undamaged material) is a consequence of the anisotropy of the expansion of ZrO(8) polyhedra. Larger anisotropic displacement parameters were found for Zr and O atoms, indicating that the distortion produced by alpha particle-induced localized defects mainly affects the ZrO(8) unit. The overall shape of SiO(4) tetrahedra remains essentially undistorted, while Si-O bonds are found to lengthen by 0.43%.

Magnetic resonance imaging in HTLV-I associated myelopathy.

Magnetic resonance imaging of the brain and spinal cord were carried out for seventeen consecutive patients with HTLV-1 associated myelopathy (HAM). Eight patients had brain abnormalities and four had decreased thoracic spinal cord diameter. Brain lesions were mostly located in subcortical and periventricular areas. Our data suggest that diffuse central nervous system lesions are present in patients with HAM.
