Effect of temperature on the retention of ionizable compounds in reversed-phase liquid chromatography: application to method development.

The analysis of pharmaceutical compounds is often a difficult challenge which requires mathematical tools to improve the quality of the separation method. This work is an attempt to rationalize the anomalous variation of the logarithm of the retention factor with temperature in case of ionizable compounds. The effect of temperature on ionizable compounds was studied within a large range of temperature, ranging from 30 to 130 degrees C. The determination of the so-called chromatographic pKa and the study of its variation with temperature allow to explain why the forms of the van't Hoff curves are so different depending on the type of solute, the type of buffer and the type of the mobile phase. A retention model along with a computation procedure is proposed to optimize both temperature and mobile phase composition and to provide good and robust conditions as shown by illustrative examples.

A study of interlaboratory influence on column evaluation.

A liquid chromatography method for the characterization of base deactivated columns was investigated in a collaborative study involving six laboratories. This work was carried out on two chromatographic supports (Xterra RP 18 and Symmetry Shield). Different cooling systems, namely water bath and air oven, were tested and it was shown that column thermoregulation did not significantly influence chromatographic data. In order to control the mobile phase composition, the latter was prepared by weight rather than volume. Thanks to the injection of a set of selected neutral compounds, extra-column effects were evaluated in each of the participating laboratories. The results showed that chromatographic supports tested in different laboratories and following the same test protocol could be effectively compared.

Some advantages of high temperature for the separation of pharmaceutical compounds with mass spectrometry detection.

The beneficial effects of high temperature on separation and detection of basic compounds, the detection being performed by MS via ESI, are investigated. The influence of various parameters on both separation and detection performances is studied. These parameters include the mobile phase pH, the temperature, and the type of stationary phase. Experiments are performed under gradient elution conditions. The results obtained with four different supports, silica-, zirconia-, carbon-, and polymer-based columns, are compared by means of different criteria including the elution composition, the peak asymmetry, and the S/N. High temperature liquid chromatography at high pH with volatile buffers suitable for MS detection was shown to be an interesting choice for solutes with basic sites.
