STUDIES ON HUMAN ANTIBODIES. 3. AMINO ACID COMPOSITION OF FOUR ANTIBODIES FROM ONE INDIVIDUAL.

Antilevan, antidextran, antiteichoic acid, anti-"A" and normal gammaG, all isolated from a single human, as well as pooled gammaG and gammaM, were analyzed for amino acid content. Differences ranging from 7 to 32 residues were found among these antibodies in glycine, valine, leucine, tyrosine, arginine, lysine, and threonine content. These conspicuous differences in amino acid composition were not correlated with the Gm type of these antibodies. Hydroxylysine was found in two of the four antibodies, antidextran and antiteichoic acid, and in gammaM- immunoglobulin. No hydroxylysine was found in gammaG- globulins. These findings probably account for discrepancies in the literature concerning the occurrence of this residue in antibody protein.

A STUDY OF THE CROSS-REACTIVITY OF ANTIPURIN-6-OYL SERUM WITH DEOXYRIBONUCLEIC ACID (DNA).

The specificity of the reaction of antipurin-6-oyl sera with thermally denatured DNA has been studied by means of hapten inhibition techniques. The relative order of effectiveness of various haptens as inhibitors of the complement fixation reaction between DNA and antipurin-6-oyl serum was found to be comparable to their relative order of effectiveness as inhibitors of the precipitin reaction between purin-6-oyl-protein conjugates and antipurin-6-oyl serum. Antipurin-6-oyl antibody has been purified and has been shown to be capable of reacting with thermally denatured DNA. It is concluded that the reactivity of these purine-specific antisera with DNA is a property of antibody with specificity for the purin-6-oyl moiety. In addition, the reaction between antipurinoyl sera and DNA has been demonstrated by the techniques of radioimmunoelectrophoresis and passive cutaneous anaphylaxis.

PURINE- AND PYRIMIDINE-SPECIFIC ANTIBODIES: EFFECT ON THE FERTILIZED SEA URCHIN EGG.

Antiserums from rabbits immunized with several purine- and pyrimidine-containing albumin conjugates can penetrate and become fixed within cells of Arbacia punctulata. All of these specific antiserums were found to inhibit embryonic development of the fertilized sea urchin egg. The stage of arrestment of development was dependent in part upon the dilution of antiserum, and also upon the nature of its specificity. It is suggested that the observed results reflect the interaction of these antiserums with singlestranded DNA within the cell.

Purification of antibody to galactosyl-protein conjugates.

A rapid procedure for purification of rabbit antibody to beta-D-galacto-sylphenylazo-bovine serum albumin is described. Antibody was precipitated with a heterologous antigen and was then dissociated from the precipitated complex with hapten. The antigen was partially removed by pH adjustment, and antibody was separated from residual antigen and from hapten by partition chromatography on Sephadex G-25. The antibody, obtained in 50 to 55 percent over-all yield, was 96 percent reactive with homologous antigen. It appeared to consist, of 95 percent of a single component, based upon physicochemical measurements.
