Identification of mitophagy-related biomarkers with immune cell infiltration in psoriasis.

BACKGROUND: Psoriasis is an inflammatory disorder characterized by scaly erythematous plaques and significant comorbidities. Recent studies have suggested that impaired mitophagy, the cellular mechanism for removing dysfunctional mitochondria, may contribute to the pathogenesis of psoriasis. METHODS: In this study, we analyzed bulk RNA sequencing data from 167 healthy individuals and 177 patients with psoriasis obtained from the Gene Expression Omnibus database (GSE30999 and GSE54456). Mitophagy-related genes were isolated using weighted gene co-expression network analysis. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were performed and protein-protein interaction networks were constructed for the functional enrichment of genes associated with mitophagy. The correlations between genes associated with mitophagy, signaling pathways, and immune cell infiltration were analyzed. The potential diagnostic value of genes associated with mitophagy was evaluated using receiver operating characteristic (ROC) curves, which were validated in imiquimod-induced psoriatic skin lesions in mice. RESULTS: We identified 3,839 differentially expressed genes between healthy individuals and patients with psoriasis, and 23 genes were selected as hub genes showing a high correlation with mitophagy in psoriasis. GO and KEGG analyses revealed that hub and associated genes were significantly correlated with skin functions, such as epidermal development and keratinocyte differentiation. In addition, mitophagy-related genes were negatively associated with pro-inflammatory and pro-proliferation pathways in psoriasis. Among the immune cells, CD4+ T cells were most significantly affected by mitophagy-related genes. ROC analysis demonstrated that mitophagy-related genes, especially ACER1, C1ORF68, CST6, FLG2, GJB3, GJB5, GPRIN2, KRT2, and SPRR4 were potential biomarkers of psoriasis for use in diagnosis or treatment. CONCLUSIONS: Mitophagy-related genes play crucial roles in psoriasis and have potential use as biomarkers, providing insights into disease mechanisms and therapeutic targets. Further research may lead to the development of new strategies for psoriasis management.

From controllable attached isolation moieties to possibly highly efficient nonlinear optical main-chain polyurethanes containing indole-based chromophores.

For the first time, the indole-based NLO chromophores were embedded into the polymer main chain, and different isolation groups were attached to their donor side with the efforts of adjusting the NLO properties of the resultant main-chain polyurethanes, according to the site isolation principle. Thanks to the main-chain structure and the advantages of the indole-based chromophores, all the polymers show excellent transparency, good processability, thermal stability, and relatively good NLO effects. The tested NLO properties of the polymers demonstrate that there is a suitable isolation group present for the sulfonyl-based chromophore to boost its microscopic beta value to a possibly higher macroscopic NLO property efficiently.

Infrared emission spectra and equilibrium structures of gaseous HgH2 and HgD2.

A detailed analysis of the high-resolution infrared emission spectra of gaseous HgH2 and HgD2 in the 1200-2200 cm(-1) spectral range is presented. The nu3 antisymmetric stretching fundamental bands of 204HgH2, 202HgH2, 201HgH2, 200HgH2, 199HgH2, 198HgH2, 204HgD2, 202HgD2, 201HgD2, 200HgD2, 199HgD2, and 198HgD2, as well as a few hot bands involving nu1, nu2, and nu3 were analyzed rotationally, and spectroscopic constants were obtained. Using the rotational constants of the 000, 100, 01(1)0, and 001 vibrational levels, we determined the equilibrium rotational constants (B(e)) of the most abundant isotopologues, 202HgH2 and 202HgD2, to be 3.135325(24) cm(-1) and 1.569037(16) cm(-1), respectively, and the associated equilibrium Hg-H and Hg-D internuclear distances (re) are 1.63324(1) A and 1.63315(1) A, respectively. The re distances of 202HgH2 and 202HgD2 differ by about 0.005%, which can be attributed to the breakdown of the Born-Oppenheimer approximation.

The vibration-rotation emission spectrum of hot BeF2.

The high-resolution infrared emission spectrum of BeF2 vapor at 1000 degrees C was rotationally analyzed with the assistance of large-scale ab initio calculations using the coupled-cluster method including single and double excitations and perturbative inclusion of triple excitations, in conjunction with correlation-consistent basis sets up to quintuple-zeta quality. The nu3 fundamental band, the nu1+nu2, nu1+nu3, and 2nu2+nu3 combination bands, and 18 hot bands were assigned. The symmetric stretching (nu1), bending (nu2), and antisymmetric stretching (nu3) mode frequencies were determined to be 769.0943(2), 342.6145(3), and 1555.0480(1) cm-1, respectively, from the band origins of the nu3, nu1+nu3, and nu1+nu2 bands. The observed vibrational term values and B rotational constants were fitted simultaneously to an effective Hamiltonian model with Fermi resonance taken into account, and deperturbed equilibrium vibrational and rotational constants were obtained for BeF2. The equilibrium rotational constant (Be) was determined to be 0.235 354(41) cm-1, and the associated equilibrium bond distance (re) is 1.3730(1) A. The results of our ab initio calculations are in remarkably good agreement with those of our experiment, and the calculated value was 1.374 A for the equilibrium bond distance (re). As in the isoelectronic CO2 molecule, the Fermi resonance in BeF2 is very strong, and the interaction constant k122 was found to be 90.20(4) cm-1.

Gaseous HgH2, CdH2, and ZnH2.

Gaseous HgH2, CdH2, and ZnH2 molecules were synthesized by the direct gas-phase reaction of excited mercury, cadmium, and zinc atoms with molecular hydrogen. The molecules were identified by their high-resolution infrared emission spectra, and the metal-hydrogen bond lengths were determined from the rotational analysis of the antisymmetric stretching fundamental bands.

The vibration-rotation emission spectra of gaseous CdH2 and CdD2.

The vibration-rotation emission spectra of CdH2 and CdD2 molecules have been recorded at high resolution using a Fourier-transform spectrometer. The molecules were generated in a furnace-discharge emission source by reaction of cadmium vapor with molecular hydrogen or deuterium. The fundamental bands for the antisymmetric stretching mode (upsilon3) of CdH2 and CdD2 were detected at about 1771.5 and 1278.3 cm(-1), respectively. In addition, the 002(sigma(g)+)-001(sigma(u)+) and 01 l(pi(g))-010(pi(u)) hot bands were observed for CdH2. Spectroscopic constants were determined for each of the 12 observed isotopologs: 110CdH2, 111CdH2, 112CdH2, 113CdH2, 114CdH2, 116CdH2, 1l0CdD2, 111CdD2, 112CdD2, 113CdD2, 114CdD2, and 116CdD2. The average Cd-H and Cd-D bond distances (r0) were determined to be 1.683028(10) and 1.679161(16) angstroms, respectively.

The vibration-rotation emission spectrum of gaseous HZnCl.

Gaseous HZnCl has been synthesized for the first time in a high-temperature tube furnace with a dc discharge in a flowing mixture of pure HCl and Zn vapor. The vibration-rotation emission spectrum of HZnCl was recorded at high resolution using a Fourier transform spectrometer. The H-Zn stretching modes (nu(1)) of the H(64)Zn(35)Cl, H(66)Zn(35)Cl, H(68)Zn(35)Cl, and H(64)Zn(37)Cl species, as well as the 2nu(1)-nu(1) hot band of the most abundant isotopologue H(64)Zn(35)Cl, were observed near 1966 cm(-1). A least-squares fit was performed for each of the four observed isotopologues, and their spectroscopic constants were determined.
