[Effects of expression silencing of MAGE3 by RNA interference on location and metastasis of lung carcinoma cells].

OBJECTIVE: To construct small interfering RNA (siRNA) expression vectors targeting human MAGE3 gene and to observe the effects of gene silencing of MAGE3 by RNA interference on location and metastasis of lung carcinoma cells. METHODS: MAGE3 mRNA targeted hairpin siRNA was devised and the oligonucleotide strands of DNA fragments encoding the above siRNA were synthesized. After annealing of the complementary strands, the DNA fragments were cloned into pSUPERneoGFP, followed by amplification and DNA sequencing, then transfected into human lung carcinoma NCI-H446. The expression of MAGE3 gene mRNA and protein were examined by RT-PCR and Western blotting. Colony formation assay and Boyden chamber assay were performed to detect the effects of MAGE3 on colony formation and metastasis. RESULTS: The DNA fragments encoding MAGE3-targeted siRNA were cloned into the pSUPERneoGFP and confirmed by restrictive enzyme digestion and DNA sequencing. RT-PCR and Western blotting revealed a strongly decreased expression level of MAGE3. The lung carcinoma cells transfected by siRNA group was significantly lower than others, an effect on its colony formation and invasiveness. The colony formation of lung carcinoma cells transfected by siRNA in soft agar and the number of cells penetrating matrigel both reduced, there is significant difference compared with untransfected group and transfected empty vector. CONCLUSION: An siRNA vector targeting human MAGE3 gene has been successfully constructed. Expression silencing of MAGE3 by RNA interference could reduce location and metastasis of lung carcinoma cells effectively.

[Correlation between premature ejaculation and redundant prepuce].

OBJECTIVE: To investigate the correlation between redundant prepuce and premature ejaculation. METHODS: Fifty-two cases suffering from premature ejaculation and redundant prepuce were treated with circumcision. They were asked to fill the investigating questionnaire about the changes of ejaculatory latent period, patients' and their wives' satisfaction with sexual life before and after the treatment. RESULTS: During 12 months after circumcision, 28 cases were cured and 11 cases were efficacious. The curative rate was 54.9% and effective rate was 76.5%. Twelve cases with no responding continued to be treated with routine methods, such as psychotherapy, daub narcotic to glans of penis and taking medicine to treat chronic prostatitis, which were used before circumcision but still no effects. During 18 months after circumcision, 4 cases were cured and 5 cases were efficacious. CONCLUSION: Redundant prepuce had direct or indirect relationship with premature ejaculation. The circumcision is one of the effective methods to treat premature ejaculation.

[Analysis on the anatomic features of 47 cases of hepatic hydatidosis complicated with biliary fistula].

OBJECTIVE: To propose a criterion and its significance of clinical classification of hepatic hydatidosis complicated with biliary fistula. METHODS: 47 hepatic hydatidosis with biliary fistula cases who were given a subadventitial pericystectomy were observed from 2000 to 2005 in a retrospective study. The methods included observation of the different anatomic features of hepatic hydatidosis complicated with biliary fistula during the surgical operation and evaluation of the curative effect. RESULTS: All the 47 patients recuperated successfully and had no complication. Based on the anatomic features of hepatic hydatidosis complicated with biliary fistula, a criterion on clinical classification was proposed as three types: tangential, transfixional and terminal types. CONCLUSIONS: Hepatic hydatidosis complicated with biliary fistula can be classified as three types according to its anatomic features.

[Expression of osteopontin in the pericystic layer of hepatic hydatid cyst].

OBJECTIVE: To investigate the expression and distribution of osteopontin (OPN) in the pericystic layer of Echinococcus granulosus cyst. METHODS: 60 surgically excised cysts were studied with HE and immunohistochemistry staining by using antibodies specific for OPN. Immunofluorescence double labeling technique and Media Cybernetics Image (Pro Plus 5.1 software) were used to appraise the relationship between the OPN and macrophages (CD68). RESULTS: Expression of OPN was found at various degree in hydatid cysts, 75% (45/60) was distributed in the intra-layer at the parasite side, 8.3% (5/60) distributed in the extra-layer near the side of liver parenchyma ("exocyst"-layer vs "adventitia"-layer, P<0.01). Macrophages were identified between the intra-layer and extra-layer, and OPN was observed in most of the macrophages. Meanwhile, OPN was concurrently found with the deposit of calcium in a similar distribution in the hydatid cyst. CONCLUSION: Osteopontin mainly distributes in the intra-pericystic layer of hepatic hydatid cyst.

[Prevalence study of age-related macular degeneration in Caojiadu blocks, Shanghai].

OBJECTIVE: To investigate the prevalence of age-related maculopathy (AMD) in Shanghai residents older than 50 years. METHODS: 1023 individuals were examined in the years 2002 and 2003 for clinical and subjective signs of dry and exudative AMD. A interview concerning clinical symptoms of AMD as also a complete eye exam was performed. Non-mydriatic retinoscopy was done by use of a direct opthalmoscopy. Fundus photography was used to confirm and document the diagnosis. The diagnosis of both forms of AMD was made according to the criteria of the Sub-society of Fundus Disease, Chinese Ophthalmological Society. RESULTS: 15.5% of the included population had AMD and 19 of them were exudative AMD. The prevalences of AMD in the age groups of 50-59, 60-69, 70-79 and more than 80 were identified as 5.7%, 13.5%, 20.2% and 23.5%, respectively (chi(2) = 27.97, P < 0.01). No statistical differences were found concerning atrophic and exudative AMD related to gender, education, daily focus time, smoking, alcohol consumption and systematic diseases. In the AMD patients, 5.1% and 36.2% of the eyes were identified as blind or with low vision. In exudative AMD patients, 23.3% and 63.3% of the eyes were identified as blind or with low vision. The visual acuity of eyes with exudative AMD was significantly lower than the visual acuity of atrophic AMD eyes. (chi(2) = 15.4, P < 0.01). CONCLUSIONS: The prevalence of AMD is high, and increasing with aging. The AMD, especially the exudative AMD, leads to severe visual impairment.

[Formation mechanisms of the fibrous capsule around hepatic and splenic hydatid cyst].

OBJECTIVE: To investigate the formative mechanisms of the fibrous capsule around hepatic and splenic hydatid cyst. METHODS: HE stain was used to observe the histopathologic changes of the fibrous capsules around hepatic and splenic hydatid cysts and the adjacent parenchyma. The expression of collagen IV, fibronectin (FN), laminin (LN), transforming growth factor-beta1 (TGF-beta1) and tumor necrosis factor-alpha (TNF-alpha) were detected by using immunohistochemistry or in situ hybridization in fibrous capsules around 40 hepatic hydatid cysts and 15 splenic hydatid cysts including adjacent parenchyma. RESULTS: A special delaminated phenomenon was observed in the fibrous capsules around hepatic hydatid cysts. Granuloma-like pathologic changes were found in the fibrous capsules near worm and there were many depressed Glisson capsules and hepatic vein system in the fibrous capsules near hepatic parenchyma while these phenomena could not be observed in splenic hydatid cysts. All parameters were highly expressed in the fibrous capsules near hepatic parenchyma and the difference between the two fibrous capsules was significant (P<0.01). CONCLUSION: The mechanisms of forming fibrous capsule around hepatic and splenic hydatid cysts are different. The fibrous capsule around hepatic hydatid cyst is a granuloma-like structure, covered by the pressed Glisson system and hepatic vein system with a small gap between them; while the fibrous capsule around splenic hydatid cyst is formed by granuloma-like tissue covering the worm and there is no gap between the capsule and the splenicparenchyma.

[A clinicopathological study on 3 cases of severe acute respiratory syndrome].

OBJECTIVE: To study the pathological characteristics of severe acute respiratory syndrome (SARS) and its relationship to clinical manifestation. METHODS: Tissue specimens from 3 autopsy cases of diagnosed SARS were studied under microscopy and the clinical data were reviewed. RESULTS: The typical pathological changes of lungs were diffuse hemorrhage on surface. A mixture features of serous, fibrinous and hemorrhagic inflammation were seen in most pulmonary alveoli with engorgement of capillary and there were microthrombosis in some capillary. Pulmonary alveoli became thick with interstitial mononuclear inflammatory infiltration, diffused alveoli damage, desquamation of pneumocytes and hyaline-membrane formation. Fibrinoid materials and erythrocytes could be found in alveolar spaces. There were thrombo-embolisms in some bronchial artery. Meanwhile, haemorrhagic necrosis was showed in lymph nodes and spleen with attenuation of lymphocytes. Other atypical pathological changes, such as hydropic degeneration, fatty degeneration, interstitial cell proliferation and some lesions observed in liver, heart, kidney, pancreas may have existed before the hospitalization. CONCLUSION: Severe damages of pulmonary and immunological system damage are responsible for clinical features of SARS and may lead to death of patients.

[Detection of hepatic progenitor cells in patients with severe hepatitis and their distribution].

OBJECTIVES: To identify hepatic progenitor cells (HPCs) in patients with severe hepatitis (SH) by detecting their markers and investigate the features of their distribution and location. METHODS: Liver tissues taken from 59 SH patients were tested for the receptor of stem cell factor (c-kit), pi-class glutathione S-transferase (GST-pi), cluster of differentiation 34 (CD34), cytokeratin 19 (CK19), cytokeratin 18 (CK18) and alpha fetoprotein (AFP) by immunohistochemistry (IHC). Meanwhile, 58 patients with acute or chronic hepatitis were also detected to act as controls. RESULTS: Hepatic progenitor cells could be seen in SH patients. Most of them existed as ductular cells that had been called "typical ductular proliferation (ADP)" or "typical ductular reaction" in previous research. These ductular cells were mainly located at the portal areas, fibro septa, periportal parenchyma and the border of the pseudolobuli and inflammatory foci. Further, c-kit, GST-pi, CK19 and CK18, but not CD34 and AFP could be detected in these cells. Another kind of HPC was the small hepatocyte-like cell (SHLC), which could express c-kit, GST-pi, and CK18, but not CK19, CD34 and AFP. The semi-quantitative analysis showed that the scope of ADP in SH patients was significantly larger than that in acute and chronic hepatitis patients (chi2= 63.62, P<0.05), and the scope of ADP in subacute severe hepatitis and chronic severe hepatitis patients was also significantly larger than that in acute severe hepatitis patients. CONCLUSION: In the course of regeneration of viral hepatitis, different types of pathology have different features. In acute and chronic hepatitis (G1-2), the regeneration is mainly owing to the proliferation of mature hepatocytes, and in chronic hepatitis (G3-4), there is the participation of HPCs, although they are limited. In severe hepatitis, however, since the replicative capacity of normal hepatocytes is impaired or prohibited, liver regenerates and restores mainly by the means of hepatic stem cells activation and proliferation. But the hepatic stem cells don't differentiate into their mature functional compartments directly at all. There are several intermediary or transition populations. In human severe hepatitis, they are mainly ductular cells, and parts of them are small hepatocyte-like cells.
