Multiple sclerosis--relation between HLA haplotype A25, B18 and disease progression.

The HLA typing of 100 patients with multiple sclerosis (MS) and their differentiation according to the dynamics of the disease, the type of course and the degree of defect showed the following results: a significant increase in frequency of HLA B7 (P less than 0.05), independent of the severity of illness, a 4-fold increased frequency of the HLA haplotype A25, B18 with augmented occurrence in slight to moderate severity of the disability degree (P less than 0.05), less so in the dynamics of the disease, and only partially so in the relapsing-remittent course. These HLA constellations provide only a hint at the prognosis of MS; the findings could be an expression of the heterogeneity of the disease.

[Immunostaging of patients with chronic pancreatitis. I. A study of humoral and cell-mediated immunity (author's transl)].

Immunostaging was performed in patients with chronic pancreatitis of different etiologic groups. Immune responifveness was assessed by immunoglobulin serum levels, non specific autoantibody formation, C3 levels, antistreptolysin reaction, antibody titer against Toxoplasma gondii, E. coli, Herpes simplex-, Mumps- and Cytomegalovirus and screening for soluble immune-complexes (PEG precipitation assay), number of T- and B-lymphocytes, lymphocyte responsiveness to PHA, PPD, tetatoxoid and mumps-antigen in a whole blood assay, skin testing with candidin, trichophytin, tuberculin and streptokinase and HLA-typing. In some cases we found a depressed cell-mediated immune responsiveness. Patients were divided into two groups based on this main finding. Cellular hyporesponsiveness was correlated to the severity of the disease and a high incidence of HLA-B5 but not to any etiologic factor of the disease.

[3H-thymidine paper strip modification of the lymphocyte transformation test. III. Whole blood lymphocyte mixed culture using a cryopreserved stimulating cell mixture].

In comparison to the classical mixed lymphocyte culture (MLC) method a MLC technique using whole blood cultures of responding cells and a pool of prepared stimulating cells from 10 healthy blood donors is described. The reproducibility of cryopreservation of stimulating lymphocytes was proven by means of cell electrophoresis.

[Sperma- and HLA-antibody demonstration in sterility].

Sperma antibodies were detected by means of passive haemagglutination, the immobilisation test and immunofluorescence in the serum of 16 females from among 150 infertile married couples. These serums (experimental group) and 16 serum samples containing no sperma antibodies (control group) were investigated for the presence of cytotoxic HLA-antibodies against lymphocytes from their married partners and from 20 selected donors by means of the micro-lymphocyte toxicity test. HLA-antibodies against the lymphocytes of the husbands involved were found in four women from the experimental group. Two of these four serums also reacted with 20% of the test lymphocytes. In the control group, lymphocyte-toxic antibodies against test subjects were also found in all three serums which cross-reacted with the lymphocytes of the husbands. No new diagnostic or therapeutic criteria for the treatment of sterility can be derived from the combined humoral sperma and cytotoxic HLA-antibodies in the serum.

Application of teem-test for investigations of cellular immune phenomena in multiple sclerosis.

Some evidence is presented for usableness of tanned erythrocytes in electrophoretic mobility (TEEM)-test. Two possibilities of application of TEEM-test for immunological investigation in multiple sclerosis are discussed: detection of lymphocyte sensitization against a soluble antigen (3 M KCl extracted) derived from a normal brain and measurement of mixed lymphocyte reactin (MLR) after a short-time lymphocyte culture. Results show some connection between autosensitization (TEEM-test using normal tissue antigen) and predisposition (TEEM-MLR-test).

[Immunologic donor-recipient matching in living kidney transplantation].

The better results of transplantation in kidneys of relatives in contrast to cadaveric kidneys are above all traced back to a compatibility in the HLA-D-Locus which is more frequently present in relatives and which can be established by a lacking stimulation in the mixed culture of lymphocytes. Therefore, the selection of donor and recipient in transplantations of living kidneys where there is no pressure of time should apart from the serological standardization of lymphocytes should also comprise the mixed culture of lymphocytes lasting 5 to 7 days.

[HLA-antigens in patients with rheumatoid arthritis].

109 patients with rheumatoid arthritis (RA) from the north of the GDR were HLA typed and the antigen frequencies compared with those of a control group. Even in our patients the HLA-DR4 association was demonstrable. But in all studies DR4 association is not strong enough for taking HLA typing as a diagnostic test for RA. There was an immunogenetic heterogeneity in correlation to side effects of therapy with sodium aurothiomalate or D-penicillamine. HLA-DR5 was significantly increased in patients with side effects in comparison to patients without side effects in therapy. Before recommending HLA-DR typing for selection of high risk patients in D-penicillamine therapy these results should be proven in a large patient group or in a second study.

[HLA antigens in cleft lip-palate patients].

HLA typing of 50 patients with cleft lip and/or cleft palate (50 HLA-ABC and 35 HLA-DR tests) and, in part, also of their families showed an increased frequency of the antigens HLA-AII, DRw6 and the haplotype HLA-A11, B35 as against the normal population. Disorders of wound healing were observed more frequently in patients with HLA antigens A11 and B15 than in those without these antigens. Based on these hints of an HLA association (p values after correction are not significant), and the frequent coincidence of HLA-DR antigen in the patients' parents, as well as the increased rate of HLA-DR homozygosity of the patients, it is assumed that genetic factors of the HLA complex or an HLA-linked complex play a part in the etiology of clefts of the lip and cleft palate.
