Adsorption of arsenic(V) by activated carbon prepared from oat hulls.

The efficiency of self-manufactured activated carbon (AC) produced from oat hulls in adsorbing arsenic(V) was tested in a batch reactor. The results indicated that the adsorptive capacity of AC was affected by initial pH value, with adsorption capacity decreasing from 3.09 to 1.57 mg As g(-1) AC when the initial pH values increased from 5 to 8. A modified linear driving force model conjugated with a Langmuir isotherm was created to describe the study's kinetics. The test results show that rapid adsorption and slow adsorption exist simultaneously when AC is used to remove arsenic(V).

Enhanced brain levels of 8,12-iso-iPF2alpha-VI differentiate AD from frontotemporal dementia.

OBJECTIVE: To quantify the isoprostane 8,12-iso-iPF2alpha-VI in brain tissues obtained from patients with AD, patients with frontotemporal dementia (FTD), and controls. BACKGROUND: Enhanced brain oxidative stress with secondary damage to cellular macromolecules may play a role in the pathogenesis of AD and FTD. The isoprostane 8,12-iso-iPF2alpha-VI is a specific and sensitive marker of in vivo lipid peroxidation and is increased in AD. METHODS: Levels of this isoprostane were determined by gas chromatography/negative ion chemical ionization mass spectrometry. RESULTS: Levels of 8,12-iso-iPF2alpha-VI were markedly elevated in both frontal and temporal cortex of AD brains compared to the corresponding areas of FTD and controls. No significant difference in brain 8,12-iso-iPF2alpha-VI levels for any regions considered was observed between FTD and controls. CONCLUSIONS: Lipid peroxidation is a feature of AD, but not FTD. The generation of 8,12-iso-iPF(2alpha)-VI in the brain is not a general or final common pathway of neurodegeneration, but may be relatively specific for disease processes in AD and not FTD.

Ammonia inhibition on thermophilic aceticlastic methanogens.

The inhibition effects of total ammonia nitrogen (TAN) on aceticlastic methanogenic activity using biomass from thermophilic anaerobic reactors were investigated in this study. Anaerobic Toxicity Assays (ATA) were conducted after the biomass was acclimated to different levels of TAN. The TAN background concentrations in the reactors were 400, 1,200, and 3,050 mg/L. The results from ATA showed: 1) high TAN concentrations could cause inhibition of aceticlastic methanogens; 2) biomass acclimated to higher TAN concentrations could alleviate the inhibition effect due to the increase of TAN concentration; 3) the lethal TAN concentration for methanogens was approximately 10,000 mg/L regardless of the background TAN concentration; 4) ATA results also revealed the role played by pH. At a given TAN concentration, methanogenic activity varied with the pH values. The highest methanogenic activity was always observed at a pH of 7.0-7.5. 5) It was also observed that acclimation could increase the pH tolerance range, which made methanogens less sensitive to pH changes.

Biohydrogen production as a function of pH and substrate concentration.

The conversion of organics in wastewaters into hydrogen gas could serve the dual role of renewable energy production and waste reduction. The chemical energy in a sucrose rich synthetic wastewater was recovered as hydrogen gas in this study. Using fractional factorial design batch experiments, the effect of varying pH (4.5-7.5) and substrate concentration (1.5-44.8 g COD/L) and their interaction on hydrogen gas production were tested. Mixed bacterial cultures obtained from a compost pile, a potato field, and a soybean field were heated to inhibit hydrogen-consuming methanogens and to enrich sporeforming, hydrogen-producing acidogens. It was determined that the highest rate (74.7 mL H2/(L*h)) of hydrogen production occurred at a pH of 5.5 and a substrate concentration of 7.5 g COD/Lwith a conversion efficiency of 38.9 mL H2/(g COD/L). The highest conversion efficiency was 46.6 mL H2/(g COD/L).

Palladium-catalyzed cross-coupling reactions of in situ generated allylindium reagents with aryl halides.

[reaction: see text] In situ generated allylindium reagents from the reaction of 1 equiv of indium with 1.5 equiv of allyl halides could be effective cross-coupling partners in palladium-catalyzed cross-coupling reactions to aryl halides. The best results were obtained with 2% Pd(2)dba(3)CHCl(3) and 16% Ph(3)P in the presence of 3 equiv of LiCl in DMF at 100 degrees C.

Intratracheal priming with ovalbumin- and ovalbumin 323-339 peptide-pulsed dendritic cells induces airway hyperresponsiveness, lung eosinophilia, goblet cell hyperplasia, and inflammation.

Dendritic cells (DC) are the primary APC responsible for the capture of allergens in the airways and the shuttling of processed allergens to the draining lymph nodes where Ag presentation and T cell activation take place. The mechanism of this Ag handling and presentation in asthma is poorly understood. In addition, the feasibility of asthma induction by DC priming has not been directly tested. In this report an asthma model using intratracheally (i.t.) injected splenic DC was used to address these issues. DC pulsed with a model Ag OVA or the major MHC class II-restricted OVA T epitope peptide OVA(323-339) and instilled i.t. primed mice to exhibit asthma-like diseases. With OVA as the Ag, mice exhibit airway hyperresponsiveness (AHR), lung eosinophilia and inflammation, and pulmonary goblet cell hyperplasia. In OVA(323-339)-immunized mice, AHR and goblet cell hyperplasia were noted, with little eosinophilia and parenchymal inflammation. The latter finding provides evidence for dissociating AHR from eosinophilia. In both cases mediastinal node hypertrophy occurred, and high levels of Th2 cytokines were produced by the lung and mediastinal lymph node cells (LNC). Interestingly, mediastinal LNC also produced high levels of Th1 cytokines. Lung cells produced much less Th1 cytokines than these LNC. These results demonstrate that DC when introduced i.t. are potent in inducing asthma-like diseases by recruiting lymphocytes to the lung-draining lymph nodes and by stimulating Th2 responses and also suggest that the lung environment strongly biases T cell responses to Th2.

Development of anaerobic migrating blanket reactor (AMBR), a novel anaerobic treatment system.

A novel anaerobic treatment system, the anaerobic migrating blanket reactor (AMBR), was developed after completing a parallel study with upflow anaerobic sludge blanket (UASB) and anaerobic sequencing batch reactor (ASBR) processes. Using sucrose as the main component of a synthetic wastewater, the AMBR achieved a maximum chemical oxygen demand (COD) loading rate of 30 g.l-1.day-1 at a 12-h hydraulic retention time (HRT). This resulted in a standard methane production rate (SMPR) of 6.51.l-1.day-1 and an average methane-based COD (MCOD) removal efficiency of 62.2%. A key element in granular biomass formation was migration of the biomass blanket through the reactor. Although a carbohydrate-rich wastewater was used, no separate pre-acidification was required for the AMBR, because of high mixing intensities and wash out of acidogenic bacteria. In contrast, the absence of pre-acidification created "bulking" problems (caused by abundant acidogenic bacteria at the surface of granules) in a UASB reactor, operated under conditions similar to that of the AMBR. As a result, a maximum COD loading rate and SMPR of 21 g.l-1.day-1 and 4.91.l-1.day-1 were achieved, respectively, for the UASB reactor at a 12-h HRT. These values were 18 g.l-1.day-1 and 3.71.l-1.day-1, respectively, for an ASBR at a 12-h HRT. Hence, the performance of the AMBR in treating a carbohydrate-rich wastewater was found to be superior in terms of maximum loading rate and SMPR.

Microsatellite diversity and the demographic history of modern humans.

We have examined differences in diversity at 60 microsatellite loci among human population samples from three major continental groups to evaluate the hypothesis of greater African diversity in this rapidly evolving class of loci. Application of a statistical test that assumes equal mutation rates at all loci fails to demonstrate differences in microsatellite diversity, while a randomization test that does not make this assumption finds that Africans have significantly greater microsatellite diversity (P < 10(-8)) than do Asians and Europeans. Greater African diversity is most apparent at loci with smaller overall variance in allele size, suggesting that the record of population history has been erased at repeat loci with higher mutation rates. A power analysis shows that only 35-40 microsatellites are needed to establish this difference statistically, demonstrating the considerable evolutionary information contained in these systems. On average, African populations have approximately 20% greater microsatellite diversity than do Asian and European populations. A comparison of continental diversity differences in microsatellites and mtDNA sequences suggests earlier demographic expansion of the ancestors of Africans.

An allograft kidney showing both features of IgA nephropathy and membranous glomerulonephritis--a case report.

We report a case of glomerular disease with both mesangial IgA and subepithelial IgG deposits in the allograft kidney. The patient was a 36 year-old man who had received a renal allograft 1 year previously. Fifteen days before admission, he discovered a microscopic hematuria without clinical evidences of allograft rejection. Light microscopy showed diffuse increase of mesangial matrix without mesangial cell proliferation. Capillary walls were diffusely and mildly thickened. Immunofluorescence microscopy demonstrated both granular deposits of IgA in the mesangium and IgG along the capillary walls. On electron microscopy, electron-dense deposits were identified not only in the mesangium but also on the epithelial side of the glomerular basement membrane.

Initial clinical experience with a single pass VDDR pacing system.

Although ventricular rate adaptive pacing (VVIR) improves exercise capacity and cardiac output compared to constant rate ventricular pacing (VVI), this pacing mode does not provide benefit of atrioventricular (AV) synchrony. We evaluated the use of a custom-built VDDR pacing system using a single pass, ventricular lead, which detects endocavity P wave using a pair of diagonally arranged atrial bipolar (DAB) electrodes. In the VDDR mode, AV synchrony is enabled and the P wave rate is used in conjunction with an accelerometer based activity sensor for rate adaptive pacing. A VDDR pacemaker was implanted in three patients with complete AV block (mean age 63 +/- 1 year) and the mean implantation time was 29 minutes. Mean P wave amplitude was 2.4 mV (1.2-4.2 mV) at implantation and telemetered P wave amplitude was stable over a follow-up of 6 months. At a sensitivity of 0.2 mV, stable P wave sensing was observed during breathing maneuvers, arm swinging, myopotential induction, and Holter recording. Paired exercise tests performed in the VDDR and VVIR modes showed higher cardiac output at rest, during exercise, and in the recovery period in the VDDR pacing mode. Thus VDDR pacing using a single pass lead is superior to VVIR pacing by enabling P synchronous ventricular pacing without adding to the complexity of implantation.

Endothelin receptors in human placenta: relationship to vascular resistance and thromboxane release.

This investigation was performed to study the potential role of endothelin in the modulation of fetoplacental vascular resistance in the human placenta. Full-term placentas from uncomplicated pregnancies were studied within 30 min of delivery. The umbilical artery and vein to a single placental cotyledon were cannulated and the artery perfused with RPMI media (0.82 ml/min). Endothelin 1 caused a sustained dose-dependent increase in perfusion pressure. Infused endothelin 1 (50 nM) stimulated thromboxane release 2.3-fold compared with basal values. Thromboxane release persisted for 15 min after discontinuation of endothelin. Properties of human placental endothelin 1 receptors were defined in binding studies performed on a crude membrane fraction of placental cotyledons. Binding was saturable, reached steady state by 3 h at 25 degrees C, and was linear with protein concentration. Scatchard analysis of binding data indicated a single class of high-affinity binding sites with a Kd of 36.1 +/- 9.7 pM and a density of 185.4 +/- 9.6 fmol/mg protein (n = 5). The potency order for competitive inhibition of the binding of 125I-labeled endothelin 1 was endothelin 1 greater than endothelin 2 = endothelin 3 = sarafotoxin S6b greater than big endothelin (human) = big endothelin (porcine). Phenylephrine, bradykinin, norepinephrine, atrial natriuretic factor, diltiazem, U46619, and angiotensin II did not displace 125I-endothelin 1 from its receptors. These experiments demonstrate that endothelin 1 is a potent pressor substance in the human fetoplacental cotyledon. Pressor effects of endothelin may be mediated by a combination of direct effects and stimulation of vasoconstrictor prostanoids.

Anaerobic migrating blanket reactor treatment of low-strength wastewater at low temperatures.

The feasibility of the compartmentalized anaerobic migrating blanket reactor (AMBR) was studied for the treatment of low-strength soluble wastewater under low-temperature conditions. During an operating period of 186 days, a 20-L AMBR was fed nonfat dry milk substrate as a synthetic wastewater at low temperatures (15 and 20 degrees C). The concentration of the influent was constant at chemical oxygen demand (COD) and 5-day biological oxygen demand (BOD5) concentrations of 600 and 285 mg/L, respectively. The soluble COD (SCOD) removal efficiency was 73% at the end of the operating period (15 degrees C) at a 4-hour hydraulic retention time (HRT), while the total COD (TCOD) removal efficiency was 59%. At a 4-hour HRT, staged conditions promoted complete removal of propionic acid in the final compartments of the reactor. The specific methanogenic activity of granules increased slowly until the end of the operating time, improving the removal rate. Biomass was retained effectively, as evidenced by the solids retention time (SRT) that was always greater than 50 days even during step decreases of the reactor HRT from 12 hours to 4 hours. A long SRT also promoted system stability during changes in flow, which was observed by SCOD removal efficiencies staving greater than 70%. During a hydraulic stress test, the HRT was reduced from 4 hours to 1 hour for one day (24 HRTs) in which volatile suspended solids (VSS) in the effluent increased from an average background level of 8.7 g/d to 35 g/d and the SRT decreased from 50.5 days to 12.6 days. However, mixed liquor volatile suspended solids concentration decreased only by 1 g/L, and hence a similar COD removal efficiency and biogas production was found one day after the hydraulic stress (as compared to one day before the hydraulic stress).
