[A canine model of pancreas transplantation recipient following total pancreatectomy and associated lymphoproliferation].

An attempt was made to use a diabetic dog as a model recipient of pancreas transplantation. The diabetic state was induced by total pancreatectomy, and was shown to be stable by oral and intravenous glucose tolerance tests. In dogs that underwent total pancreatectomy under isoflurane anesthesia (GOI), blastogenesis of peripheral blood lymphocytes had a tendency to be reduced in comparison with no-treated dogs until 2 weeks after surgery. However, from 3 weeks after surgery, their responses became comparable to those of non-treated dogs. These results suggest that stable diabetes mellitus is induced, in dogs, by total pancreatectomy, and that this animal is a useful model of a pancreas transplantation recipient.

[Effect of habitual physical activity on physical fitness and serum cholesterol in middle-aged male workers].

The effects of habitual physical activity on physical fitness parameters and serum cholesterol profile were studied in middle-aged male workers. The subjects were 3376 middle-aged men (age: 30-59 yrs) who worked at two enterprises in HIROSHIMA Prefecture. As for physical fitness parameters, grip strength, vertical jump, side step, and step test scores were measured. Atherogenic index was calculated as (total Chol-HDL Chol)/(HDL Chol). Because daily physical activity mainly consists of physical activities during leisure time and working time, by using a questionnaire, physical activity during leisure time was assessed and categorized by frequency into three categories: 'frequently', 'sometimes', and 'seldom': and physical activity during work was categorized as 'high' or 'low' according to the average walking time per day during work. The effect of these physical activities on physical fitness parameters and serum cholesterol profile was evaluated by analysis of covariance (PC-SAS: GLM procedure) adjusting for several confounding factors such as age, body mass index, type of job, smoking and drinking habits. The main results are summarized as follows: 1) Physical activity during leisure time was positively related to vertical jump, side step, and step test score, independent of physical activity during working time, and 2) physical activity during leisure time was negatively associated to LDL-C and AI in the group with 'low' physical activity during work. These results show that in middle-aged male workers, even moderate or light physical activity during leisure time has some beneficial health promoting effects through changes in physical fitness and serum cholesterol profile.

[Cigarette smoking and physical fitness].

In this review, previous studies regarding the effects of cigarette smoking on parameters of physical fitness such as endurance performance, muscle strength, step test score, body mass index, blood pressure, serum lipoprotein profile, etc., have been summarized with a special reference to the quality of life for middle-aged and elder men, including the result of recent studies by the present authors. Additionally the hypothesized mechanisms of cigarette smoking to atherosclerosis have been shown. It is concluded mainly from our recent studies that: 1) the smoking has an effect to impair not only the aerobic power, but also the anaerobic power, 2) the abstention from chronic smoking would be expected to improve the parameters of physical fitness, and 3) even though the duration from the onset of smoking is less than one year, i.e., a relatively short-term smoking habit, the smoking has a serious damage to the several parameters of physical fitness.

Changes in blastogenic responses of lymphocytes and delayed type hypersensitivity responses after vaccination in dogs.

To clarify the immunologic effects of vaccination in dogs, we monitored total leukocyte and lymphocyte counts, humoral antibody responses, blastogenic responses of lymphocyte, and delayed type hypersensitivity (DTH) responses after vaccination. Mixed vaccines were administered on day 0 except for canine parvovirus (CPV) vaccine which was readministered on day 21. The puppy and adult dogs had a significant decrease in leukocyte and lymphocyte counts on day 7. The puppies showed a significant increase in the blastogenesis of lymphocytes after each vaccination, whereas the adult dogs had no significant changes. However, the adult dogs were divided into two groups, high responders and low responders in blastogenesis of lymphocytes. The dogs with higher or lower response in SI values on day 0 tended to show decrease or increase after the first vaccination, respectively. Since almost all dogs developed high titers of humoral antibody, it is considered that vaccination acts in an immunomodulative fashion. DTH responses to phytohemagglutinin (PHA) and CPV vaccine monitored at 0, 3, and 8 weeks after the first vaccination produced strong reactions, in particular those to CPV vaccine rose significantly after vaccination and maintained the higher responses for at least 2 months. These results suggest that DTH responses to PHA and CPV vaccine are helpful to monitoring non-specific and specific immune functions in vivo, therefore, DTH could be used as simple and rapid immunologic tests in canine practice.

Immunosuppression in dogs by pretransfusions with ultraviolet (UV)-irradiated whole blood.

Ultraviolet (UV)-irradiated whole dog blood prevented the initiation of proliferative responses in allogeneic mixed lymphocyte reactions (MLR). When dogs were given 4 weekly intravenous transfusions of UV-irradiated allogeneic donor whole blood, peripheral blood lymphocytes (PBL) of the recipients responded less significantly to donor PBL in MLR for over one week after the final transfusion. Red blood cell (RBC)-crossmatching of these dogs was negative. Dogs treated with UV-irradiated blood did not produce anti-donor PBL antibody, or IgG, IgM and C3 determined by the indirect Coombs test. These dogs also had negligible delayed type hypersensitivity (DTH) responses to donor PBL at the end of the treatment period, recognized as slightly suppression of skin graft rejection. In contrast, six dogs receiving injections of untreated allogeneic whole blood evidenced high proliferation in MLR, agglutination in RBC-crossmatching, acute rejection of skin grafts and three of them produced C3 in the Coombs test.

The effect of lectin from Taro tuber (Colocasia antiquorum) given by force-feeding on the growth of mice.

In earlier experiments in our laboratory, a lectin from the Kintoki bean (Phaseolus vulgaris) was found to have not only erythrocyte agglutinating activity but also toxicities for mice and rats, including growth inhibitory activity and even lethal activity. A number of studies on legume lectins have been carried out in other laboratories as well. But relatively little attention has been paid to lectins from non-leguminous foods. In the present study, we chose Taro tuber as a source of non-leguminous lectins and prepared two types of Taro tuber lectin. One was crude lectin precipitated with ammonium sulfate from the aqueous extract and the other was pure lectin isolated as we described previously. The two were compared with regards to the antinutritional functions in mice. The daily doses were 100 mg for either intact or autoclaved crude lectin, which was a maximum amount available to give to mice in 1 ml, and 30 mg for the pure lectin which was equivalent to 100 mg of the crude lectin in hemagglutinating activity. Control mice were given 1 ml of water and the experiment was conducted for 6 days. Growth retardation was found in the mice given either lectin, but no significant difference was found in the weight increase between the control group and the autoclaved lectin group. For 3 days during the experimental period, physical activity was measured as an index of vigor of mice. The activities of the crude lectin and the pure lectin groups leveled down to 62.9 and 64.2% of that of the control group, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)
