[Study on the stigma/pollen vigor and self-compatibility of Platycodon grandiflorum].

OBJECTIVE: To provide the basal data for the breeding and cultivation of Platycodon grandiflorum. METHOD: The field investigation and pollination by bagging were carried out. TTC(2,3,5-triphenyl tetrazolium chloride) solution was used to test the pollen vigor. RESULT: The stigma life-span of P. grandiflorum was about 9 days, however the optimal time for pollination is 4-6 days after the petals opening, with the stigma was splitting lightly or significantly. When the petals opened, the anther began scattering pollen, and finished in the same day. The pollen vigor was about 81.4% at the beginning, and decreases to 27.6% three days later. The pollen vigor still remains 64.4% three days later, when the flower was kept in the desiccant. The natural fructification rate of self-flower-pollination was 4.8%. The fructification rate and compatible index was about 62.7% and 54.6, respectively, when the self-plant-pollination performed by hand. They decreased to 12.8% and 6.5 when the pollination was implemented during the flowering period. CONCLUSION: Low fructification percentage of self-flower-pollination attributes to the difference of maturing period of pistil and stamen, as well as the short pollen life-span of P. grandiflorum. The compatibility of self-plant-pollination is high during flowering period when pollination performed by hand. The life-span of the pollen can be prolonged significantly when keeping in the dry environment.

[Review of bioactivities of natural cycloartane triterpenoids].

Cycloartane triterpenoids, which exist widely in nature, are mainly distributed in Astragalus (Leguminosae) species, Trib. Cimicifuga (Ranunculaceae) and Thalictrium (Ranunculaceae) species and possess various bioactivities. Along with the development of isolation techniques of phytochemistry, more and more this kind of compounds are isolated and identified. However, bioactivity researches on the compounds are relatively lagged behind. Most researches are still in screening level, deficient in mechanism elucidation, short of action proven in vivo and SAR analysis. The author summarized the bioactivity of this kind of compounds from all aspects: anti-tumor, anti-virus, antibacterial, anti-inflammation, immune-regulatory, cardiovascular system, hepatic protection and so forth. This will be benefit for the further research and development of the compounds.

[Esophagectomy combined with radical lymphadenectomy by video-thoracoscopy].

OBJECTIVE: To explore the value of video-thoracoscopy in the thoracoscopic mobilization of the thoracic esophagus combined with radical lymphadenectomy. METHODS: Between March 2002 and May 2003, thoracoscopic mobilization of the thoracic esophagus combined with radical lymphadenectomy was attempted in 25 patients (test group) and 22 cases received routine open thoracotomy (control group). Mean age was 55 years (range 34 - 73). The cancers were T(1)-T(3). Dissection of the thoracic esophagus was attempted via a right-sided approach, followed by a laparotomy and a cervical incision. RESULTS: The thoracoscopic procedure was successful in all patients. There was no post-operative death in two groups. Mean node harvest was (7.8 +/- 1.7) nodes for test group and (7.5 +/- 1.3) nodes for control group (P > 0.05). Mean blood lo of the thoracic component was (130 +/- 83) ml for test group and (350 +/- 135) ml for control group (t = 6.83, P < 0.05). Median post-operative stay was (10.9 +/- 2.5) days for test group and (14.6 +/- 1.7) days for control group (t = 5.87, P < 0.05). CONCLUSION: Video-thoracoscopy could potentially provide an oncologically sound means for resecting the thoracic esophagus without the need for a thoracotomy. Radical thoracoscopic mobilization of the esophagus is feasible.

[Induction of angiogenesis in ischemic myocardium by adenovirus mediated angiopoietin-1 gene transfer, an experimental study].

OBJECTIVE: To investigate the effect on angiogenesis in ischemic myocardium of adenovirus mediated angiopoietin-1 gene transfer. METHODS: Ang-1 cDNA was obtained from human spleen by RT-PCR and then cotransfected into 293 host cells together with Adv5-CAG, E1 and E3 substituted adenovirus genome, thus constructing recombinant adenovirus Adv5-CAG/Ang-1. Recombinant adenovirus Adv5-CAG/LacZ containing LacZ gene was constructed too. Thirty six male New Zealand white rabbits were randomly divided into 3 group of 12 rabbits: DMEM group, Ang-1 group, and LacZ group and underwent high-positioned double-ligation of the left anterior descending coronary artery and then myocardial injection of DMEM, Adv5-CAG/Ang-1, or Adv5-CAG/LacZ respectively. Fourteen days after the operation 2 rabbits in each group were killed, the myocardial tissues where injected was given were taken to detect the expression of Ang-1 by RT-PCR. Coronary angiography was conducted 28 days postoperatively upon 5 rabbits in each group to observe the angiogenesis in the ischemic myocardium. Five rabbits in each group were killed at the 14 th and 28 th postoperative days to observe the capillary density by immunohistochemical staining. RESULTS: The Ang-1 cDNA cloned in the laboratory was 1,515 bp in length containing the signal peptide structure in accordance with the report in literature. Fourteen days after operation, Ang-1 gene was positive in the myocardium of Ang-1 group and negative in the other 2 groups. New vessel formation was obvious at the 28 th postoperative day in the Ang-1 group and not in the other 2 groups. Capillary density increased after operation in all 3 groups, however, more significant in the Ang-1 group, especially 28 days after. CONCLUSION: Adenovirus-mediated angiopoietin-1 gene effectively promotes the neovascularization in ischemic myocardium of rabbits.

Genotyping and species identification of Fritillaria by DNA chips.

AIM: To investigate the genetic polymorphism of several species of Fritillaria and to develop a DNA chip for the genotyping and identification of the origin of various species of Fritillaria at molecular level. METHODS: Genomic DNA from bulbs of several Fritillaria species was extracted and the polymorphisms of the D2 and D3 regions inside the 26S rDNA gene were identified by direct sequencing. Oligonucleotide probes specific for these polymorphisms were designed and printed on the poly-lysine coated slides to prepare the DNA chip. PCR products from the Fritillaria species were labeled with fluorescence by incorporation of dye-labeled dideoxyribonucleotides and hybridized to the immobilized probes on the chip. RESULTS: The polymorphisms were used as markers for discrimination among various species. Specific oligonucleotide probes were designed and immobilized on a DNA chip. Differentiation of the various Fritillaria species was accomplished based on hybridization of fluorescent labeled PCR products with the DNA chip. CONCLUSION: The results demonstrated the reliability of using DNA chips to identify different species of Fritillaria, and the DNA chip technology can provide a rapid, high throughput tool for genotyping and quality assurance of the plant species verification.
