[Analysis of genomic polymorphism in leptospira by polymerase chain reaction with random primers].

A test system for genetic typing of Leptospirae is developed, based on the polymerase chain reaction (PCR) with arbitrary primers. Thirteen strains of 4 Leptospira species were examined: L. interrogans, L. parva, L. illini, and L. inadai. Analysis of polymorphism of amplicon length (PAL) by the PCR with short Sh1 and Sh2 primers revealed genotypical differences at the inter- and intraspecias levels, as well as at the subserovar level. PAL of L. interrogans strains Rga and M-20, serovars icterohaemorrhagiae and copenhageni, were identical both with Sh1 and Sh2 primers. Moreover, PCR with Sh2 primer showed genotypical similarity between strains Moscow V and M. oeconomus, serovar grippotyphosa of the same serogroup. Analysis of PAL by the PCR with long Lgn1 and Lgn2 primers showed similar results. Analysis of the PAL values obtained by the PCR with all primers permitted us to differentiate 9 L. interrogans strains into 8 PAL genotypes and identify a different degree of genotypical relation between strains of different serovars of this species. Complete genotypical relationship between strains L. inadai N 10 and EMJH 86 (serovar lyme) was confirmed by the new test system. Therefore, PCR-based test system with different primers can be used to differentiate between closely related Leptospira strains and to investigate the genotypical relationships between the strains at the intra- and interspecies and inter- and subserovar levels.

[The theoretical and practical aspects of the nonspecific prevention of infectious diseases among the troops].

As possible alternative to a complex of sanitary-hygienic measures, directed on the infectious diseases prevention, not specific prophylaxis with the aid of immunomodulative preparations is offered. Prospects of the immunomodulative preparations application is defined by transition to popularized epidemiological thinking planned in modern conditions. Strategy of total and selective not specific prophylaxis, results of own researches on estimation of preventive efficiency of dibazolum and prodigiosanum during acute respiratory disease and virus hepatitis A are described.

[Selective inhibition of DNA amplification in nonadhesive cultures of Mycoplasma pneumoniae].

Inhibition of amplification of various genome regions of Mycoplasma pneumoniae was observed in the polymerase chain reaction, and was dependent on cultivation conditions. A protein stably associated with DNA is responsible for the inhibitory effect. It is assumed that when the protein selectively associates with separate DNA regions, it can inhibit genes encoding pathogenicity factors, thus promoting mycoplasma transformation into persistent variants.

[Test-systems of polymerase chain reaction for determining the tuberculosis pathogen].

Two polymerase chain reaction-based test systems were used to identify Mycobacteria tuberculosis by using a clinical material for practical health purposes. The former test system strictly specifically enables M. tuberculosis to be detected. The latter also allows one to differentiate a BCG vaccine strain from the M. tuberculosis. The test systems were tested by using 11 clinical sputum samples, three of them were taken from patients with fibrocavernous tuberculosis, two from those with focal tuberculosis and one from that with infiltrative one, 5 from those with non-specific pulmonary diseases (chronic bronchitis and asthma). In addition, one serum sample from a patient with fibrocavernous tuberculosis was examined. All tests obtained from patients with tuberculosis were positive, whereas those from patients with non-specific pulmonary diseases were negative. This suggests that differentiation of BCG vaccine strain from M. tuberculosis can be used both in model experiments and in the study of clinical material lysates.

[The ultrastructure of a new species of Chlamydia--Chlamydia pneumoniae].

The dynamic study of a new Chlamydia species, C. pneumoniae (strain TWAR, isolate TW-480), inoculated into the monolayer culture of cells L-929 was made 24, 48, 72 and 96 hours after inoculation. When compared with C. trachomatis and C. psittaci, C. pneumoniae were found to stand between these two species with respect to the morphology of their intracytoplasmic microcolonies (inclusions): they were round, almost bubble-like, but more densely packed with chlamydiae, surrounded by an undulate membrane, preserving its integrity until the late stages of their development cycle. In cells L-929 C. pneumoniae had a typical development cycle accompanied by the formation of vegetative and spore-like cells, reticular and elementary bodies, as well as intermediate cells, though this process was slower than in C. trachomatis and C. psittaci. Besides normal elementary bodies, many altered ones were formed in the process of the development of C. pneumoniae in cells L-929. Most of these alterations were similar to the process of bacterial L-transformation and could be regarded as the manifestation of chlamydial pathology related to the adaptation to new host cells.

[The efficacy of exogenous and endogenous interferonization in preventing influenza].

In this work the results of using interferon (IFN), Dibasol and the combination of these preparations for the urgent prophylaxis of influenza and acute respiratory diseases (ARD) among the employees of the Gamaleia Research Institute of Epidemiology and Microbiology (USSR Acad. Med. Sci.) are summarized. Reaferon and Dibasol decrease ARD morbidity 2 times and leukocytic IFN decreases it 1.4 times, while the combined administration of Dibasol and IFN has proved to be ineffective. Reaferon is mainly a prophylactic remedy; it has been found to bring about almost no decrease in the number of patients at the peak of morbidity, while pronouncedly decreasing it in two weeks after the administration of the preparation. Dibasol has a curative effect, sharply interrupting the beginning rise of morbidity. Reaferon normalizes the characteristics of the IFN status, decreasing the amount of circulating IFN and enhancing the capacity of leukocytes for producing alpha-IFN and gamma-IFN. For the prophylaxis of respiratory infections the use of Reaferon is advisable 3-4 weeks prior to the beginning of the epidemic and then, when the first cases of infection are registered, the course of prophylaxis with Dibasol should be carried out.

[A mathematical model of the growth of a population of Legionella pneumophila in the presence of Tetrahymena pyriformis protozoa].

The mathematical model describing the dynamics of the growth of L. pneumophila in aqueous environment in the presence of protozoa has been worked out. The model has demonstrated considerable heterogeneity of the initial population of virulent L. pneumophila strains. The number of bacteria capable of multiplication in Infusoria is no more than 0.1% of the initial population. The time of the generation of the infective agent inside Tetrahymena pyriformis is 2.8 hours.

[The creation of a test system for detecting Mycoplasma pneumoniae based on a method of directed DNA amplification].

A system for the rapid detection of M. pneumoniae, the causative agent of pneumonia in humans, has been developed. This system is based on the amplification of M. pneumoniae chromosomal DNA sequences in the polymerase chain reaction (PCR). The use of two primer sets, for nucleotide sequences of adhesion protein P1 and for nucleotide sequences of variable regions of the 16S ribosomal RNA, has permitted the detection of individual M. pneumoniae cells. The application of this technique for the study of simulated clinical material has shown that PRC is a sensitive and reliable assay and may be useful for the early detection of M. pneumoniae in infectious clinical material (blood and sputum samples).

[A method for the passive agglutination of polymer dispersions for the diagnosis of legionellosis].

The method of synthetizing dispersions of gelatin-modified polyacrolein microspheres 1.5-2.5 micrograms in diameter, used as a solid-phase carrier for the preparation of immunodispersion diagnostica, has been developed. The possibility of using immunodispersion diagnosticum in the passive agglutination test with hyperimmune rabbit sera has been demonstrated. High activity and specificity of immunodispersion diagnosticum, combining methodological simplicity and rapidity characteristic of agglutination, has been shown.

[The genomic fingerprinting of the causative agents of sapronoses].

The genome polymorphism of the causative agents of sapronoses (Vibrio cholerae, Legionella and Leptospira) has been studied. The use of the method of genome fingerprinting [correction of dactyloscopy] has been shown to permit the differentiation of closely related strains of such causative agents. The epidemically significant strains of the causative agents of sapronoses, isolated in different geographical regions, have been found to be genotypically related, i.e., they are probably of clonal origin. Avirulent and nontoxigenic strains are genotypically heterogeneous and differ both from one another and from epidemically significant strains. Using V. cholerae as an example, the hypothesis of the appearance of potentially dangerous variants at the epidemic period in the absence of their release at the period between epidemics is considered.

[Social significance of Mycoplasma infections. Prospects of the research].

The paper analyzes the data on the prevalence of human Mycoplasma infections in the USSR and on their diagnostic methods. It also provides evidence for the pathogenicity and biological properties of M. incognitus, a new species, and defines the ways and prospects for further investigations in theoretical and practical Mycoplasma research.

[The splitting of the acceptor proteins of the protein kinase system in eukaryotic cells by Legionella cytolysin].

In the process of protein kinase reaction carried out in the mixture consisting of tris-HCl buffer, EDTA, MgCl2, gamma-32P-ATP and the cytoplasmic fraction of rabbit pulmonary cells the phosphorylation of proteins with molecular weights of 150 and 55 kD took place. The addition of L. pneumophila culture fluid to the reaction mixture resulted in the splitting of phosphorylated proteins with the formation of the component having a molecular weight of 45 kD. These disturbances in protein kinase reaction were found to occur due to the involvement of Legionella cytoplasm, a previously characterized protein with a molecular weight of 37 kD, into the process. In this connection, the participation of cytolysin in the pathogenesis of Legionella infection may also be considered from the viewpoint of the effect produced by cytolysin on the regulatory processes affecting the metabolism of target cells.

[Serological and epidemiological characteristics of acute pneumonias caused by Mycoplasma pneumoniae in Belgorod].

The etiology of the outbreaks of acute pneumonia in Belgorod was established and their epidemiological features were studied. The mycoplasmal etiology of all cases of acute pneumonia in children and adults, appearing alongside acute respiratory infections resulting from the preceding outbreaks caused by influenza viruses A/Prague/, B/Leningrad/369/75, and all types of parainfluenza viruses, was shown. The droplet mechanism of the transfer of infection was established, which was confirmed by the severity of the outbreak and a high rate of infection of the medical staff at hospitals for children and adults, where patients with mycoplasma-induced pneumonia were treated.

[The immune structure of occupational groups of the population in relation to Legionella pneumophila serogroup 1].

The immune structure of different professional groups with respect to L. pneumophila has been studied. A wide spread of Legionella infection among subway construction workers, subway personnel and railroad conductors has been established, which makes it possible to regard these professions as high risk groups susceptible to Legionella infection. The risk factors for this group are as follows: work in the atmosphere of high dust content with air supply by means of air conditioners, contact with soil in the process of earthwork. The bacteriological study of water samples obtained in dining cars has resulted in the isolation of two strains of L. pneumophila, serogroup I, which confirms the possibility of cases of Legionella infection among railroad personnel.
