The antagonism of cholecystokinin octapeptide-8 to the peroxynitrite oxidation on a diabetic cataractal rat model.

BACKGROUND: Cataracts is considered be formed because of an abnormal glucose metabolic pathway or oxidative stress. We explored the damaging role of ONOO- and antagonism of cholecystokinin octapeptide-8 (CCK-8) in diabetic cataractal rat lenses. METHODS: A diabetic cataractal animal model was established by peritoneal injection of streptozotocine (STZ). Thirty-six normal SD rats were taken as control group; seventy-two were given STZ (45 mg/kg) and then divided into STZ group and CCK-8 group (peritoneal injection CCK-8). STZ induced diabetic rats were treated with CCK-8 for 60 days. Lenses were examined with slit lamp at 20, 40 and 60 days. Immunofluorescent staining and Western blot analysis were used for determining nitrotyrosine (NT, a marker for ONOO-). PT-PCR and gene array analysis were used for determining the expression of inducible nitric oxide synthetase mRNA (iNOS mRNA) in lens epithelium (LEC). RESULTS: STZ group rats developed lens opacity by 20 days that reached a high level by 60 days after STZ injection. CCK-8 group rats delayed the cataract formation. CCK-8 group rats delayed the cataract formation. There was no distinct expression of NT and iNOS mRNA in control group. In STZ group, there were distinct expression of NT and upregulation of iNOS mRNA; however, CCK-8 group showed weak expression of NT and downregulation of iNOS mRNA. CONCLUSIONS: NT, which may be a new form of oxidative stress, was expressed in diabetic rat LEC although CCK-8 could reverse NT damage in LEC. The results suggested that CCK-8 might be a useful therapeutic agent against diabetic cataract. The antagonizing mechanism of CCK-8 may be related to direct antagonism of ONOO- as well as its inhibition of the expression of iNOS mRNA for production of NO and therefore decrease in the formation of ONOO-.

[New trends of customizing intraocular lens choice].

Definition and basic principle of customizing intraocular lens choice are described. The importance of the choicing intraocular lens for individual patient is illustrated. The review emphasizes that it is surgeon's responsibility to choice the appropriate intraocular lens for the best postoperative outcome and patient's satisfaction.

[The dynamic study of retinoic acid and its alginate sodium microspheres in rabbit eye].

OBJECTIVE: To obtain a intravitreous delay release system by injection of biodegradable polymers (alginate sodium-retinoic acid microspheres, AGS-RA microspheres) to the rabbit vitreous and study the release character of RA delivery system in vitro. METHODS: RA was mixed with 1.5% alginate sodium and microspheres was made by a special electrostatic generator after dissolved in organic solvent; The content of AGS-RA in the microspheres and its release from the microspheres were measured by spectrophotometer. The characteristic of the metabolic dynamics of AGS-RA in the microspheres was analyzed by HPLC. RESULTS: The size of microspheres we manufacture is (95.2443 +/- 8.6265) microm; Medicine contain quantity of RA is (1.7644 +/- 0.0453) microg/mg; Vitro test show that RA in the microspheres is release evenly in the 28 days of observation; Inject medicine to vitreous cavity show no poisonous side effect; The medicine dynamics research of microspheres showed that medicine is being even in 6 weeks releasing [the aqueous RA density of 1, 3 d, 1, 2, 3, 4, 5, 6 weeks is (23.79 +/- 0.15), (33.45 +/- 0.48), (19.95 +/- 0.79), (21.12 +/- 0.47), (19.65 +/- 0.35), (20.01 +/- 0.25), (18.24 +/- 0.27), (18.5 +/- 0.68) ng/ml, respectively]. Just in the 3rd day there is a release summit. CONCLUSION: In vivo and in vitro tests show that AGS-RA microspheres can delay RA release uniformity.

[Outline of benefit and disadvantage on intraocular refractive surgery].

The history, classification and current applications of intraocular refractive surgery (IRS) are expatiated. This review emphasizes the importance of the advantages and the disadvantages of IRS and patients selection criteria. Advices is brought forward to advance healthful expansion of IRS.

[Recent advances of cataract surgery and intraocular lens studies].

Recent advances of cataract surgery and intraocular lens (IOL) studies are reviewed briefly. This article reviews the progress in cataract surgery (technique, instruments and equipments), intraocular refractive surgery, IOL, the advantages and the disadvantages of various new techniques in the last 5 years. The importance of establishment of a patient selection criteria and a standard procedure are important for the development of a new intervention. New technology in the field of ophthalmology should be developed actively and carefully.

[Expression and signification of pigment epithelium-derived factor in experimental choroidal neovascularization of rat].

OBJECTIVE: To investigate the expression and signification of pigment epithelium-derived factor in choroidal neovascularization (CNV) induced by krypton laser on Brown Norway rat. METHODS: Five groups of 30 rats were photocoagulated by krypton laser in single eye to induce CNV. 3 days, 1 week, 2 weeks, 3 weeks and 4 weeks after photocoagulation, The retina was processed for histopathology, in situ hybridization of PEDF mRNA, immunohistochemistry of PEDF and FVIIIR:Ag. RESULTS: CNV was formed 1 week after photocoagulation, and reached its peak at 3 weeks. PEDF mRNA, PEDF expressed in the ganglion cells, inner nuclear layer, retinal pigment epithelial cells in normal BN retina. After photocoagulation, the expression of PEDF mRNA and PEDF was observed in the ganglion cells, inner nuclear layer, retinal pigment epithelial cells, and the lesions of outer nuclear layer and choroid; the expression of PEDF in the lesions was weaker than that at the edge of the lesions. From 3 days to 4 weeks, the expression density of FVIIIR:Ag gradually increased, however, the expression density of PEDF mRNA and PEDF in the lesions decreased (P < 0.05). The expression of FVIIIR:Ag was inversely correlated with that of PEDF during the development of CNV (r = -0.84, P < 0.05). CONCLUSIONS: CNV is inversely correlated with PEDF. The down-regulation of PEDF may be one of the main reasons for the formation and development of CNV.

[Peroxynitrite-induced formation of diabetic cataract and its prevention by puerarin in rat].

OBJECTIVE: To investigate the effect of peroxynitrite on the formation of diabetic cataract and its reversal by puerarin. in rat. METHODS: Normal Sprague-Dawley (SD) rats were equally divided into control group, streptozotocin group (STZ) and treatment group. STZ and treatment group were treated with STZ to establish animal model of diabetic rat cataract and in treatment group puerarin was given by peritoneal injection. General condition and lens shape of rats were examined at 20th, 40th, 60th day respectively. Lens epithelial cells (LEC) were observed with optical microscope. percentage of apoptotic cells, and fluorescent intensity of positive cells for nitrotyrosine (NT) which is the symbol of peroxynitrite were tested by flow cytometry. The expression of NT in the lens was analyzed by Western blot analysis. RESULTS: On 20th, 40th, 60th day of the experiment, clear, mild and severe lens opacity were found in control group, treatment group, and STZ group respectively and the lens opacity was gradually increased in all of rats included in the study. Under optical microscope, no changes were presented in control group, but mild changes were showed in treatment group and remarkable changes were found in STZ group. On all the specified days, distinguishable differences were found in the amount and percentage of apoptotic cells compared with that of control group in treatment group and STZ groups as well as between treatment and STZ groups (P < 0.05). There was significant increase in the fluorescent intensity and amount of NT in treatment and STZ groups compared with that of control group (P < 0.05), and the similar results were revealed between treatment group and STZ. The amount of expression of NT protein at different time points in different groups as described above was also significant difference (P < 0.001). CONCLUSIONS: Peroxynitrite plays an important role in the formation of diabetic rat cataract and is a participant in the pathogenesis of oxidative stress induced cataract. Puerarin is an effective antioxidative agent in the treatment of early diabetic rat cataract.

[Expression of vascular endothelial growth factor and its receptor in experimental choroidal neovascularization in rat].

OBJECTIVE: To study the expressions of vascular endothelial growth factor and its receptor FLK1 in krypton laser-induced choroidal neovascularization (CNV) in Brown Norway rat. METHODS: Thirty anesthetized male Brown Norway rats received krypton laser (647 nm, 360 mW, 50 micro m, 0.05 s) to induce CNV. Fundus fluorescein angiography examinations were performed just before euthanasia on days 3, 7, 14, 21, 28 and 56 after laser photocoagulation. The retina was processed for histopathology analysis. VEGF mRNA, VEGF and FLK1 expressions were demonstrated using in situ hybridization and immunohistochemistry staining respectively. RESULTS: VEGF mRNA expression was mainly observed in the vascular endothelial cells, the ganglion cells, the inner nuclear layers, the retinal pigment epithelial cells in normal retina and the vascular endothelial cells of normal choroid of BN rat. VEGF mRNA expressed in the vascular endothelial cells, the ganglion cells, the inner nuclear layers, lesion of outer nuclear layers in retina on days 3 after photocoagulation, but CNV wasn't detected. The level of VEGF mRNA expression in retina was decreased after 3 days (P < 0.01). CNV was firstly observed on day 7 after photocoagulation by FFA and histopathology. The area and density of positively stained cells for VEGF mRNA in CNV were increased during the development of CNV (P < 0.01) and no significant change after days 21 (P > 0.05). FLK1 was detected in the vascular endothelial cells, the ganglion cells of normal retina and the vascular endothelial cells of choroid. The staining of FLK1 was positive in laser induced CNV after days 7, the level of the receptor expression was increased during the development of CNV (P < 0.01) and was not significantly changed after days 21 (P > 0.05). CONCLUSIONS: VEGF and its receptor FLK1 accumulation and combination in the lesion of retina and choroid are related to choroidal neovascularization induced by laser.

[Study on the experimental model of krypton laser-induced choroidal neovascularization in the rats].

OBJECTIVE: To evaluate the feasibility of krypton laser-induced choroidal neovascularization (CNV) model in the Brown Norway rats in order to establish the foundation for studying the mechanism of CNV and for the development of treatment for CNV. METHODS: Experimental eyes in 33 rats were received a series of 10 krypton red laser (647 nm) lesions per eye (360 mW, 50 micro m, 0.05 s). Fundus fluorescein angiography (FFA) and indocyanine green angiography (ICGA) were performed, then the rats were sacrificed immediately, the eyes were enucleated and processed for histopathologic examination and transmission electron microscopy on days 3, 7, 14, 21, 28 and 56. RESULTS: CNV was firstly appeared on day 7 after photocoagulation, reaching the peak on day 21. The incidence of CNV was 76.0%. Disciform leakage staining appeared in the FFA. ICGA showed that the disciform area was filled with CNV. CNV was ascertained by light microscopy and electron microscopy. The thickness of laser-induced CNV was increased from day 7 to day 21 (P < 0.01) and no significant progress occurred after day 21 (P > 0.05). CONCLUSIONS: The present studies demonstrated that krypton laser photocoagulation could be used to produce choroidal neovascularization experimental model in the Brown Norway rat. CNV can be induced rapidly by the method, persists for a long period and at a high successful rate.

[Optical coherence tomography features of central exudative chorioretinopathy].

OBJECTIVE: To study the morphological features of submacular choroidal neovascularization in central exudative chorioretinopathy (CEC) using optical coherence tomography (OCT). METHODS: Using OCT, we examined 21 consecutive eyes (20 patients; 8 men, 12 women; mean age 38 years) with central exudative chorioretinopathy. Results of OCT were compared with fundus fluorescein angiography (FFA) and indocyanine green angiography (ICGA). RESULTS: Choroidal neovascularization appeared in subsensory retinal space anterior to the retinal pigment epithelium as a highly or moderately reflective mass, which protruded from the retinal pigment epithelium in 16 of 21 eyes. In five eyes, choroidal neovascularization appeared as a highly reflective fusiform or irregular mass at the level of retinal pigment epithelium and choriocapillaris. Nine of 21 eyes were associated with serous neurosensory retina detachment, 6 eyes with hemorrhagic pigment epithelium detachment (PED) and 14 eyes with edema and thickening of sensory retina. In 3-12 months (mean 6 months), choroidal neovascularization of 17 eyes with PDT regressed and its morphological features changed. The results obtained from OCT, FFA and ICGA could be supplemented with each other. CONCLUSIONS: OCT can be used to confirm the morphological features, location and size of submacular choroidal neovascularization. Choroidal neovascularization appears as a highly or moderately reflective round mass which protrudes from the retinal pigment epithelium in most cases. The morphology and the size of this mass may change after treatment and during follow-up.
