Quantification of phase synchronization phenomena and their importance for verbal memory processes.

In the past years, interest in brain oscillations and their possible role in perceptual and cognitive processes has greatly increased. The two oscillations that have received the most attention are the theta and the gamma rhythm. In this study, the functioning and properties of phase synchronization parameters for these two frequency bands estimated by means of Gabor expansion were demonstrated with simulations for the phase-locking index (PLI) and the 1:1 as well as n:m phase synchronization indices. In order to demonstrate the importance of phase synchronization phenomena for memory performance, power, PLI and the 1:1 as well as n:m phase synchronization indices were calculated for EEG data on verbal memory encoding. These parameters showed various dissociations for recalled versus not-recalled nouns. In particular, the calculation of phase synchronization among different frequencies either at the same electrode or at different electrodes provided a completely new picture of dynamic neuronal interaction accompanying memory processing.

New concepts of multiple tests and their use for evaluating high-dimensional EEG data.

Recently, new concepts of type I error control in multiple comparisons have been proposed, in addition to FWE and FDR control. We introduce these criteria and investigate in simulations how the powers of corresponding test procedures for multiple endpoints depend on various quantities such as number and correlation of endpoints, percentage of false hypotheses, etc. We applied the different multiple tests to EEG coherence data. We compared the memory encoding of subsequently recalled and not recalled nouns. The results show that subsequently recalled nouns elicited significantly higher coherence than not recalled ones.

Increased neuronal communication accompanying sentence comprehension.

The main purpose of this study was to examine large-scale oscillatory activity and frequency-related neuronal synchronization during the comprehension of English spoken sentences of different complexity. Therefore, EEG coherence during the processing of subject-subject (SS)- and more complex subject-object (SO)-relatives was computed using an adaptive fitting approach of bivariate auto-regressive moving average (ARMA) models which enabled the continuous calculation of coherence in the course of sentence processing with a high frequency resolution according to the dynamic changes of the EEG signals. Coherence differences between sentence types were observed in the theta (4-7 Hz), beta-1 (13-18 Hz) and gamma (30-34 Hz) frequency ranges, though emerging during the processing of different parts of these sentences: gamma differences were evident mainly during the relative clause while theta and beta-1 differed significantly following the end of the relative clause. These findings reveal no simple one to one map between EEG frequencies and cognitive operations necessary for sentence comprehension. Instead, they indicate a complex interplay and dynamic interaction between different EEG frequencies and verbal working memory, episodic memory, attention, morpho-syntactic and semantic-pragmatic analyses, which though distinct often co-occur.

Multivariate tests for the evaluation of high-dimensional EEG data.

In this paper several multivariate tests are presented, in particular permutation tests, which can be used in multiple endpoint problems as for example in comparisons of high-dimensional vectors of EEG data. We have investigated the power of these tests using artificial data in simulations and real EEG data. It is obvious that no one multivariate test is uniformly most powerful. The power of the different methods depends in different ways on the correlation between the endpoints, on the number of endpoints for which differences exist and on other factors. Based on our findings, we have derived rules of thumb regarding under which configurations a particular test should be used. In order to demonstrate the properties of different multivariate tests we applied them to EEG coherence data. As an example for the paired samples case, we compared the 171-dimensional coherence vectors observed for the alpha1-band while processing either concrete or abstract nouns and obtained significant global differences for some sections of time. As an example for the unpaired samples case, we compared the coherence vectors observed for language students and non-language students who processed an English text and found a significant global difference.

Interferons in enteroviral heart disease: modulation of cytokine expression and antiviral activity.

Interferon (IFN)-beta has a more than 120-fold higher antiviral activity than the closely related IFN-alpha in human myocardial fibroblasts infected with the cardiotropic enterovirus coxsackievirus B3 (CVB3). CVB3 replication induces interleukin (IL)-6 and IL-8 expression in myocardial fibroblasts, and suppresses the expression of monocyte chemoattractant protein-1 (MCP-1). We investigated whether the higher antiviral activity of IFN-beta compared to IFN-alpha was a result of a suppression of IL-8 expression by IFN-beta since previous studies had indicated that IL-8 stimulates enterovirus replication. Human myocardial fibroblasts were treated with either IFN-alpha, IFN-beta or IFN-gamma (0, 10, 100, or 1,000 IU/ml) and the concentrations of IL-6, IL-8 and MCP-1 were measured in culture supernatants by immunoassays. Both IFN-beta and IFN-gamma reduced IL-6 and IL-8 expression significantly. In addition, neutralization of IL-8 in culture supernatants of myocardial fibroblasts using a monoclonal antibody demonstrated a significant reduction of CVB3 titers. Antiproliferative effects of all three IFNs were very low (<30% with 1,000 IU/ml), indicating that the suppression IL-6 and IL-8 was not related to cytotoxicity. MCP-1 expression was increased only by high concentrations of IFN-gamma (1,000 IU/ml). By contrast, IFN-alpha had no significant effect on IL-6, IL-8 and MCP-1 expression. In conclusion, suppression of IL-8 expression is an "immuno-modulating" feature of IFN-beta in human myocardial fibroblasts, which is similar to the activity of IFN-gamma. This feature of IFN-beta contributes to its high antiviral activity against CVB3 and may be useful in the treatment of enteroviral heart disease.

Cerebral information transfer during word processing: where and when does it occur and how fast is it?

Different regions of the brain have to interact to perform language processing. Such neural integration processes can be studied by measuring synchronization of oscillations. Coherence is the best known algorithm to study synchronization between two sites of the cortex with regard to selected oscillation frequencies. Phase coherence quantifies phase synchronization, in particular. In addition, cross phase (or simply phase) gives information about the direction and speed of the spread of oscillations. We use a new method to study short-time phases between different sites of the cortex in order to explore transient neural networks during word processing. Particularly, processing of abstract and concrete nouns was investigated by dynamic phase analysis of the alpha1 frequency band (8-10 Hz). Abstract and concrete nouns turned out to induce different dynamic networks of information transfer. Whereas processing of concrete nouns excites a widespread network for about 800 msec, the interhemispheric interactions during abstract noun processing are restricted to 300-500 msec after word presentation. Further, the direction of cerebral information transfer differs for the two word categories. Additionally, it could be shown that the propagation speed of information is slower for concrete nouns than for abstract nouns.

The contribution of EEG coherence to the investigation of language.

The contribution of EEG coherence analysis to the investigation of cognition and, in particular, language processing is demonstrated with examples of recent EEG studies. The concept of EEG coherence analysis is explained, and its importance emphasized in the light of recent neurobiological findings on frequency-dependent synchrony as a code of information processing between nerve cell assemblies. Furthermore, EEG coherence studies on naturally spoken and written word and sentence processing are reviewed and experimental results are presented giving new insights into the occurrence of "transient functional language centers" within the brain.

Detergent extraction identifies different VirB protein subassemblies of the type IV secretion machinery in the membranes of Agrobacterium tumefaciens.

The VirB/D4 type IV secretion system of Agrobacterium tumefaciens translocates virulence factors (VirE2, VirF, and the VirD2-T-DNA complex) to plant cells. The membrane-bound translocation machinery consists of 12 proteins (VirB1-11 and VirD4) required for substrate translocation. Protein-protein interactions in the membranes were analyzed after extraction with the mild detergent dodecyl-beta-d-maltoside followed by separation under native conditions. Incubation of the membranes with increasing concentrations of the detergent differentially extracted virulence proteins. Separation of the solubilized proteins by blue native electrophoresis revealed cofractionation between two classes of protein complexes containing VirB7. The first class, consisting of major T-pilus component VirB2 and associated proteins VirB5 and VirB7, comigrated in the low molecular mass portion of the gel of about 100 kDa. The second class contains putative translocation complex core components VirB8, VirB9, and VirB10 in the high molecular mass portion of the gel larger than 232 kDa, as well as VirB7. Solubilized proteins were characterized further by gel filtration chromatography. This procedure separated T-pilus-associated proteins VirB2, VirB5, and VirB7 in the low molecular mass range from the other components of the translocation machinery and the substrates VirE2 and VirD2. Fractionation of VirB7-containing complexes (VirB7-VirB7 homodimers and VirB7-VirB9 heterodimers) suggested that they may link the T-pilus components to the core of the translocation machinery. Based on previously described VirB protein interactions and biochemical analysis of C58 wild type as well as of virB5 and virB6 deletion mutants, a model of T-pilus assembly in A. tumefaciens is suggested.
