Library-based, multiplexed strategy for mapping protein interaction networks via crosslinking.

BACKGROUND: Protein-protein interactions are fundamental to cellular function, yet resolving their interaction interfaces and dynamic behaviors in native biological contexts remains challenging, particularly for weak or transient interactions. Crosslinking strategies based on noncanonical amino acids offer an effective means to capture such interactions; however, traditional single-site incorporation provides limited coverage and may overlook critical interaction hotspots. RESULTS: By employing a mutagenesis library, multiple interaction partners and cross-linking sites of a target protein can be simultaneously screened in a single experiment, without prior knowledge of its precise structural or functional features, enabling effective and unbiased analysis of its interaction network. In this study, we constructed an amber codon-scanning mutagenesis library of PSMD10, facilitating independent incorporation of the photocrosslinking ncAA p-azido-phenylalanine at multiple distinct residues. This approach allowed us to systematically interrogate and precisely map potential interaction regions across the protein surface. Coupled with crosslinking mass spectrometry, we identified multiple residues involved in intermolecular interactions, as well as previously unreported interaction partners, including T2FA, TBA1C, and ATRIP. CONCLUSIONS: These findings expand our understanding of PSMD10-associated proteasome interactome, demonstrate a multiplexed strategy for in situ mapping of protein interaction interfaces with broad coverage, and offer a valuable platform for developing therapeutics that target protein-protein interactions.

Comparative studies on histological and ultra-structure of the pituitary of different ploidy level fishes.

The histological and ultra-structure of the pituitary in diploid red crucian carp (Carassius auratus red var.), triploid crucian carp and allotetraploid hybrids within and after the breeding season were comparatively studied. The result showed that there were six endocrine cell types in the pituitary of these three kinds of fishes, and there was an obvious difference in cell size among different ploidy level fishes. As for the same type of pituitary cells, the cell size was increased gradually with the increasing ploidy level. In the breeding season, the allotetraploid hybrids had higher proportion of gonadotropin cells (GTH) than triploids, and the triploids had higher proportion of GTH than diploids. The results were related to the earlier sexual maturity of allotetraploid hybrids and sterility of triploid crucian carp. On the other hand, among the three kinds of fishes, the proportion of somatotropin (STH) cells in triploids crucian carp was the highest, whereas that in allotetraploid hybrids was the lowest. The results might be connected with the faster growth rate of triploids and slower growth rate of allotetraploid hybrids. In addition, in GTH cells of meso-adenohypophysis after the breeding season, there were many endocrine particles in triploids, while those endocrine particles were released from the cells in allotetraploids and diploids. This result showed that the sterility of triploid crucian carp might be related to the hormone which was not released from the GTH cells. In a word, the present study indicated that the differences in the structure of pituitary among different ploidy level fishes contributed to their difference in the growth rate and gonadal development.

Penetrating keratoplasty combined with vitreoretinal surgery for severe ocular injury with blood-stained cornea and no light perception.

OBJECTIVE: To evaluate the effects of treatment of severe ocular injury with blood-stained cornea and no light perception by combined penetrating keratoplasty and vitreoretinal surgery, and to analyze the relevant factors. METHODS: Records of 7 severely injured eyes of 7 patients with blood-stained cornea and no light perception who underwent penetrating keratoplasty combined with vitrectomy using a temporary keratoprosthesis were evaluated retrospectively. The preoperative visual acuity was no light perception in all injured eyes with a mean intraocular pressure of 3 mm Hg and a range from 2 to 5 mm Hg. The average interval from emergency wound closure to vitrectomy was 18 days with a range from 12 to 21 days. The mean follow-up was 28 months with a range from 26 to 30 months. RESULTS: The postoperative visual acuity was better than light perception in 5 eyes with the best corrected visual acuity from light perception to 0.06. The retina was attached in 5 eyes. The postoperative intraocular pressure ranged from 5 to 15 mm Hg with a mean of 12 mm Hg; it was significantly higher than the preoperative one (p < 0.05). The postoperative complications mainly included temporary intraocular elevation (1 eye), corneal neovascularization (4 eyes), corneal rejection (4 eyes), retinal detachment (2 eyes) and ocular atrophy (2 eyes). CONCLUSION: Penetrating keratoplasty combined with vitrectomy using a temporary keratoprosthesis is a safe and effective method in treating severe ocular injury with blood-stained cornea and no light perception.

Reduction of bronchopulmonary dysplasia after participation in the Breathsavers Group of the Vermont Oxford Network Neonatal Intensive Care Quality Improvement Collaborative.

OBJECTIVE: The objective of this study was to compare the primary and secondary outcomes of very low birth weight infants before and after participation in the Breathsavers Group of the Vermont Oxford Network-sponsored Neonatal Intensive Care Quality Collaborative. METHODS: Hospitals that participated in the Breathsavers Group contributed clinical data on the outcomes of their very low birth weight infants to the Vermont Oxford Network using standardized clinical definitions, data forms, and inclusion criteria. Outcomes from the last year of the collaborative, 2003, were compared with those from the baseline year, 2001. Models for treatment practices and outcomes measures were adjusted for within-hospital correlation (clustering) and standard risk factors that were present at birth. RESULTS: Bronchopulmonary dysplasia dropped significantly in 2003 compared with the baseline year. Survival improved but not significantly. In addition, severe retinopathy of prematurity, severe intraventricular hemorrhage, and supplemental oxygen at discharge dropped significantly. The use of conventional ventilation at any time during the initial hospitalization, postnatal steroids, and time to first dose of surfactant all decreased significantly. The use of nasal continuous positive airway pressure at any time during hospitalization increased. The use of high-frequency ventilation, delivery room intubation, and surfactant at any time during hospitalization did not change. CONCLUSIONS: The Breathsavers Group improved both clinical care processes and clinical outcomes during the Neonatal Intensive Care Quality Collaborative.

Constitutive overexpression of multiple cytochrome P450 genes associated with pyrethroid resistance in Helicoverpa armigera.

Our previous studies showed that enhanced cytochrome P450-mediated detoxification is a major mechanism responsible for pyrethroid resistance in a laboratory-selected strain (YGF) of Helicoverpa armigera (Hübner). Two new cytochrome P450 genes, CYP9A12 and CYP9A14 (encoding 531 and 530 amino acid residues, respectively) were isolated from the fat body of the YGF strain of H. armigera. The mRNA expression levels of these two CYP9 P450 genes, together with CYP6B7 and CYP4G8 (previously reported to be overexpressed in pyrethroid-resistant strains), were assessed by real-time quantitative reverse transcription-polymerase chain reaction in the final instars of a field-derived YG strain (with seven-fold resistance to the pyrethroid fenvalerate) and the YGF strain (selected from the YG strain with fenvalerate in the laboratory, with 1,690-fold resistance to fenvalerate). The mRNA expression levels of CYP9A12, CYP9A14, and CYP6B7 in the fat body of the YGF strain increased to 433-, 59-, and 9.3-fold, respectively, compared with the YG strain, whereas no overexpression was revealed for CYP4G8. CYP9A12 and CYP9A14 had 19- and 4.3-fold overexpression in the midgut of the YGF strain compared with the YG strain, respectively, but CYP6B7 and CYP4G8 were not overexpressed. The current study provided evidence that constitutive overexpression of multiple cytochrome P450 genes (CYP9A12, CYP9A14, and CYP6B7) is associated with pyrethroid resistance in H. armigera.

[Analysis of apposition on differently treated implant surface at early stage of implantation].

OBJECTIVE: To compare the surface apposition and the bone response at early period of implantation in two differently treated implants. METHODS: The implants were subject to double acid-etched-H2O2/HCl-heat treatment and double acid-etching treatment, and then randomly implanted into the tibia of rabbits. After 2, 4, 8 weeks of follow up, the bone specimens containing implants were prepared and examined by a field emission SEM and EDX. RESULT: A layer rich with calcium and phosphorus was clearly demonstrated on the implants surface of both groups after 2 weeks of implantation, but it was mostly disappeared after 4 weeks. There were large amounts of osteoblasts cells on double acid-etched-H2O2/HCl-heat treated implants surface indicating the initiation of osteogenesis. After 8 weeks of implantation some new bones were attached on the implants surface in both groups, more bones attached were shown on double acid-etched- H2O2/HCl-heat treated implants surface. CONCLUSION: A calcium and phosphorus-rich layer was formed on the implants surface of both groups at early period of implantation.

[Experimental study of fast formation of biomimetic apatite coatings on pure titanium surface].

OBJECTIVE: To investigate calcium-phosphate coating on commercial pure titanium substrate using a fast biomimetic procedure. METHODS: The titanium specimens were divided into two groups, group A and group B. The specimens of group A were only treated with the mixture of H2SO4/HCl for 30 min. The specimens of group B were treated with the mixture of H2SO4/HCl for 30 min, immersed into 5 mol/L NaOH solution at 60 degrees C for 24 h, and then heated to 600 degrees C and maintained for 1 h. All specimens were soaked into simulated body fluid-A (SBF) and SBF-B for each day. The surface morphology was observed by field scanning electron microscopy. RESULTS: Dense calcium-phosphate coating deposited on all the titanium surfaces of two groups. The calcium-phosphate coating consisted of spheric structure with a diameter of 1 approximately 3 microm, which was proved to be hydroxycarbonate apatite with the analysis of X-ray diffraction. CONCLUSIONS: Thin hydroxycarbonate apatite coating can deposited on pure titanium using a fast biomimetic procedure.

Importance of polyunsaturated acyl chains in chlorpromazine interaction with phosphatidylserines: a 13C and 31P solid-state NMR study.

The polyunsaturated fatty acid docosahexaenoic acid (DHA, c22:6, n-3) is found at a level of about 50% in the phospholipids of neuronal tissue membranes and appears to be crucial to human health. Dipalmitoyl phosphatidylcholine (DPPC, 16:0/16:0 PC), 1-palmitoyl-2-oleoyl phosphatidylserine (POPS) and the DHA containing 1-stearaoyl-2-docosahexenoyl phosphatidylserine (SDPS) were used to make DPPC (60%)/POPS (29%)/SDPS (11%) bilayers with and without 10 mol% chlorpromazine (CPZ), a cationic, amphiphilic phenothiazine. The T1 relaxation measurements make it clear that the saturated acyl chains carbons (palmitic, stearic and most of the oleic chain) and the choline head group are not affected by CPZ addition. The observed increased signal intensity and T1-values of DHA indicate reduced mobility of C4 and C5 due to CPZ binding. 31P NMR spectra confirm that CPZ binding to the phosphatidylserines in the bilayer enhances phospholipid head group mobility.

Multigene tracking of quasispecies in viral persistence and clearance of hepatitis C virus.

AIM: To investigate the evaluation of hepatitis C virus (HCV) quasispecies in the envelope region and its relationship with the outcome of acute hepatitis C. METHODS: HCV quasispecies were characterized in specimens collected every 2-6 mo from a cohort of acutely HCV-infected subjects. We evaluated two individuals who spontaneously cleared viremia and three individuals with persistent viremia by cloning 33 1-kb amplicons that spanned E1 and the 5' half of E2, including hypervariable region 1 (HVR1). To assess the quasispecies complexity and to detect variants for sequencing, 33 cloned cDNAs representing each specimen were assessed by a combined method of analysis of a single-stranded conformational polymorphism and heteroduplex analysis. The rates of both synonymous and nonsynonymous substitutions for the E1, HVR1 and E2 regions outside HVR1 were analyzed. RESULTS: Serum samples collected from chronic phase of infection had higher quasispecies complexity than those collected from acute phase of infection in all individuals examined. The genetic diversity (genetic distance) within HVR1 was consistently higher than that in the complete E1 (0.0322+/-0.0068 vs -0.0020+/-0.0014, P<0.05) and E2 regions outside HVR1 (0.0322+/-0.0068 vs 0.0017+/-0.0011, P<0.05) in individuals with persistent viremia, but did not change markedly over time in those with clearance of viremia. For individuals with persistent viremia, the rate of nonsynonymous substitutions within the HVR1 region (2.76X10(-3)+/-1.51X10(-3)) predominated and gradually increased, as compared with that in the E1 and E2 regions outside HVR1 (0.23X10(-3)+/-0.15X10(-3), 0.50X10(-3)+/-0.10X10(-3)). By contrast, the rates of both nonsynonymous and synonymous substitutions for the E1 and E2 regions including HVR1 were consistently lower in individuals with clearance of viremia. CONCLUSION: HCV persistence is associated with a complexity quasispecies and positive selection of HVR1 by the host immune system.

Correlation between fenvalerate resistance and cytochrome P450-mediated O-demethylation activity in Helicoverpa armigera (Lepidoptera: Noctuidae).

Cytochrome P450 monooxygenases are a major metabolic mechanism responsible for pyrethroid resistance in Helicoverpa armigera (Hübner) from Asia. Cytochrome P450-mediated O-demethylation activity toward p-nitroanisole (PNOD) of individual fourth instars was determined in five strains of H. armigera by using a microplate reader. The four resistant strains of YS, HD, YGF, and YG59 had 6-, 71-, 2540-, and 11,800-fold resistance, respectively, to fenvalerate in comparison with the susceptible BK77 strain. Their mean PNOD activity was 4-, 10-, 24-, and 60-fold, respectively, compared with the BK77 strain. A strong positive correlation (correlation coefficient r = 0.98) between PNOD activity and fenvalerate resistance was found. Of 48 larvae from each strain, only 4% larvae of the susceptible BK77 strain had detectable PNOD activity, whereas 25, 33, 79, and 96% of larvae from the resistant strains YS, HD, YGF, and YG59 exhibited PNOD activity, respectively. There was a clear discrimination of patterns of PNOD frequency distribution between H. armigera strains and their magnitudes of fenvalerate resistance. The PNOD activity can be used as a biochemical marker for monooxygenase-mediated pyrethroid resistance in field populations of H. armigera.

[Clinical and pathological characterization of the overlap syndrome of primary biliary cirrhosis and autoimmune hepatitis].

OBJECTIVE: To analyze the clinical and pathological characteristics of primary biliary cirrhosis (PBC) and autoimmune hepatitis (AIH) overlap syndrome. METHODS: Of our 68 patients with the diagnosis of PBC, we identified 9 overlap syndrome cases strictly using the revised descriptive criteria and scoring system for diagnosis of AIH proposed by the International Autoimmune Hepatitis Group Report. The clinical manifestations and pathological changes shown in liver biopsies from the overlap syndrome and pure PBC were analyzed and compared. RESULTS: The mean aggregate scores of the 9 cases (13.2%) of the overlap syndrome group and 59 cases of pure PBC (86.8%) were 10.2+/-0.2 and 4.7+/-0.7 respectively among the 68 total. The serum levels of ALT and AST, immunoglobulin G, gammaglobulin, in the overlap syndrome group were significantly higher than those in the pure PBC group, and also there was a frequent presence of antinuclear antibody and/or smooth muscle antibody positivity in the overlap syndrome group. Histopathologically the livers in the overlap syndrome group showed combined features of interface hepatitis and piecemeal necrosis, characterizing the two diseases. CONCLUSION: The overlap syndrome group showed combined features of both PBC and AIH. There were differences in clinical aspects, serology and histology between the overlap syndrome and pure PBC groups.

[Analysis of protamine content in patients with asthenozoospermia].

OBJECTIVE: To study the significance of the change of protamine content in the management of male infertility. METHODS: Sperm nuclear proteins extracted from 199 infertile patients were analyzed by polyacrylamide gel-electrophoresis and scanning microdensitometry. RESULTS: Forty-two (21%) cases of the total number had normal sperm nuclear proteins and 157 (79%) had aberrant ones, which mainly presented the interruption of HPRR and abnormality of P2 protamine. Thirty patients were selected at random from 157 abnormal cases for clinical treatment. After the treatment, the sperm nuclear proteins were extracted and analyzed and the results demonstrated that 11 cases (36.6%) improved markedly, 5 (16.6%) restored to normal and the other 16 (46.6%) remained unchanged. CONCLUSION: There is a reduced level or selective absence or even complete selective absence of protamines in infertile patients. Protamines may act as a parameter for evaluating the treatment effect of infertile males and protamine content can be influenced by a certain or several factors.

[The preparation of the multilevel holes and analysis of the pure titanium surface].

PURPOSE: Using sandblast, double acid-etching and H(2)O(2)/HCl solution treatment to establish a new implant surface, and to analyze the surface's morphology. METHODS: After being polished, sandblasted, cleaned, the titanium specimens were divided into six groups, i.e.A to F group. The specimens of group A were treated with low concentration of HF/HNO(3) solution, Group B was treated with low concentration of HF/HNO(3) solution and then H(2)SO(4)/HCl solution, Group C was treated after Group B and further with H(2)O(2)/HCl solution at 80degrees centigrade for 30 minutes, and subsequently heated at 400degrees centigrade for 1 hour. Group D was treated with high concentration of HF/HNO(3) solution, Group E was treated after Group D and then H(2)SO(4)/HCl solution, Group F was treated after E group and further with H(2)O(2)/HCl solution at 80degrees centigrade for 30 minutes, and subsequenty heated at 400degrees centigrade for 1 hour. RESULTS: The titanium surface sandblasted and ultrasonically cleaned exhibited a great deal of particles. The titanium plates treated with low concentration of HF/HNO(3) solution still had some particles, but there were much nano-holes on the titanium surface. The titanium plates treated with high concentration of HF/HNO(3) solution had no particles on the surface, but showed much micron-holes on the titanium surface. The titanium plates treated with H(2)SO(4)/HCl solution after HF/HNO(3) solution etching had all particles removed and got multilevel holes on the surface. The last H(2)O(2)/HCl method produced an anatase titania gel layer on the specimen surface. CONCLUSION: By altering the concentration of HF/HNO(3) solution, we could make different sizes of apertures on the titanium surface. Double acid-etching can remove all sandblasting particles embedded in titanium surfaces and form multilevel holes. The H(2)O(2)/HCl method can turn the rough non- crystallographic TiO(2) layer into the anatase titania gel layer.

An improved, inexpensive procedure for the large-scale purification of recombinant human erythropoietin.

A rapid and simple chromatographic procedure has been developed for the large-scale purification of therapeutic-grade rHuEPO (recombinant human erythropoietin) from medium-conditioned cell cultures, which includes ion-exchange, hydrophobic-interaction and gel-filtration chromatography. A combination of these well-connected steps results in highly purified rHuEPO (> 99%), as revealed by SDS/PAGE and HPLC analyses, with a total yield of 38%. The specific activity of purified rHuEPO was 160,104 i.u./mg. Immunoblotting studies revealed that the protein possesses native EPO immunity. N-terminal sequencing of rHuEPO shows that the first 15 amino acids coincide with those of native EPO reported previously.
