Large-scale purification and characterization of the Escherichia coli rep gene product.

We report a procedure for the large-scale purification of the Escherichia coli Rep protein, a helicase that is involved in the replication of the E. coli chromosome as well as a number of single-stranded bacteriophages. The procedure starts with E. coli cells harboring an overproducing plasmid, pRepO, in which the E. coli rep gene is under transcriptional control of the inducible lambda PL promoter (Colasanti, J., and Denhardt, D. T. (1987) Mol. Gen. Genet. 209, 382-390). The purification procedure results in greater than 98% pure Rep protein, which is free of contaminating nuclease activity, with yields of 40-50 mg of Rep protein/50 g of induced MZ-1/pRepO cells. We also show that cell death occurs upon inducing such a large overproduction of the E. coli Rep protein in MZ-1/pRepO. The Rep protein purified by this procedure has high specific single-stranded DNA-dependent ATPase activity, as well as helicase activity, with an apparent 3' to 5' directionality. The extinction coefficient of purified E. coli Rep protein is epsilon 280 = 1.16 +/- 0.04 ml mg-1 cm-1 (8.47 +/- 0.28 X 10(4) M-1 cm-1) in 10 mM Tris (pH 7.5), 20% (v/v) glycerol, 0.10 M NaCl at 25 degrees C. The solubility properties of the purified Rep protein have been examined as a function of glycerol, NaCl, MgCl2, ATP, and ADP concentrations at 25 and 37 degrees C (pH 7.5). Rep protein solubility decreases significantly with decreasing concentrations of glycerol and monovalent salt and increasing temperature; however, the presence of 1.5 mM ATP or ADP or MgCl2 at low NaCl concentrations increases the solubility. At 4 degrees C, in the presence of 20% glycerol and greater than or equal to 50 mM NaCl, the free Rep protein exists as a stable monomer under all conditions examined (+/- ATP and +/- MgCl2). The single-stranded DNA-dependent ATPase activity decreases with increasing glycerol concentration, such that in 25% (v/v) glycerol it has approximately 40% of its activity as compared to solutions that contain no glycerol. The dependence of the single-stranded DNA-dependent ATPase activity on salt concentration for a series of monovalent salts indicates the presence of both cation and anion effects, with decreasing activity in the order glutamate greater than acetate greater than chloride. The ability to obtain highly purified E. coli Rep protein in large quantities with relative ease will greatly facilitate physical characterizations of the protein and its interactions with DNA.

[Comparative studies of the effect of mictonorm (propiverin hydrochloride) and Spasuret (flavoxate hydrochloride) on the bladder detrusor muscle].

46 patients suffering from urgency/urge incontinency were treated with Mictonorm and Spasuret in a crossing-over study. In consideration of the placebo effect in this disease both agents were tested versus a non-verum. The application period was 4 weeks and the dosage was 45 mg/d Mictonorm and 300 mg/d Spasuret, respectively. Both with Mictonorm and with Spasuret a significant reduction of micturition frequency and an increase of the compliance could be observed, whereas the placebo was ineffective. A markedly growth of the maximal bladder capacity (16.9%) was obtained only by Mictonorm. Both agents keep likewise to an improvement of the symptoms or urgency/urge incontinency. Side effects could observed in a small size without a breaking-off of the treatment.

[One-step urethroplasty--indications, results and surgical peculiarities].

In a representative patient group the long-term results were examined after one-stage urethroplasty with a subcutaneous pedicle skin flap. The used modification in which an intact strip of epithelium is buried shows good to satisfactory results in 80% of the cases, but in more than 60% a diverticulum of the neourethra was found. Therefore, in 25% of all urethroplasties a secondary resection of the diverticulum was necessary. First experiences with the use of a skin patch to reconstruct the urethra have shown that formation of diverticula can be prevented by this technique. The pre-, intra- and postoperative measures (epilation, secretion drainage, temporary urinary diversion) which are necessary to achieve good functional results are presented in detail.

Follow-Up Studies

[Personal experiences with phenoxybenzamine (Dibenzyran) in the treatment of functional disorders of bladder emptying].

The influence of phenoxybenzamine on micturition was examined in 46 patients with functional bladder neck obstruction. Due to blockade of the alpha-adrenergic receptors at the bladder neck an increase of the maximal flow rate from a mean value of 13.6 to 24.6 ml s-1 was found during the treatment. In addition, there was a relaxation of the functional urethral length and a reduction of the maximal urethral closure pressure from 96.3 to 74.4 cm H2O. The most significant changes resulting from the application of Dibenzyran were observed in the neuropathic bladder with a reduction of the functional urethral length from 24.3 to 20.5 mm and a decrease of Pura max from 100.5 to 82.7 cm H2O. Furthermore, we achieved an increase of the maximal flow rate from 13.4 to 22.5 ml s-1.

[Transurethral electrostimulation in the therapy of postoperative disorders of bladder emptying following extended hysterectomy].

As a suitable method of the treatment of postoperative voiding disturbance following gynaecological surgery (hysterectomy) the indirect electrostimulation of detrusor vesicae is presented. It is possible with this method to treat the congenital (myelomeningocele) and the obtained (spinal cord dysfunction) but also the disturbances following hysterectomy successfully. In cases with such a hypotonic bladder the stimulation of the bladder wall is made with an intermittent application of exponential current so that sufficient contractions to the voiding will occur after the treatment.

[Operative measures in recurrence of urinary incontinence and advanced displacement of pelvic organs in the female].

The individual approach in the stress incontinence of the woman requires the consideration of general pathological and social factors, the locoregional topical anatomy of the pelvic organs, results of functional investigations and aimed metaphylactic measures. The possibility of selecting incontinent women for the abdomino-vaginal approach is demonstrated with the help of a model of diagnostics and the tactics of operation as well as metaphylactic measures are described. In 92% of the women with highly pathological topical anatomy of the pelvic organs or recidivations of incontinence of urine a continence could be proved on an average three years after operation. 25% of the female patients showed a component of urgent desire to pass water. The combined approach with application of chorium material contains three operative suspensory steps and the correction of further factors evoking incontinence. The technique tested on account of the results obtained is an alternative method if indication is given.

Developments in infants' search for displaced objects.

In an exploratory study of infants' search for displaced objects, 13-month-olds and 21-month-olds were tested on three kinds of displacement problems--visible displacements, invisible displacements, and transpositions--as well as single-hiding problems. Two aspects of performance on the displacement problems were distinguished: (1) searching within the locations involved in the displacement rather than at a control location, and (2) selecting between the displacement locations. Both the younger and the older infants were able to select the correct location on visible displacement problems and to search within the displacement locations on invisible problems. Neither age group was able to solve the transposition problems, but the older infants did at least search within the relevant locations on those problems. Age differences appeared to be due primarily to improved skill at identifying relevant locations rather than to improvements in selecting among those locations. Other factors contributing to the age differences in performance were a decrease in response biases and an increase in skills for coping with multiple possibilities.

[Therapy of detrusor hyperactivity with propiverin (mictonorm)].

In the context of treating hyperactivity of the detrusor the newly developed preparation Propiverin (Mictonorm) is presented. It is postganglionic and anticholinergic and mainly acts peripherally and selectively on the bladder. At the same time a musculotropic relaxing effect with a specific affinity to the unstriated musculature of the bladder can be demonstrated in experiments on animals. In our urodynamic investigations the application of the medicament resulted in an increase in bladder capacity, a reduction of urge symptoms, a decrease in bladder tension and a depression of the uroflow. These results justify the clinical use of Propiverin after appropriate urodynamic diagnostic checks.

[Significance of phenacetin abuse in urology].

With the help of the biotransformation the dangerousness of phenacetine is explained. The sequels of the phenacetine abuse for the urinary system are demonstrated. Conditioned by the increase of the consumption of analgesics during the next years in the GDR must be reckoned with an increased number of diseases of drug-conditioned chronic interstitial nephritis and tumours of the urinary tracts.
