A Mo(VI) alkylidyne complex with polyhedral oligomeric silsesquioxane ligands: homogeneous analogue of a silica-supported alkyne metathesis catalyst.

A highly active alkyne metathesis catalyst is realized by replacing the amide ligands of a molybdenum(VI) trisamide alkylidyne complex with silanol groups from incompletely condensed POSS (polyhedral oligomeric silsesquioxane) ligands. This catalyst serves as an effective homogeneous mimic of an amorphous silica-supported catalyst. Reactivities of various catalytic mixtures are reported along with an X-ray structure of the aniline-coordinated amidodisiloxymolybdenum(VI) alkylidyne complex.

Crystal structures of Aedes aegypti alanine glyoxylate aminotransferase.

Mosquitoes are unique in having evolved two alanine glyoxylate aminotransferases (AGTs). One is 3-hydroxykynurenine transaminase (HKT), which is primarily responsible for catalyzing the transamination of 3-hydroxykynurenine (3-HK) to xanthurenic acid (XA). Interestingly, XA is used by malaria parasites as a chemical trigger for their development within the mosquito. This 3-HK to XA conversion is considered the major mechanism mosquitoes use to detoxify the chemically reactive and potentially toxic 3-HK. The other AGT is a typical dipteran insect AGT and is specific for converting glyoxylic acid to glycine. Here we report the 1.75A high-resolution three-dimensional crystal structure of AGT from the mosquito Aedes aegypti (AeAGT) and structures of its complexes with reactants glyoxylic acid and alanine at 1.75 and 2.1A resolution, respectively. This is the first time that the three-dimensional crystal structures of an AGT with its amino acceptor, glyoxylic acid, and amino donor, alanine, have been determined. The protein is dimeric and adopts the type I-fold of pyridoxal 5-phosphate (PLP)-dependent aminotransferases. The PLP co-factor is covalently bound to the active site in the crystal structure, and its binding site is similar to those of other AGTs. The comparison of the AeAGT-glyoxylic acid structure with other AGT structures revealed that these glyoxylic acid binding residues are conserved in most AGTs. Comparison of the AeAGT-alanine structure with that of the Anopheles HKT-inhibitor complex suggests that a Ser-Asn-Phe motif in the latter may be responsible for the substrate specificity of HKT enzymes for 3-HK.

MS2(Me2PC2H4PMe2)2 (M = Mo, W): acid-base properties, proton transfer, and reversible protonolysis of sulfido ligands.

The acid-base reactivity of MS(2)(dmpe)(2), where M = Mo (1) and W (2) and dmpe = Me(2)PCH(2)CH(2)PMe(2), was examined. Compounds 1 and 2 arise via the one-pot reaction of (NH(4))(2)MS(4) and dmpe. Protonation of these species gives the stable salts [MS(SH)(dmpe)(2)]X. The pK(a)'s of the Mo and W compounds are estimated to be 16.5 and 15.5, respectively. Protonation causes the M=S distances to diverge from 2.24 A to 2.06 and 2.57 A, whereas the Mo-P distances do not change appreciably. (1)H and (31)P NMR studies for [1H]BAr(F)(4) reveal that the proton exchange is competitive with the NMR time scale; at low temperatures, individual signals for both the parent disulfide and its conjugate acid can be observed. Treatment of 1 with excess HOTf liberates H(2)S to afford [MoS(OTf)(dmpe)(2)]OTf, which forms an adduct with CD(3)CN and regenerates 1 upon treatment with SH(-)/Et(3)N solutions. Consistent with its ready protonation, complex 1 is methylated, and the use of excess MeOTf gives [MoS(OTf)(dmpe)(2)](+) and Me(2)S in a rare example of double alkylation at a sulfido ligand.

Electron-rich diferrous-phosphane-thiolates relevant to Fe-only hydrogenase: is cyanide "nature's trimethylphosphane"?

The two-step one-pot oxidative decarbonylation of [Fe2(S2C2H4)(CO)4(PMe3)2] (1) with [FeCp2]PF6, followed by addition of phosphane ligands, led to a series of diferrous dithiolato carbonyls 2-6, containing three or four phosphane ligands. In situ measurements indicate efficient formation of 1(2+) as the initial intermediate of the oxidation of 1, even when a deficiency of the oxidant was employed. Subsequent addition of PR3 gave rise to [Fe2(S2C2H4)(mu-CO)(CO)3(PMe3)3]2+ (2) and [Fe2(S2C2H4)(mu-CO)(CO)2(PMe3)2(PR3)2]2+ (R = Me 3, OMe 4) as principal products. One terminal CO ligand in these complexes was readily substituted by MeCN, and [Fe2(S2C2H4)(mu-CO)(CO)2(PMe3)3(MeCN)]2+ (5) and [Fe2(S2C2H4)(mu-CO)(CO)(PMe3)4(MeCN)]2+ (6) were fully characterized. Relevant to the H(red) state of the active site of Fe-only hydrogenases, the unsymmetrical derivatives 5 and 6 feature a semibridging CO ligand trans to a labile coordination site.

Characterization of a diferrous terminal hydride mechanistically relevant to the Fe-only hydrogenases.

Reduction of diferrous dithiolato complexes with hydride donor reagents affords the first example of the previously elusive terminal diferrous hydride, [Fe2(edt)(mu-CO)(H)(CO)(PMe3)4]PF6 (edt = S2C2H4). Crystallographic characterization shows that this model contains an asymmetrical semi-bridging CO trans to a terminal hydrido, as indicated in the Hred state in the D. desulfuricans enzyme. The model reacts with protons to yield H2 and rearranges via an intramolecular process to the isomeric mu-hydrido isomer, which is unreactive toward protons.

Organo-tricyanoborates as tectons: illustrative coordination polymers based on copper(I) derivatives.

The first systematic study on the use of tricyanoborates as ligands is presented. The tricyanoborates [RB(CN)3]- (R = oct and Ph) can be prepared by direct cyanation of RBCl2 precursors as well as by thermolysis of the corresponding isocyanides [RB(NC)3]-. The first organo-cyanogallates [RGa(CN)3]- (R = Bu, C6H2-2,4,6-Me3) were prepared from the corresponding dichloride, the structure of Et4N[mesGa(CN3] being confirmed crystallographically. The reaction of equimolar [RB(CN)3]- (R = oct, Ph) and [Cu(MeCN)4]+ afforded two-dimensional polymers [RB(CN)3Cu(NCMe)]. The sheets arise via conjoined hexagonal B3Cu3(CN)6 rings with chair conformations. The reaction of excess [PhB(CN)3]- and [Cu(MeCN)4]+ gives the polymer [K(18-crown-6)]{Cu[PhB(CN)3]2}. Treatment of [PhB(CN)3]- with [Cu(PCy3)2(NCMe)x]PF6 gave the one-dimensional polymer [PhB(CN)3Cu(PCy3)2], wherein two of the three BCN substituents are coordinated.

Diferrous cyanides as models for the Fe-only hydrogenases.

The first systematic study of diferrous dicyano dithiolates is described. Oxidation of [Fe2(S2C2H4)(CN)2(CO)4](2-) in the presence of cyanide and tertiary phosphines and of Fe2(S2C2H4)(CO)4(PMe3)2 in the presence of cyanide affords a series of diferrous cyanide derivatives that bear a stoichiometric, structural, and electronic relationship to the H(ox)(air) state of the Fe-only hydrogenases. With PPh3 as the trapping ligand, we obtained an unsymmetrical isomer of Fe2(S2C2H4)(mu-CO)(CN)2(PPh3)2(CO)2, as confirmed crystallographically. This diferrous cyanide features the semibridging CO-ligand, with Fe-muC bond lengths of 2.15 and 1.85 A. Four isomers of Fe2(S2C2H4)(mu-CO)(CN)2(PMe3)2(CO)2 were observed, the initial product again being unsymmetrical but more stable isomers being symmetrical. DFT calculations confirm that the most stable isomers of Fe2(S2C2H4)(mu-CO)(CN)2(PMe3)2(CO)2 have cyanide trans to mu-CO. Oxidative decarbonylation also afforded the new tetracyanide [Fe2(S2C2H4)(mu-CO)(CN)4(CO)2]2-. Insights into the oxidative decarbonylation mechanism of these syntheses come from the spectroscopic characterization of the tetracarbonyl [Fe2(S2C2H4)(mu-CO)(CN)3(CO)3](-). This species reacts with PEt3 to produce the stable adduct [Fe2(S2C2H4)(mu-CO)(CN)3(CO)2(PEt3)](-).

Properties and reactivity of chlorovinylcobalamin and vinylcobalamin and their implications for vitamin B12-catalyzed reductive dechlorination of chlorinated alkenes.

Vitamin B12-catalyzed reductive dechlorination of perchloroethylene (PCE) and trichloroethylene (TCE) is a potential strategy for cleanup of polluted environments. Presented are crystal structures of vinylcobalamin 2 and cis-chlorovinylcobalamin 1. They show a strong resistance toward photolysis. Reduction of 2 is difficult, but reduction of 1 occurs readily and produces 2. The mechanism of this latter reaction involves acetylene as an intermediate. These and other findings are discussed in the context of environmental studies on B12-catalyzed dechlorination of PCE and TCE and investigations of the haloalkene reductive dehalogenases that catalyze similar reactions.

Microporous porphyrin solids.

Metalloporphyrins are exceedingly useful building blocks for the design and synthesis of molecularly based solids. The use of hydrogen bonding or metal ion coordination provides a wide range of framework solids. Using various polyfunctionalized porphyrins, we have created porous solids that are thermally robust and that retain their internal porosity upon loss of solvates. Their pore dimensions are comparable to zeolites, and they show shape and size selectivity in sorption of guest molecules and in epoxidation of alkenes.

[Fe2(SR)2(mu-CO)(CNMe)6]2+ and analogues: a new class of diiron dithiolates as structural models for the H(ox)Air state of the fe-only hydrogenase.

Low-temperature oxidation of Fe(2)(S(2)C(n)H(2n)(CNMe)(6-x)(CO)x (n = 2, 3; x = 2, 3) affords a family of mixed carbonyl-isocyanides of the type [Fe(2)(S(2)C(n)H(2n)(CO)x(CNMe)(7-x)](2+). The degree of substitution is controlled by the RNC/Fe ratio, as well as the degree of initial substitution at iron, with tricarbonyl derivatives favoring more highly carbonylated products. The structures of the monocarbonyl derivatives [Fe(2)(S(2)C(n)H(2n))(mu-CO)(CNMe)(6)](PF(6))(2) (n = 2,3) established crystallographically and spectroscopically, are quite similar, with Fe---Fe distances of ca. 2.5 A, although the mu-CO is unsymmetrical in the propanedithiolate derivative. Isomeric forms of [Fe(2)(S(2)C(3)H(6))(CO)(CNMe)(6)](PF(6))(2) were characterized where the CO is bridging or terminal, the greatest structural difference being the 0.1 A elongation of the Fe---Fe distance when MeNC (vs CO) is bridging. In the dicarbonyl species, [Fe(2)(S(2)C(2)H(4))(mu-CO)(CO)(CNMe)(5)](PF(6))(2), the terminal CO ligand is situated at one of the basal sites, not trans to the Fe---Fe vector. Oxidation of Fe(2)(S(2)C(2)H(4))(CNMe)(3)(CO)(3) under 1 atm CO gives the deep pink tricarbonyl [Fe(2)(S(2)C(2)H(4))(CO)(3)(CNMe)(4)](PF(6))(2). DFT calculations show that a bridging CO or MeNC establishes a 3-center, 2-electron bond within the two Fe(II) centers, which would otherwise be nonbonding.

Dihydrogen activation by a diruthenium analogue of the Fe-only hydrogenase active site.

The photochemical reaction of Ru2(S2C3H6)(CO)4(PCy3)2 (1) and H2 gives the dihydride Ru2(S2C3H6)(mu-H)(H)(CO)3(PCy3)2 (2). NMR and crystallographic studies reveal mutually trans basal phosphine ligands and both bridging and terminal hydrides. Ru2(S2C2H4)(CO)4(PCy3)2 behaves similarly. Other HX substrates undergo photoaddition to 1, affording Ru2(S2C3H6)(mu-H)(X)(CO)3(PCy3)2 for X = OTs (3a), Cl (3b), and SPh (3c). Treatment of Ru2(S2C3H6)(mu-H)(H)(CO)3(PCy3)2 with [H(OEt2)]BArF4 (ArF = B(C6H3-3,5-(CF3)2) in CD2Cl2 gives [Ru2(S2C3H6)(mu-H)(CO)3(PCy3)2(H2)]+ (4), which catalyzes H2-D2 exchange. The reaction of 2 with [D(OEt2)]BArF4 gave [Ru2(S2C3H6)(mu-H)(CO)3(PCy3)2(HD)]+ (JH-D = 31 Hz). These studies provide the first models for the Fe-only hydrogenases that bear dihydrogen and terminal hydrido ligands.

A robust microporous zinc porphyrin framework solid.

A robust microporous zinc(II) metalloporphyrin framework solid has been synthesized. The proposed structural model developed from X-ray single crystal data has an interpenetrated three-dimensional framework of zinc trans-biscarboxylate tetraarylporphyrins whose carboxylates coordinate the six edges of tetrahedral Zn(4)O(6+) clusters, maintaining a charge-neutral framework. This cubic framework has 74% free volume and 4 x 7 A pores. N(2) adsorption gives a type I isotherm with a surface area of 800 m(2)/g, which is greater than that of a typical zeolite. Experimental evidence indicates that the interpenetrated frameworks of the evacuated solid remain intact and retain a microporous structure. This is a versatile framework system: alteration of the metal in the porphyrin may create a catalytically active solid, and modification of the 10-, 20-substituents of the porphyrin can provide control over both the polarity and the size of the pores.

Radical polymerization initiated by Bergman cyclization.

The diradical generated by the Bergman cyclization of 3,4-benzocyclodec-3-ene-1,5-diyne is shown to initiate the radical polymerization of several monomers. Methacrylates are polymerized to high molecular weight by the diradical initiator much more efficiently than other monomers. The relation between the rate of polymerization and the degree of polymerization indicates that the polymer primarily propagates as a monoradical. This monoradical growth is in agreement with established theory predicting that diradical initiators can produce high polymer only through chain transfer followed by monoradical growth due to the rapid intramolecular termination of short diradical chains. In agreement with this mechanism, the polymerization rate of acrylonitrile initiated by the diradical is shown to increase by more than 20-fold upon addition of a chain transfer agent. Small molecule products consistent with intramolecular termination of diradical oligomers were isolated, and the structures of these molecules suggest how the diradical self-terminates in the absence of chain transfer.

Ruptured splenic abscess: a cause of pneumoperitoneum in a patient with AIDS.

We encountered a case of pneumoperitoneum caused by a gas-forming splenic abscess in a patient with acquired immunodeficiency syndrome (AIDS). Plain abdominal films and computed tomography demonstrated a large amount of free air. Pneumoperitoneum was eventually shown to represent gas liberated from fermentation by gas-forming organisms within the splenic abscess. Gas-containing necrotic tissue from the ruptured spleen mimicked the spillage of feces from colon perforation. The authors emphasize that a ruptured abscess should be included in the differential diagnosis of acute abdomen in an immunocompromised patient.

Structural analogues of the bimetallic reaction center in acetyl CoA synthase: a Ni--Ni model with bound CO.

Models for the active site of the acetyl CoA synthase (ACS) were synthesized by attachment of Cu+ and Ni(0) to nickel diaminodithiolate (S2N2) and diamidodithiolate (S2N2') complexes. The Ni-Ni species form stable CO adducts, i.e., [{(CO)2Ni}{NiS2N2'}]2-, whereas the Cu-NiS2N2 and Cu-NiS2N2' models do not. These results provide supporting evidence for a biological role for reduced nickel in ACS.

Characterization of chlorovinylcobalamin, a putative intermediate in reductive degradation of chlorinated ethylenes.

The first X-ray structure of a vinylcobalamin is reported. Chlorovinylcobalamin is formed in the reaction of cob(I)alamin with chloroacetylene. Subsequently, cob(I)alamin catalyzes the reduction of chlorovinylcobalamin to vinylcobalamin in the presence of excess titanium(III)citrate. Introduction of a chlorine onto the vinyl group of vinylcobalamin greatly changes its reduction potential. These results are discussed with respect to vitamin B12-catalyzed dechlorination of perchloroethylene, a pollutant on the priority list of the EPA.

Aggregation of PMe3-stabilized molybdenum sulfides and the catalytic dehydrogenation of H2S.

The reactivity of [MoS(4)](2-) (1) toward PMe(3) was explored in the presence and absence of proton donors. Whereas MeCN solutions of (Et(4)N)(2)[MoS(4)] and PMe(3) are stable, in the presence of H(2)S such solutions catalyze formation of H(2) and SPMe(3). Addition of NH(4+) to such solutions afforded MoS(2)(PMe(3))(4) (2), which can be prepared directly from (NH(4))(2)[1]. Compound 2 is reactive toward thiols via a process proposed to involve the initial dissociation of one PMe(3) ligand, a hypothesis supported by the relative inertness of trans-MoS(2)(dmpe)(2). Benzene solutions of 2 react with EtSH to give Mo(2)(mu-S)(mu-SH)(PMe(3))(4)(SEt)(3) (3Et). Analogous reactions with thiocresol (MeC(6)H(4)SH) and H(2)S gave Mo(2)(mu-S)(mu-SH)(PMe(3))(4)(SR)(3) (R = tol, H). Crystallographic analyses of 3Et, 3H, and 3tol indicate dinuclear species with seven terminal ligands and a Mo(2)(mu-SR)(mu-S) core (r(Mo)(-)(Mo) = 2.748(1) A). From reaction mixtures leading to 3Et from 2, we obtained the intermediate Mo(IV)(2)(mu-S)(2)(SEt)(4)(PMe(3))(2) (4), an edge-shared bis(trigonal pyramidal) structure. Compounds 3H and 3Et react further with H(2)S to give Mo(4)(mu(2)-S)(4)(mu(3)-S)(2)(PMe(3))(6)(SH)(2) (5H) and Mo(4)(mu(2)-S)(4)(mu(3)-S)(2)(PMe(3))(6)(SEt)(2) (5Et), respectively. Analogously, W(4)(mu(2)-S)(4)(mu(3)-S)(2)(PMe(3))(6)(SH)(2) was synthesized from a methanol solution of (NH(4))(2)WS(4) with H(2)S and PMe(3). A highly accurate crystallographic analysis of (NH(4))(2)MoS(4) (R(1) = 0.0193) indicates several weak NH.S interactions.
