[The mechanisms of carbon monoxide inhalation on the apoptosis of pulmonary cells in rats with acute lung injury induced by lipopolysaccharide].

OBJECTIVE: To investigate the effects of carbon monoxide (CO) inhalation on the apoptosis of pulmonary cells in rats with acute lung injury (ALI) induced by lipopolysaccharide (LPS) and to investigate its mechanisms. METHODS: Eighteen male SD rats were randomly divided into three groups. The ALI group received LPS 5 mg/kg intravenously, while the control group received normal saline, and the CO inhalation group received an inhalation of 2.5 x 10(-4) CO (V/V) after ALI induced by LPS 5 mg/kg. The animals were sacrificed by exsanguinations and lung tissue was harvested at 3 h of observation for determination of the heme oxygenase-1 (HO-1) mRNA with semiquantitative reverse transcription-polymerase chain reaction (RT-PCR), the level of tumor necrosis factor-alpha (TNF-alpha), interleukin-6 (IL-6) and IL-10 with enzyme-linked immunosorbent assay (ELISA) and the maleic dialdehyde (MDA) concentration, myeloperoxidase (MPO) and superoxide dismutase (SOD) activity with chemical methods. The extent of cell apoptosis was observed by using the flow cytometric method. RESULTS: The change of the levels of HO-1 mRNA, TNF-alpha, IL-6, IL-10, MDA, MPO, SOD and apoptotic cells of the ALI group was significant as compared with the control group [1.002 +/- 0.004, (0.47 +/- 0.06) pg/mg prot, (0.49 +/- 0.12) pg/mg prot, (0.42 +/- 0.08) pg/mg prot, (0.79 +/- 0.14) nmol/mg prot, (6.0 +/- 1.0) U/mg prot, (74 +/- 7) U/mg prot, (0.12 +/- 0.03)%, P < 0.05 or < 0.01], and lung injury was severe. The levels of TNF-alpha, IL-6, MDA, MPO, and apoptotic cells of the CO inhalation group [(0.91 +/- 0.25) pg/mg prot, (0.64 +/- 0.05) pg/mg prot, (1.02 +/- 0.23) nmol/mg prot, (7.2 +/- 1.6) U/mg prot, (1.60 +/- 0.34)%] were decreased, while HO-1, IL-10 and SOD expression [5.433 +/- 0.921, (0.26 +/- 0.07) pg/mg prot, (60 +/- 10) U/mg prot] were increased compared with the ALI group [(1.48 +/- 0.23) pg/mg prot, (1.16 +/- 0.26) pg/mg prot, (1.27 +/- 0.33) nmol/mg prot, (8.2 +/- 1.5) U/mg prot, (3.18 +/- 0.51)%, 3.081 +/- 0.823, (0.15 +/- 0.03) pg/mg prot, (51 +/- 7) U/mg prot, P < 0.05 or < 0.01], and lung injury was attenuated. CONCLUSION: CO inhalation protects lung from injury by regulating oxidative/anti-oxidative and inflammatory/anti-inflammatory disorders, inhibiting excessive cell apoptosis and up-regulating HO-1 expression, which may play an important role in the pathogenesis of LPS-induced ALI.

[Experimental study on the phenotype of human embryonic lung fibroblasts transfected with Ad-BMP-2].

OBJECTIVE: To investigate the effects on the phenotype, especially the mineralization ability of human embryonic lung fibroblasts(HELFs) by transfection with adenovirus vector ecoding human bone morphogenetic protein-2 gene (Ad-BMP-2). METHODS: The HELFs were primarily cultured, then transfected with Ad-BMP-2. The morphologic characteristics of the cells were observed. The cell proliferation, alkaline phosphatase activities, BMP-2 protein expression, and the mineralization ability were detected with the methods of MTT, ALP staining, Western blot, and alizarin red S staining, respectively. RESULTS: After transfection, the shape of HELFs changed from silm spindle to multifigure, the cells became bigger than before. The colonies changed from unilaminar into multilaminar. The proliferation of HELFs was severely inhibited after transfection. An obvious BMP-2 lane was shown in Western blotting. Most cells presented positive in ALP staining, and large number of nacarat mineralized nodes were observed after alizarin red S staining. CONCLUSION: HELFs were capable of transforming into osteoblast-like phenotype, and were endowed with the ability of mineralization while being transfected with Ad-BMP-2.

[Diagnosis and treatment of 23 cases with stylohyoid syndrome].

PURPOSE: To assess the diagnosis and treatment of stylohyoid syndrome. METHODS: The clinical data of 23 patients with stylohyoid syndrome were analyzed retrospectively,the symptoms,age,gender, length of styloid process, and clinical signs were investigated in their relationship to the disease onset , the value of styloidectomy via transoral and extraoral approach was compared. RESULTS: (1) Pain and foreign body sensation in oropharyngeal area accounted for 83%(19/23) of the symptoms ,followed by 43%(10/23) of headache, tongueache and periauricular discomfort. (2) Females were more often affected than males. The age peak was 50s and 30s ,accounting for 43%(10/23) and 30%(7/23) respectively. (3) 91%(21/23) of the excised styloid processes were longer than 3cm in length, 9% (2/23) of them shorter than 3cm but with abnormality in shape. (4) 57%(13/23) of the elongated styloid processes could be palpated transorally. (5) Through extraoral approach, all kinds of diseased styloid processes could be excised thoroughly and safely. CONCLUSIONS: Stylohyoid syndrome is one of the causes of head and neck pain. A complete medical history , careful palpation of the oral pharyngeal area, and radiographic examination are essential for the diagnosis. Styloidectomy by extraoral approach is effective for curing of the disease.

[Study on mRNA expression of fibronectin and integrin beta1 during fracture healing].

OBJECTIVE: To investigate the spatial and temporal expression changes of fibronectin and integrin beta1 mRNA during fracture healing. METHODS: Using in situ RT-PCR technique, the spatial and temporal expression pattern of both fibronectin and integrin beta1 mRNA was detected on paraffined slices in different rabbit mandibular fracture healing phases. RESULTS: (1) The mRNA of integrin beta1 and fibronectin was widely expressed in fractured bone cells. The positive stain existed in cytoplasm and nucleolus. (2) After 7 days of fracture, increaseded integrin beta1 mRNA expression was observed, and it reached maximal levels approximately 14- 30 days post-fracture and returned to normal levels after 60 - 90 days. (3) Fibronectin mRNA was detected on 3 days post-fracture, it reached maximal levels at 7 - 14 days, faint expression was detected at 30 days, undetectable 60 days afterwards. CONCLUSION: During fracture healing, the mRNA expression of integrin beta1 and fibronectin increased locally, FN and Itg beta1 cooperate with each other, it is suggested that they have an important influence on fracture healing.

[Study of the expression of integrin alphaV beta3 in mandibular distraction osteogenesis].

OBJECTIVE: To investigate the expression and distribution pattern of Integrin alphaV beta3 (ItgalphaV beta3) in course of distraction 44 adults New Zealand white osteogenesis and to quest for the function of ItgalphaV beta3 in distraction osteogenesis (DO). METHODS: rabbits were selected and divided randomly into 3 groups: DO, bone fracture and normal group. There were 20, 20 and 4 rabbits in each group separately. Animal models were established and animals were killed at different time points. Sections were stained by IHC method. Distribution and expression pattern of ItgalphaV beta3 were observed by semi-quantitative technique, and the results were managed with statistic methods. RESULTS: The expression of ItgalphaV beta3 was found both in the DO and bone fracture groups. The positive staining was seen mainly in vascular endothelial cells on cytomembrane and in cytoplasm. The staining intensity of group of DO was higher than that of the bone fracture group. CONCLUSION: ItgalphaV beta3 plays an important role in promoting the process of DO.

[Investigation of primary cell culture for dental pulp: comparing of coronal pulp and root pulp].

PURPOSE: To compare and establish an ideal method for the culture of human primary dental coronal and root pulp cells. METHODS: The whole dental pulp tissues were obtained from healthy and young human teeth extracted for orthodontic purposes. The cells in coronal and root pulp were separately cultured by the tissue explant and tissue explant-collagenase digestion(0.1% type I collagenase) methods. By evaluating the attachment efficiency,viability and the successfulness of the cultured cells, the optimal primary cell culture process of human dental coronal and root pulp was determined. RESULTS: The tissue explant culture had more successfulness,more viability and less attachment efficiency than that of tissue explant-collagenase digestion method in the culture of human coronal and root dental pulp cells. CONCLUSIONS: Tissue explant method was superior to tissue explant-collagenase digestion method for the culturing of human dental coronal and root pulp cells. The ideal method for dental coronal and root pulp cells was established in this study.

[The characteristics of cultured dental pulp cells and the localization of dental pulp stem cells].

PURPOSE: This study was aimed to investigate the localization of dental pulp stem cells (DPSCs) by comparing the characteristics of cultured dental pulp cells in coronal and root pulp. METHODS: Human dental coronal and root pulp cells were cultured in tissue-explant method, the cell culture successfulness, attachment efficiency, cell viality, morphology, proliferation pattern, and the mineralization ability were observed, the localization of DPSCs was investigated in the functional respect of DPSCs. RESULTS: The human dental root pulp cells have more culture successfulness,more attachment efficiency,more cell viality, more primary characteristics, and stronger induced mineralization ability than that of coronal pulp cells. Root and coronal pulp cells showed same proliferation patterns. CONCLUSIONS: DPSCs may exist in both dental root and coronal pulp, and the density of DPSCs in the root pulp may be higher than the coronal pulp.

[The establishment of an animal model of bilateral mandibular distraction osteogenesis in rabbits].

PURPOSE: To establish an animal model of bilateral mandibular distraction osteogenesis in rabbits. METHODS: 16 adult male white New Zealand rabbits were divided into 4 groups randomly, 4 in each group. Osteotomy was performed on mandible bilaterally before the first premolar. Bone stumps were repositioned and fixed with distractors. After 7 days of latency period, distraction was initiated at a rate of 1mm/d for 7 days. Then distractors were held in place for days. Animals were killed at different time points, and specimens were taken for X-ray and histology examination. RESULTS: The mandibles were lengthened successfully. The distraction gaps were filled with regenerated bone tissue. CONCLUSION: The method in this study is practicable and repeatable which could be used to establish an animal model of mandibular distraction osteogenesis in rabbits.

[The ALP activity of cultured dental coronal and root pulp cells in vitro].

PURPOSE: To compare the ALP activity in human dental coronal and root pulp,and investigate the localization of dental pulp stem cells in the dental pulp. METHODS: The human dental root and coronal pulp cells were cultured in tissue-explant method, the 5th generation of cells in good growth condition were subcultured with conditioned media (20% DMEM + 10 nmol/L + dexamethaone + 50mg/L ascorbic acid) to induce mineralization. At the days of 5, 10, 15 after inducing, Gomori alkaline phosphatase stain was used to investigate alkaline phosphatase (ALP) activity in root and coronal pulp cells. RESULTS: Positive staining was found in both root and coronal cells, the ALP activity was equal at 5 days of incubation, but higher in root pulp cells than that of coronal pulp cells after 10 and 15 days of incubation. CONCLUSIONS: ALP activity in dental root pulp is higher than that in coronal pulp. DPSCs may exist in both dental root and coronal pulp, and the density of DPSCs is higher in the root pulp than the coronal pulp.

[Effect of combination of nitric oxide inhalation and inverse ratio ventilation in endotoxin-induced acute respiratory distress syndrome in sheep].

OBJECTIVE: To observe the effect of combination of nitric oxide (NO) inhalation and inverse ratio ventilation (IRV) on oxygenation and hemodynamics in endotoxin-induced acute respiratory distress syndrome (ARDS) in sheep. METHODS: Sheep ARDS model was induced by an intravenous infusion of low dose endotoxin, and then animals were randomly divided into two groups. (1) NO group (n=6), inhalation of 40x10(-6) nitric oxide. (2) Combination group (n=6), receiving mechanical ventilation with IRV (inspiratory-to-expiratory ratio of 2:1) and inhalation of 40x10(-6) NO. The dynamic changes in gas exchange and hemodynamics were measured with the aid of Swan-Ganz catheter and arterial blood gas analysis before and after the onset of, ARDS and 30 minutes after treatment. RESULTS: The combination of IRV and 40x10(-6) NO inhalation rapidly reduced mean pulmonary arterial pressure (MPAP), increased PaO(2), decreased P((A-a))O(2), and Qs/Qt without inducing significant change in systemic hemodynamics, and it was a more effective method of correcting hypoxemia than inhalation of nitric oxide alone. NO inhalation did not change the airway pressure of the model, but the combined treatment resulted in reduction of peak inspiratory pressure and increase of mean airway pressure. CONCLUSION: The combined use of IRV and NO inhalation has an additive effect on improving oxygenation in endotoxin-induced acute respiratory distress syndrome in sheep.
