Scanning electron microscopy of rat erythrocytes during chronic alcoholic intoxication combined with protein and vitamin deficit.

Electron microscopy revealed morphological changes in erythrocytes from rats with chronic alcoholic intoxication kept on protein- and vitamin B-deficient rations. All animals had anisopoikilocytosis (up to 50%). Most pronounced changes in erythrocyte population attesting to accelerated erythrocyte aging (stomato- and microcytosis, discocyte swelling, and spontaneous hemolysis) were found in alcohol-fed rats kept on deficient ration.

[Ultrastructural changes in the aortic arch intima in rats with various resistance to emotional stress].

Experiments were performed on 24 male Wistar rats weighing 210 +/- 14 g. Depending on the activity in the open field test, all rats were divided into behaviorally active and passive subgroups. Water-immersion stress (2 hrs) followed by 2-hrs rest period was used as the model of acute emotional stress. Tissue samples from aortic ascending arch were taken from control and stressed rats immediately after decapitation and used for further electron microscopy. The number of micropinocytosis vesicles along the free and basal layers of the cytoplasm and the area of mitochondria were determined. Emotional stress was shown to intensify metabolic processes involving micropinocytosis vesicles between the aortic lumen and its wall. Passive rats display the most severe alterations induced by stress (damages of aortic endotheliocytes, partial regeneration of the plasmalemma by membranes of micropinocytosis vesicles, and decrease in the average area of mitochondria). Morphological equivalents of the initial stages of emotional stress-induced formation of fibrous changes in the aortic arch intima are believed to be found. Such changes can lead to the appearance of atherosclerotic patches.

Telemedicine: capabilities of telecommunications in clinical practice in Russia.

Successful implementation of telemedicine services depends on an adequate telecommunications infrastructure. At present the main infrastructure in Russia and the countries of the former Soviet Union consists of analogue telephone lines. These are sufficient for e-mail and access to the World Wide Web but do not provide sufficient bandwidth for the transmission of realtime teleconsultations. ISDN lines and fibre optic cables are becoming more widespread in the major cities but have yet to be installed in the regions.

[The mapping of the local content of water and dry matter by using ultrathin frozen sections].

The possibility and applications of quantitative dry mass and water content mapping using an image analysis of darkfield intensity digitalization were shown on ultrathin freeze-dried cryosections. The distribution map obtained may be further used both for direct measurements of local dry mass and water content under physiological state and pathological disorders and for calculation of the concentration values obtained by quantitative X-ray mapping on freeze-dried cryosections in terms of millimoles per litre of cellular water.

[Reparative processes in postischemic cerebral cortex in the early postresuscitation period (materials submitted to the International Symposium, "Central nervous system and postresuscitation pathology," Moscow, March 14-16, 1989)].

Dynamics of neuronal plasticity in rat postischemic cerebral cortex in the early postresuscitation period following complete ischemia was studied by electron microscopic methods. In this period destructive processes have proved to run parallel with reparative ones in the postischemic cerebral cortex. The phenomenon of true regeneration of cell processes in the cerebral neurons engendering growth cones observed by the third hour of the post-resuscitation period is of particular interest. Subsequently, these signs lead to the formation of new synaptic interneuronal links and new neuronal nets in the cerebral cortex. The data obtained open up new prospects for understanding the pathogenesis of postresuscitation encephalopathies.

[The digital processing of the images in x-ray microanalysis].

Image analysis of X-ray microanalytical maps of sodium, potassium, phosphorus, chloride and calcium was carried out on the IBAS image analysis system (OPTON, FRG). In the present work, algorithms were developed for getting a new semiquantitative information from the binary images of maps: the element profile on the chosen line, the isoconcentration mapping and the multiple elemental mapping. It is possible to get as many as eight different elemental maps on the same image (with 8-bit resolution of pixel) and to extract the pixels with element overlappings.

[Changes in the ionic composition and shape of erythrocytes in response to cholera toxin].

Blood of 10 inbred rats has been investigated after effect of cholera toxin for 1 h by means of roentgenospectral energetic spectrometer Revex-5100 (USA). Simultaneously, erythrocytes have been studied by means of the scanning electron microscope. Contents of sodium, phosphorus, chlorine, serum, potassium, and ferrum ions have been estimated. The cholera toxin changes the contents of the ions mentioned in erythrocytes and contributes to certain changes of their form. A suggestion is made that the cholera toxin changes permeability of the erythrocytes membranes without participation of the cell cyclase systems.

[Scanning electron microscopy and x-ray microanalysis of the small intestine of suckling rabbits exposed to cholera toxin].

Surface alterations and elemental distribution in small intestinal enterocytes of suckling rabbits were studied by scanning electron microscopy and x-ray microanalysis. The latter was performed on 10 mu thick freeze-dried cryosections. Cholera toxin was found to diminish the sodium and chloride content in the epithelial layer from the cryptae towards the tops of the villi, while in control animals, no difference in the elemental distribution was revealed. The changes in the enterocyte surface consisted in the disarrangement of the microvilli and appearance of bullous enlargements on their tops.

Simple PC-based system for morphometric analysis of synapses.

The computer system designed for synaptic morphometry of aksosomatic and aksospine synapses of brain is described in the present paper. It is based on an AT-comparable personal computer equipped with low-cost frame grabber. This hardware configuration allows to input images from any TV source such as TV camera, videorecorder for further processing. The appropriate software was written in Microsoft Quick Basic to measure the main morphometric parameters of axon ending, dendritic spines, total contact, active zone and mitochondria. Number of synaptic vesicles (total and active) were also counted using a mouse as a pointing device. The derivative parameters (vesicle density, mitochondrial density) are then calculated. All measured data are stored in ASCII format, allowing ease in editing and export into other application programs. Statistical evaluation and calculation of histograms is performed by associated program also written in Quick Basic. The advantages and disadvantages of this approach are discussed.

[The effect of the simulation of a prolonged dive to 100 m in a helium-oxygen environment on human erythrocytes].

Three volunteers participated in the experiments to study the osmotic resistance and elemental composition of the erythrocytes during a 100-meter simulated saturation dive in He-O2 environment. The 15-day hyperbaric exposure resulted in a significant increase of erythrocytes resistance in response to a changed osmotic environment which was also maintained at an early stage of readaptation. The X-ray spectral local microanalysis revealed a decrease in chlorine, potassium and sodium level's on the 2nd postdecompression day. On a prolonged hyperbaric exposure at the expense of phospholipid exchange between plasma erythrocytes and lipoproteins, the erythrocyte membrane changes which may cause disturbances in ionic permeability of the cells are expected to occur.

[A morphometric study of the ultrastructure of the axosomatic synapses in the sensorimotor cortex of rats administered the delta-sleep inducing peptide].

Qualitative estimation and quantitative morphometry of ultrastructural changes in layers III, IV, and V in rat sensomotorial cortex after delta-sleep-inducing peptide administration, provides an evidence of multiplication of active inhibitory synapses induced by DSIP. The newly activated synapses may form an axo-somatic synaptic pool participating in optimization of the balance of excitement and inhibition processes in sensomotorial cortex.
