[Influence of carbohydrate substrate feeding rate on productivity of recombinant Escherichia coli strain].

Effect of different regimes of glucose feeding on growth rate and recombinant protein synthesis of recombinant E. coli was investigated. It was estimated that the selected regime of substrate feeding provided enhanced finished product yield up to 40-50 per cent. Changes in feeding regime caused significant changes in the quantity and pattern of the glucose metabolism by- products.

Biotechnology↗

[Development of technology for isolation and purification of gentamicin using sorption without filtration].

A technology for isolation and purification of gentamicin from nonfiltered fermentation broth with the use of sorption-pulsory columns was developed as a result of the research work. The practical application of the technology will increase the yield from 30 to 57 per cent and reduce the content of the minor admixtures in the final product up to 5 per cent in comparison to the currently used process.

[Analogs of human proinsulin. Obtaining a deletion at the site of the proinsulin gene encoding the region of receptor binding].

A strain producing 5-del-proinsulin was designed on the basis of an industrial strain producing human recombinant proinsulin. 5-Del-proinsulin is an analog of human recombinant proinsulin. The new strain contains a deletion of 5 amino acid residues in the C-end region of the B-chain and a residue of tyrosine at B25. A method of oligonucleotide-directed mutagenesis was applied. Comparison of the electrophoregrams of the inclusion bodies of the initial and resulting strains made it possible to conclude that the pattern of the proteins was the same and the electrophoretic mobility of the recombinant proteins was practically identical.

Amino Acid Sequence↗

[Consumption of amino acids by Escherichia coli--a producer of recombinant proteins].

The dynamics of amino acid consumption from the medium by Escherichia coli 1864, a strain producing recombinant protein was studied. It was shown that the strain actively used glutamate and aspartate from the medium, which was determined by the leading role of the amino acids in nitrogen metabolism. The strain also consumed threonine, glycine and alanine capable of effectively providing the culture with metabolic energy.

[Experimental study of human recombinant insulin].

The experimental study of recombinant human insulin revealed its high specific activity. The kinetics of the hypoglycemic effect of various dosage forms of the insulin developed at the National Research Centre of Antibiotics and the dosage forms manufactured by Eli Lilly was similar. The pharmacological investigation of the dosage forms showed that they had no side effects on the vitally important organs and systems. It was concluded that the dosage forms of the gene engineered human insulin developed at the National Research Centre of Antibiotics did not differ by their characteristics from the analogous dosage forms manufactured in other countries.

[Derivatives of C-branched monosaccharides studied by 13C NMR].

13C NMR spectra of some 3-C branched D-allofuranoses and D-ribofuranoses were obtained and interpreted. The impact of attaching the alkyl substitute to the monosaccharides on chemical shifting of the adjacent carbon atoms was shown. The experimental data are useful for elucidating structures of analogous compounds by 13C NMR.

[Exopolysaccharide formation during the cultivation of Bacillus polymyxa on different carbohydrate substrates].

The effect of the carbohydrate source in the cultivation medium on the intensity of the synthesis of exopolysaccharides of two variants of Bacillus polymyxa was studied. No significant effect of the nature of the carbohydrate source on accumulation of the culture biomass was observed, while the fermentation broth viscosity, the yield of the extracellular polysaccharides and their monosaccharide composition, as well as the ratio of the neutral and acid components significantly changed. Differences in the composition and properties of the exopolysaccharides produced in the cultures of the variant strain under identical conditions were noted. Certain correlation between the exoproduct yield and the quantity of the accumulated biomass, as well as between the composition of the exopolysaccharides and viscosity of the fermentation broth was shown, which ensures a goal-oriented effect on the synthesis of the exopolysaccharide complex by variation of the medium composition.
