Pharmacokinetics of novel immunostimulating drug kamantane was studied by using gas-liquid chromatography in experiments on rats. It was found that kemantane biotransformed rapidly after oral administration with the forming of active metabolite. Kemantane and its metabolites are distributed rapidly from the blood to organs. The drug is eliminated from the organism of rats as metabolite.
In a series of 130 patients with asthenic conditions related to borderline forms of neuro-mental disturbances the authors compared clinico-pharmacological action of the actoprotector bemitil with that of nootropic drugs (piracetam and piridotol). The treatment with bemitil was either continuous or intermittent, being conducted according to the scheme elaborated in the course of the study. Bemitil proved to be more effective than piracetam and piriditol in relation to its influence on the manifestations of the asthenic symptom complex in general. Its application was characterized by a faster onset of the therapeutic effect and a peculiar mild psychostimulating action, primarily manifested in the impact on obligate manifestations of the asthenic disorders. The degree of the psychotropic action of the drug was found to be subject to changes depending on the scheme of its use.

It has been established in experiments on mini pigs of Siberian origin that phenazepam given at a dose of 1 mg/kg per os during organogenesis has no embryotoxic or teratogenic action. The drug content in the blood of pregnant animals was determined simultaneously. A conclusion is drawn about perspectiveness of using mini pigs for testing embryotoxic activity of drugs.
A well-known protective effect of aminooxyacetic acid against thiosemicarbazide convulsions was confirmed; it was shown that a similar, although somewhat weaker activity, was exerted by sodium hydroxybutyrate. Surprisingly, the effect of aminooxyacetic acid was diminished by sodium hydroxybutyrate. Sodium hydroxybutyrate in combination with aminooxyacetic acid decreased the protective activity of the latter against thiosemicarbazide convulsions and diminished the extent of GABA accumulation characteristic of aminooxyacetic acid action. This effect is attributed to the competition between the aminooxyacetic acid, sodium hydroxybutyrate and GABA for alpha-ketoglutarate-GABA-transaminase and possible for the GABA-ergic receptor.

Dynamics of nonachlazine, a new anti-anginal agent, distribution in organs following its single and repeated introduction in the form of a potion and aqueous solution was studied in tests with albino rats. It was ascertained that on introduction of a ready for use medicinal preparation (potion) the drug is absorbed by the blood and enters internal organs at a quicker pace than does its aqueous solution. The type of the medicinal preparation of the drug has no relevance to the rate of its elimination from the organism. With its repeated introduction (over a space of 12 days) the nonachlazine level in the heart and kidneys goes up, whereas its content in the blood and liver experiences but a slight change.
The psychotropic activity, the content in the organs and tissues, the acute and chronic toxicity of phthorphenazine-depot - (phthorphenazine-decanoate)-2-triphthormethyl-10-/3/1-(beta-caprionyl-oxyethyl)-piperazinyl-4/-propy/phenothiazine - were studied as contrasted against phthorphenazine dehydrochloride. Phthorphenazine-depot has been found to display a prolonged action, a single injection being sufficient to depress conditioned avoidance reflexes for a period of 45-50 days. The antagonism of the drug to phenamine is manifest for 30 days after its administration. After 17 days from the instant of its introduction phthorphenamine is capable of prolonging the anesthetic action of sodium thiopental. Upon a single administration phthorphenazine-depot continues to be in evidence in the tissues of organs and in the blood over a period of 4-5 weeks. The drug is little toxic, this having been ascertained in the course of acute and chronic experiments.

As a result of studies into the pharmacokinetics of pentoxyfillin, no significant differences were established in the magnitudes of its non-model parameters in normal pregnancy and chronic placental insufficiency. However, there were differences in the metabolite formation rate in the groups under study. Distinctions were detected in the antiaggregation effect of pentoxyfillin. They consisted in a slower recovery of platelet aggregation parameters to the initial values in the first group examined. There was no good agreement between the concentrations of pentoxyfillin, its major pharmacologically active metabolite in blood plasma of pregnant women and the antiaggregation effect, which reaffirms the indirect action of this drug on platelets.
Significant interspecific differences in the pharmacokinetics of noopept are manifested by a decrease in the drug elimination rate on the passage from rats to rabbits and humans. Very intensive metabolism of noopept was observed upon intravenous administration in rats. In these animals, presystemic elimination mechanisms lead to the formation of a specific metabolite representing a product of drug biotransformation hydroxylated at the phenyl ring. In rabbits, unchanged noopept circulates in the blood for a longer time upon both intravenous and peroral introduction, biotransformation proceeds at a much slower rate, and no metabolites analogous to that found in rats are detected. The noopept pharmacokinetics in humans differs from that in animals by still slower elimination and considerable individual variability. No drug metabolites are found in the human blood plasma, probably because of a relatively small dose and low concentration.
The pharmacokinetics of a new psychotropic drug bemithil was studied after single and long-term administrations under monotherapy and in combination with phenazepam in patients with asthenoneurotic disturbances. A high degree of correlation for some pharmacokinetic parameters of bemithil following the administration of the drug test dose and its global therapeutic effect was established.
There were found significant differences in the pharmacokinetics of a new psychotropic drug with nootropic action ONC-10 following a long-term administration to the offspring of the intact animals as compared with the pharmacokinetics after a single administration: an increase of the drug content in plasma with a simultaneous decrease of clearance and the volume of distribution. A long-term administration of ONC-10 to the offspring of the alcoholized animals led to a decrease of the plasma drug level that was related to not only an increase of the constant of elimination and clearance of the drug but also an enhancement of the processes of its biotransformation and the appearance of the main metabolite.
The pharmacokinetics of a new Soviet-made immunostimulant kemantane, a derivative of adamantine, was studied by gas-liquid chromatography in patients with bronchial pathology. It was found that in the blood of the patients kemantane was not practically detected due to a pronounced effect of the "first pass" through the liver. Kemantane kinetics was evaluated by the kinetics of its major active metabolite. On the basis of the performed studies the scheme of using kemantane in the patients with decreased immunity was recommended.
