Molecular study of the germinal reversions induced at the white-ivory locus in Drosophila melanogaster.

The white-ivory somatic mutation test of Drosophila melanogaster is based on the reversion of the X-linked eye colour recessive mutation white-ivory to wild-type. Although the exact mechanism of white-ivory reversion is not quite understood, it has been suggested that such reversion, both in somatic and germ-line cells, could be due to the precise excision of the tandemly duplicated 2.96 kb DNA fragment characteristic of the white-ivory mutation. We have attempted to confirm this hypothesis analysing, at the molecular level, different germinal revertants induced by chemical treatment with three well known alkylating agents: ethyl methanesulphonate, methyl methanesulphonate and N-nitroso-N-ethylurea. The molecular analysis of these germ-line revertants, using Southern blot hybridization and polymerase chain reaction techniques, shows that such reversions are associated with the deletion of the 2.96 kb tandemly duplicated DNA sequence of the white-ivory locus.

Adhesion of K88ab to guinea pig erythrocytes: effect on membrane enzyme activities.

Nontoxigenic Escherichia coli strains bearing K88 fimbriae have been associated with diarrhea in piglets. We have used guinea pig erythrocytes as a model of the host cell to study the cellular alterations after adherence of purified K88ab fimbriae. Although Mg2+-dependent ATPase was inhibited (up to 61%), Na/K ATPase was not. Metabolic enzymes were not significantly affected.

Gene toxin patterns of Escherichia coli isolated from diseased and healthy piglets.

DNA probes specific for genes coding for the heat stable enterotoxins (ST-I and ST-II), the heat labile enterotoxins (LT-I and LT-II), Shiga like cytotoxins and for the enterohemorrhagic factor (EHF), were used to examined 150 fecal Escherichia coli isolates from both diarrheic and healthy piglets. Thirty seven percent of the isolates hybridized with the LT-I probe, seventy one of them did so with the LT-II probe, while seventy six percent of the isolates possessed genes coding for the heat stable enterotoxins. No SLT-I positive hybridization was found and four percent of the isolates possessed SLT-II genes.

Involved factors in the intrafamilial spread of hepatitis C virus.

To investigate the risk of non-parenteral HCV infection, sera from 302 relatives of 120 anti-HCV positive subjects were tested for the presence of anti-HCV antibodies. For the sake of comparison, sera from 17,000 blood donors were also assayed. The prevalence of HCV positivity was 4.3% in household contacts, compared to 0.78% in the donor population, indicating a significantly higher risk of infection for family members. Close personal contact may not be as critical a factor for infection as is duration of the disease.

Unusual serotypes of Escherichia coli from piglet diarrhea in north Spain.

Eighteen strains of Escherichia coli isolated from piglet diarrhea were examined with a wide range of antisera and were found not to belong to the common serotypes found in this disease. Several different serotypes were found; nine strains belonged to O153, and an O group (not commonly associated with piglet diarrhea) belonged to four different serobiotypes.
