Preparation and biological evaluation of 153Sm-DOTMP as a potential agent for bone pain palliation.

BACKGROUND: Designing ideal radiopharmaceuticals for use as bone pain palliative agents requires the use of a moderate energy beta emitter as the radionuclide and a polyaminophosphonic acid as the carrier molecule. Cyclic polyaminophosphonic acid ligands are known for endowing higher thermodynamic stability and kinetic inertness of the radiolabelled agent when complexed with radiolanthanides. AIM: To use Sm (T1/2=46.27 h, Ebeta,max=0.81 MeV, Egamma=103 keV) as the radioisotope, obtainable at an adequate specific activity and high radionuclidic purity by irradiation of a natural Sm2O3 target, and 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetramethylene phosphonic acid (DOTMP) as the carrier ligand. RESULTS: The radiolabelling yields under optimized conditions were near quantitative with the additional merit of using a relatively low ligand:metal ratio of 2:1 unlike the 250-fold excess of ligands used in the case of the established agent, Sm-EDTMP. Radiochemical purity was retained with insignificant dissociation on storage up to 10 d at room temperature. Biodistribution studies in Wistar rats demonstrated selective skeletal uptake (4.52%+/-0.49% of injected activity per gram in tibia at 30 min post-injection) with rapid blood clearance and minimal uptake in any of the major organs. No leaching of skeletal activity was observed up to 48 h post-injection. Scintigraphic studies carried out in rabbits also showed significant skeletal accumulation and almost no retention of activity in other vital organs/tissues.

Serum prolactin in seizure disorders.

This study aimed to determine the post-ictal prolactin (PL) response in different types of seizures and seizure-like events in children, and correlate with the post-ictal duration. Patients were divided into group I (generalized tonic-clonic seizures, complex partial seizures or simple partial seizures), group II (febrile convulsions) and group III (conditions mimicking seizures). Group IV consisted of 25 controls. Blood was collected within 2 hours of the seizure and PL levels assayed. PL levels were significantly high only within group I; highest and baseline levels were attained within 10 minutes and by 100 minutes respectively. The sensitivity and specificity of elevated PL for epileptic seizures were 64 percent and 98 percent respectively. It is concluded that a high prolactin level within 100 minutes of a seizure is suggestive that a generalized or complex partial seizure has occurred.

Development of a radioimmunoassay procedure for aflatoxin B1 measurement.

A radioimmunoassaay (RIA) procedure to measure aflatoxin B(1) (AfB(1)) in agricultural commodities was developed. AfB(1) oxime derivative was synthesized, characterized, and used for preparation of (125)I-labeled AfB(1). Antiaflatoxin B(1) serum was raised in-house using AfB(1)-bovine serum albumin conjugate as immunogen. The assay system was optimized in the range of 0.2-5 ng/mL, using a liquid phase (PEG) as well as a solid phase (coated polystyrene beads) separation system. Inter-assay and intra-assay variations, recovery, and parallelism studies validated the assay. AfB(1) analysis was carried out in nearly 130 samples of different agricultural commodities. The correlation coefficient was determined using commercial ELISA and in-house-developed RIA methods.

99mTc-labeling studies of a modified metronidazole and its biodistribution in tumor bearing animal models.

A cysteine-based bifunctional chelating agent viz. N-(2'-hydroxybenzyl)-cysteine with a free carboxylic acid group (CAA) was synthesized. Bearing in mind the affinity of localization of nitroimidazoles in hypoxic tissues, this bifunctional chelating agent was coupled to metronidazole (MNZ). The (99m)Tc labeling studies of the novel agent (MNZCAA) thus obtained, was carried out and the radiolabeled product was subsequently purified. The complexation yield under optimized condition was approximately 85%. Biodistribution studies carried out in Swiss mice bearing 'barcl-95' tumors showed selective accumulation of the injected activity in the tumor (1.70%/g at 30 min p.i.) with renal as well as hepatobiliary clearance. High tumor/muscle ratio (14.7 at 3 h post-injection) of the novel agent indicates considerable promise towards further evaluation.

177Lu-labeled cyclic polyaminophosphonates as potential agents for bone pain palliation.

177Lu (T 1/2 = 6.71 d, Ebeta(max) = 497 keV) has radionuclidic properties suitable for use in palliative therapy of bone pain due to metastasis. 177Lu was produced in high-specific activity (3-4TBq/g) and excellent radionuclidic purity (100%) by thermal neutron bombardment of natural Lu target. Two cyclic tetraaminomethylene phosphonate ligands, namely DOTMP and CTMP were synthesized and radiolabeled with 177Lu. The 177Lu-DOTMP complex was formed with very high yield (> 99%) and showed excellent stability (up to 40 d at room temperature). Biodistribution of 177Lu-DOTMP was carried out in Wistar rats and the complex showed significant bone uptake (4.23%/g in femur and 5.23% in tibia at 3 h p. i.), rapid clearance from blood (no activity at 3 h p. i.) and minimum uptake in soft tissues.
