Pd(CH3CN)2Cl2-catalyzed oxidative heterodimerization reaction of 2,3-allenamides and 1,2-allenyl ketones: an efficient synthesis of 4-(furan-3'-yl)-2(5H)-furanimines.

The Pd(II)-catalyzed oxidative heterodimerization reaction of 2,3-allenamides and 1,2-allenyl ketones was studied. It provides an efficient route for the synthesis of the polysubstituted 4-(furan-3'-yl)-2(5H)-furanimines, which are not readily available from the known methods. Due to the application of benzoquinone, the loadings of both the palladium catalyst and ketone have been greatly reduced for the oxidative heterodimerization of 2,3-allenamides and 1,2-allenyl ketones in acetic acid.

Efficient two-step synthesis of 3-halo-3-enals or 2-halo-2-alkenyl ketones from propargylic bromides via a unique cationic 1,2-aryl or proton shift in electrophilic addition reaction of 2,3-allenols with X+.

The reaction of readily available 1-substituted 2,3-allenols with Br2, NBS, or I2 afforded the not-easily-available but synthetically useful 3-halo-3-alkenals or 2-halo-2-alkenyl ketones in good yields via a sequential electrophilic interaction of X+ with the allene moiety , a 1,2-aryl or proton shift, and a H+-elimination process; the structures of the products were established by X-ray diffraction study.

PdCl2-catalyzed two-component cross-coupling cyclization of 2,3-allenoic acids with 2,3-allenols. An efficient synthesis of 4-(1',3'-dien-2'-yl)-2(5H)-furanone derivatives.

Cross-coupling cyclization reaction between 2,3-allenoic acids 1 and 2,3-allenols 2, in which two allenes functioned differently, was realized to afford 4-(1',3'-dien-2'-yl)-2(5H)-furanone derivatives 3. The reaction may proceed via an oxypalladation, insertion, and beta-hydroxide elimination process. A high E-stereoselectivity of the new formed C=C double bond was observed.

Highly regio- and stereoselective synthesis of 2(E),4-Alkadienoates via the Pd(0)-catalyzed reaction of aryl halides with 3,4-alkadienoates.

[reaction: see text] 2(E),4-Alkadienoates were prepared highly stereoselectively via the Pd(0)/Ag(2)CO(3)-cocatalyzed reaction of 3,4-alkadienoates and aryl halides. The reaction is believed to proceed via the oxidative addition-carbopalladation-beta-H elimination process. Compared to the other reported methods for the synthesis of 2,4-alkadienoates, in which usually only disubstituted C=C bonds were formed, the current reaction forms the trisubstituted or even tetrasubstituted C=C bond highly stereoselectively.

Pd(II)-catalyzed oxidative dimeric cyclization-coupling reaction of 2,3-allenoic acids: an efficient synthesis of bibutenolide derivatives.

Three sets of convenient catalytic systems have been developed for the oxidative dimeric cyclization coupling of differently substituted 2,3-allenoic acids catalyzed by Pd(II), affording bibutenolides that are not otherwise readily available. The advantages and disadvantages of these systems are discussed. Although the diastereoselectivity for the bicyclization of racemic 2,3-allenoic acids is low, excellent diastereoselectivity was realized in the bicyclization reaction of optically active 2,3-allenoic acids, leading to the optically active bibutenolides in high yields and ee. Based on a mechanistic study, it is believed that the reaction may proceed by means of a double oxypalladation and reductive elimination to yield butenolide 3 and Pd(0) species, which may be reoxidized to the catalytically active Pd(II) species in the presence of alkyl iodide/air, metallic iodide/air, or benzoquinone.

Studies on Pd(II)-catalyzed synthesis of (Z)-alpha-haloalkylidene-beta-lactones from cyclocarbonylation of 2-alkynols and the subsequent coupling reactions.

[reaction: see text] A good regio- and stereoselectivity was observed for the PdCl2-catalyzed cyclocarbonylation of 2-alkynols with CuCl2 affording (Z)-alpha-chloroalkylidene-beta-lactones. The highly optically active (Z)-alpha-chloroalkylidene-beta-lactones could be easily prepared from the readily available optically active propargylic alcohols. The Pd(II)-catalyzed cyclocarbonylation of 2-alkynols with CuBr2 was also studied. Although the yields of (Z)-alpha-bromoalkylidene-beta-lactones were low, due to the relatively higher activity of the C-Br bond, the coupling reactions of (Z)-alpha-bromoalkylidene-beta-lactones were quite smooth to afford the corresponding products in high yields. A rationale for this reaction is discussed.

PdCl2-catalyzed efficient transformation of propargylic amines to (E)-alpha-chloroalkylidene-beta-lactams.

[reaction: see text] The PdCl2-catalyzed cyclocarbonylation reaction of propargylic amines with CuCl2 and benzoquinone afforded (E)-alpha-chloroalkylidene-beta-lactams in moderate to good yields. The formation of the corresponding Z-isomers or five-membered products was not observed. The reaction of the readily available optically active propargylic amines provides a convenient synthesis of the corresponding (E)-alpha-chloroalkylidene-beta-lactams with high ee values. The structure and the stereochemistry of the products were established by the X-ray single-crystal diffraction study of (E)-6d and (E)-6e, which indicates that the stereoselectivity in this reaction is different from what was observed with propargylic alcohols. A rationale for this reaction was proposed.

Enantioseparation of chiral allenic acids by micellar electrokinetic chromatography with cyclodextrins as chiral selector.

Enantioseparation of chiral aryl allenic acids by micellar electrokinetic chromatography (MEKC) with cyclodextrins (CDs) as chiral selectors was described. The screen of chiral selectors (beta-CD, gamma-CD, and hydroxypropyl (HP)-gamma-CD) showed that the enantioseparation was not only dependent on the type of CD but also the presence of 2-propanol in the buffer. In order to optimize the operational parameters, the effect of the concentration of CDs, sodium dodecyl sulfate (SDS), and 2-propanol, as well as the buffer ionic strength and pH on enantioseparation were studied. It was proved that the concentration of CDs, 2-propanol, and the buffer ionic strength were the critical parameters. Under optimal conditions, baseline separations of all seven allenic acid enantiomers were achieved. Furthermore, the method validation in terms of repeatability, linearity, limit of detection (LOD), and limit of quantitation (LOQ) were performed. Using the present method, the optical purity of a nonracemic sample with the enantiomeric excess (e.e.%) value of 99.65% was determined.

Highly selective synthesis of bicyclic quinolizidine alkaloids and their analogues via double RCM reaction of N-alkynyl-N-(1,omega)-alkadienyl acrylamides.

The double ring-closing metathesis reaction of N-alkynyl-N-(1,omega)-alkadienyl acrylamides 1 using first- or second-generation Grubbs' catalyst afforded, in a highly selectively manner, the fused bicyclic quinolizidine alkaloid derivatives and their analogues bearing a 1,3-diene moiety, which may further undergo a Diels-Alder reaction with a dienophile to afford N-containing polycyclic compounds. The excellent selectivity of fused/dumbbell-mode cyclization has been realized by the higher reactivity of the electron-rich C=C bond or carbon-carbon triple bond combined with the lower reactivity of the electron-deficient C=C bond toward metallocarbenes and the thermodynamically more stable nature of fused bicyclic compounds 3 vs dumbbell-type bicyclic compounds 4.

Highly regio- and stereoselective synthesis of polysubstituted cyclopropane compounds via the Pd(0)-catalyzed coupling-cyclization reaction of 2-(2',3'-allenyl)malonates with organic halides.

A new method for highly regio- and stereoselective synthesis of polysubstituted cyclopropane compounds via the Pd(0)-catalyzed coupling-cyclization reaction of 2-(2',3'-allenyl)malonates with organic halides is described. In these reactions, the starting materials are easily available and the operation is convenient. The ratios of trans-isomer/cis-vinylic cyclopropanes are up to 98:2.

Studies on the regio- and stereoselectivity of halohydroxylation of 1,2-allenyl sulfides or selenides.

It was observed that the halohydroxylation of 1,2-allenyl sulfides or selenides with Br2 (CuBr2 or NBS) or I2 and water demonstrated a fairly good regioselectivity (i.e., the C=C bond that is remote from the S or Se atom was halohydroxylated with the halogen atom connecting to the middle carbon atom and the hydroxyl group connecting to the non-S terminal carbon or Se-substituted terminal carbon atom of the allene moiety), leading to the synthesis of synthetically important 3-organosulfur or seleno-2-haloallylic alcohols. The stereoselectivity depends on the nature of X+ and S or Se, showing a Z-selectivity with the matched Lewis acid-base pair.
