A color version of the Hinsberg test: 1 degrees -3 degrees amine indicator.

The Hinsberg test to recognize the type of amine (1 degrees -3 degrees amines), which has been established for more than 100 years and well documented in textbooks, is not possible without conducting complicated organic reactions. We report for the first time unique chemosensors that are capable of showing selective color changes toward 1 degrees -3 degrees amines as a color version of the Hinsberg test. This simple and straightforward qualitative analysis, using the synthesized novel compounds herein, can be considered a new innovative tool for discriminating 1 degrees -3 degrees amines as an alternative to the historical Hinsberg test.

alpha-CD/crown-appended diazophenol for selective sensing of amines.

[reaction: see text] Azophenol dyes having the permethylated cyclodextrin and/or crown moieties have been synthesized. Compound 1 provides critical information on discriminating 1-3 degrees amines with unique color changes. Addition of 1 degrees and 2 degrees amines to 1 shifts the absorbance maximum of 1 from 380 to approximately 580 and approximately 530 nm, respectively, but no change is observed with 3 degrees amines. The high selectivity of 1 is mainly due to H-bonding between the ammonium H atoms of the amine and oxygen atoms of the crown-6.

Hydrogel behavior of a sugar-based gelator by introduction of an unsaturated moiety as a hydrophobic group.

The new sugar-based gelators 1 and 2 were synthesized, and their gelation abilities were evaluated in organic solvents and in water. Compound 1 gelates both water and organic solvents whereas 2 gelates only organic solvents. Superstructural difference between hydrogel 1 and organogel 2 was investigated by CD, TEM, AFM, 1H NMR and XRD. Hydrogel 1 displays a well-developed helical ribbon structure with 20-150 nm diameter and a length of several hundred microm whereas organogel 2 shows a twisted fiber structure of diameter 20 nm. CD measurements of hydrogel 1 and organogel 2 indicate that hydrogel 1 maintains a well-ordered chiral structure whereas organogel 2 maintains a relatively disordered chiral structure. The 1H NMR and XRD results suggest that the hydrophobic interaction in hydrogel 1 are relatively weak, with a relatively small region interdigitated between lipophilic alkyl groups. In addition, upon irradiation at 254 nm wavelength, hydrogel 1 reveals a red coloration at 540 nm. These results indicate that the self-assembled hydrogel 1 was polymerized by UV-irradiation. The intensity of the CD spectrum of the polymerized hydrogel markedly decreased. This result indicates that upon polymerization the highly ordered chiral structure of hydrogel 1 changes to a disordered molecular packing structure.

A chromogenic macrocycle exhibiting cation-selective and anion-controlled color change: an approach to understanding structure-color relationships.

[reaction: see text] A N-azo-coupled macrocycle (1) with a NO(2)S(2) donor set was synthesized as a chromoionophore and shown to exhibit Hg(2+) selectivity; anion control of the color of the Hg(2+) complex was observed. As a first approach, the nature of the color generation process was probed by determining the crystal structures of the two different colored species obtained with perchlorate and iodide anions. The results can be explained in terms of a "Push-n-Pull Process", which serves to illustrate how the coordinating ability of the anion controls the color change through formation of endo- or exo-metal complexes. The use of "simple" salts to induce color-switching of the above complex species was also reported.

MAPK regulation and caspase activation are required in DMNQ S-52 induced apoptosis in Lewis lung carcinoma cells.

6-(1-Hydroxyimino-4-methylpentyl)5,8-dimethyoxy 1,4-naphthoquinone S-52 (DMNQ S-52) was reported to have cytotoxic activity against L1210 leukemia cells. In the present study, we investigated the apoptotic mechanism of DMNQ S-52 in vitro and in vivo in murine solid cancer cells. DMNQ S-52 exerted cytotoxicity against Lewis lung carcinoma (LLC) cells (IC50=12.3 microM). DMNQ S-52 increased Annexin V positive cell population in a concentration-dependent manner. DMNQ S-52 also induced apoptosis through caspase-mediated pathway, including activation of caspase-3, cleavage of Poly(ADP-ribose) polymerase (PARP) and decreased expression of Bcl-2 in LLC cells in a time and concentration-dependent fashion. DMNQ S-52 activated the phosphorylation of c-Jun N-terminal kinase (JNK) and p38 as well as abrogated the expression of extracellular signal-regulated kinase (ERK) in a time-dependent manner at 10 microM. Similarly, cell proliferation inhibition by DMNQ S-52 was masked by caspase inhibitor Z-Asp-Glu-Val-Asp-fluoromethylketone (Z-VAD-FMK), JNK inhibitor SP600125 and p38 inhibitor SB203580, but not by MEK inhibitor U0126. Furthermore, i.p. administration of DMNQ S-52 at 5 mg/kg resulted in a potent inhibition of the growth of LLC cells implanted on the right flank of C57BL/6 mice compared to untreated control. Immunohistochemical analysis revealed the decreased tumor cell proliferation and increased tumor cell apoptosis in DMNQ S-52 treated tumor sections using terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling (TUNEL) and proliferation cell nuclear antigen (PCNA). Taken together, these findings demonstrate that DMNQ S-52 may exhibit anti-tumor activity by inducing apoptosis via caspases and mitogen activated protein (MAP) kinase-dependent pathways.

The relationship between the biogenetic temperament and character and psychopathology in adolescents.

BACKGROUND: This study was conducted to investigate the relationship between patterns of temperament and character and self-reported psychopathology in adolescents from the community. SAMPLING AND METHODS: The Junior Temperament and Character Inventory (JTCI) and Youth Self-Report (YSR) instruments were administered to 623 Korean middle school students (boys/girls = 331/292; age = 13.3 +/- 0.9 years old). Multiple linear regression analyses were used to evaluate the relationship between temperament and character based on Cloninger's biogenetic theory of personality and youth psychopathology of internalizing and externalizing problems. RESULTS: Internalizing problems of the YSR (withdrawn, somatic complaint and anxious/depressed scales) were significantly related to high Harm Avoidance and low Reward Dependence (JTCI temperament) and low Self-Directedness and high Self-Transcendence (JTCI character). Externalizing problems of the YSR (delinquency and aggression) were significantly associated with high Novelty Seeking and high Harm Avoidance (JTCI temperament) and low Self-Directedness, low Cooperativeness and high Self-Transcendence (JTCI character). CONCLUSIONS: Findings of the current study suggest that specific patterns of temperament and character potentially influence adolescents' psychopathology in the community.

Effects of mycelial culture of Phellinus linteus on ethanol-induced gastric ulcer in rats.

The gastroprotective effects of a mycelial culture of Phellinus linteus (MCPL) were evaluated by determining the ulcer index, gastric mucus content, histopathological observation and histochemical properties of mucin in an ethanol-induced ulcer model of rats. Preadministration with MCPL at doses of 20 and 60 mg/kg, showed a significant decrease of bleeding and ulcer index and alleviated the histopathological changes induced by ethanol such as hemorrhage and necrosis. Ethanol treatment decreased the gastric adhesion mucus content, but a higher level of gastric mucus persisted after preadministration of MCPL. As for the histochemical properties of mucins, marked changes were observed in both the surface and gland mucous cells in ethanol-treated rats, but these changes were detected only in the surface mucous cells in rat preadministered with MCPL. Using conventional methods for mucins, ethanol-treated rats revealed a decrease of neutral and acid mucin in the surface epithelium and mucous neck cells compared with normal rats. A marked decrease of BSL-1 by lectin histochemistry was also revealed in the ethanol-treated rats. But the MCPL preadministered rats showed similar stainabilities and lectin affinity patterns for mucins as the normal rats. These results indicate that pretreatment with MCPL provided protection of the gastric mucosa from ethanol-induced injury by maintaining the mucus barrier in rats.

Fabrication of color-tunable luminescent silica nanotubes loaded with functional dyes using a sol-gel cocondensation method.

Luminescent silica nanotubes SNT-2 (loaded with coumarion laser dye 2) and SNT-3 (loaded with anthracene laser dye 3) were prepared by sol-gel cocondensation of functional dyes and TEOS in a cholesterol-based organogel system. The emission colors of silica nanotubes were tuned by using different functional dyes. Interestingly, there is a great difference in PL spectrum of silica nanotubes loaded with functional dyes between ethanol and the solid state. In ethanol, a green light emission of SNT-2 and a bluish green light emission of SNT-3 were observed at 486 and 465 nm, respectively, because of the anchoring state between the dye and the tube, whereas in the solid state, strong blue light emissions of SNT-2 and SNT-3 were observed at 482 and 483 nm, respectively, because of the intense emission of the silica nanotube (SNT) itself rather than that of 2 and 3.

TAC-101 inhibits intrahepatic metastasis of orthotopically implanted murine hepatocellular carcinoma.

The anti-metastatic effect of 4-[3,5-bis(trimethylsilyl)benzamido]benzoic acid (TAC-101) was investigated using our established intrahepatic metastasis model. Orthotopic implantation of a fragment of CBO140C12 hepatoma into the liver resulted in the formation of a solitary tumor nodule and its intrahepatic metastasis. Daily oral administration of TAC-101 at a dose of 8 mg/kg resulted in a significant inhibition of intrahepatic metastasis, but did not affect the growth of the tumor at the implanted site. The down-regulation of transcriptional anti-activator protein-1 (AP-1) activity by TAC-101 paralleled the inhibition of cell invasion and migration through the repression of expression of the mRNAs for urokinase-type plasminogen activator (u-PA) and its receptor (u-PAR). These findings suggest that TAC-101 may improve therapeutic efficacy for liver cancer patients to prevent intrahepatic metastasis.

Antimetastatic and immunomodulating properties of a new herbal prescription, Bojung-bangam-tang.

We investigated the antimetastatic effect of Bojung-bangam-tang, a new herbal prescription, on liver metastasis by the inoculation of colon 26-L5 carcinoma cells into the portal vein. Oral administration of Bojung-bangam-tang for 15 days from day 7 before tumor inoculation significantly inhibited liver metastasis in a dose-dependent manner. Bojung-bangam-tang enhanced the mitogenic activity of BALB/c whole splenocytes induced by various mitogenic stimuli. Oral administration of Bojung-bangam-tang, by itself, could not induce the production of interleukin (IL)-12 or IFN-gamma by macrophages, but enhanced the potential of macrophages to produce cytokines in response to lipopolysaccharide (LPS). Experiments using macrophage- or natural killer (NK) cell-deficient mice revealed that the antimetastatic effect of Bojung-bangam-tang is mediated by macrophages rather than NK cells. Bojung-bangam-tang caused a marked increase of production of Th1 cytokine (IFN-gamma) and decrease of production of Th2 cytokine (IL-4) by splenocytes stimulated with concanavalin A (Con A). These results indicated that oral administration of Bojung-bangam-tang inhibited the liver metastasis of colon 26-L5 cells through a mechanism leading to a Th1 dominant immune state and activation of macrophages. Thus, Bojung-bangam-tang may be useful for the prevention of cancer metastasis.

An alternative way to individualized medicine: psychological and physical traits of Sasang typology.

BACKGROUND: Disease susceptibility and drug response of individuals are presumed to be different depending on their personality traits. The Sasang typology, a traditional Korean medical typology, explains the individual differences of vulnerability to pathology and proposes guidelines for the safe and effective use of medical herbs depending on individual traits. OBJECTIVE: The purpose of the present study was to evaluate psychologic and physical characteristics of Sasang types from the perspective of personality theory. DESIGN: After determining the Sasang type of 79 college students based on the Questionnaire for the Sasang Constitution Classification, the psychologic and physical traits of each type were analyzed by the Meyers-Briggs Type Indicator (MBTI) and Bioelectrical Impedance Analysis, respectively. RESULTS: Each of the Sasang types showed significantly different profiles based on the MBTI scores (generalized estimation equation, coefficient = 11.88, z = 2.13, p = 0.033) and could be distinctively classified based on their MBTI scores (discriminant analysis Wilks' lambda = 0.611, df = 8, chi(2) = 36.7, p < 0.001). Subjects with the So-Eum type (Introversion and Judging) and the So-Yang type (Extroversion and Perceiving) showed contrasting psychologic features. However, they had similar anthropometric characteristics. Subjects with Tae-Eum type had relatively higher body fat mass. CONCLUSION: Current results demonstrated distinctive personality traits associated with Sasang types using reproducible psychometric and anthropometric instruments. With further study, the Sasang typology could serve as a scientific tool for individualized and integrative medicine.

Decrease in genu of the corpus callosum in medication-naïve, early-onset dysthymia and depressive personality disorder.

BACKGROUND: The purpose of this study was to detect differences in regional areas of the corpus callosum (CC) in subjects with early-onset minor depression (dysthymia or depressive personality disorder) and healthy comparison subjects. Based on previous reports that have suggested reduced frontal lobe volume and reduced hemispheric lateralization in the frontal regions of the brain in depression, we hypothesized that the area of the CC that interconnects the frontal regions of the brain, i.e., the genu, will be smaller compared to that of healthy comparison subjects. METHODS: Forty female subjects with early-onset dysthymia or depressive personality disorder, as defined by the Structured Clinical Interview for DSM-III-R and the Diagnostic Interview for Depressive Personality, respectively, and age- and gender-matched healthy comparison subjects (n = 42) were recruited (age: 21.4 +/- 2.1 and 20.9 +/- 2.8 years, respectively). All subjects were psychotropic medications-naïve and right-handed. A 1.5T GE Sigma scanner was used to acquire 124 1.5-mm-thick contiguous coronal images. Midsagittal slice images were carefully selected from reconstructed magnetic resonance images both from native and stereotaxic space to measure seven regional areas of the CC. RESULTS: There were significant diagnosis by CC region interactions [F(6,480) = 4.06, p <.001; F(6,480) = 3.30, p =.003, native and stereotaxic space, respectively]. Early-onset minor depression subjects had a 9.9% (native space) and 6.9% (stereotaxic space) smaller genu of the CC compared to the healthy comparison subjects (the Newman-Keuls post hoc test, p =.005 and.019, native and stereotaxic space, respectively). Early-onset minor depression subjects also had a 7.8% smaller posterior midbody relative to the comparison subjects (the Newman-Keuls post hoc test, p =.033) only in the native space. Severity of current depressive symptoms or duration of illness did not correlate with the size of the genu or the posterior midbody parts of the CC. CONCLUSIONS: These results suggest frontal lobe structural, and possibly functional, abnormalities in the brain in young female adults with a milder spectrum of depression, i.e., DSM-IV early-onset dysthymia or depressive personality disorder. The present findings point out the possible role of frontal lobe abnormality in pathophysiology of early-onset minor depression.

Relationship between T-wave normalization on exercise ECG and myocardial functional recovery in patients with acute myocardial infarction.

BACKGROUND: Several studies suggested that T-wave normalization (TWN) in exercise ECG indicates the presence of viable myocardium. But the clinical implication of this phenomenon in patients with acute myocardial infarction who received proper revascularization therapy was not determined. Precisely the aim of this study was to investigate the relationship between TWN in exercise ECG and myocardial functional recovery after acute myocardial infarction. METHODS: We studied 30 acute myocardial infarction patients with negative T waves in infarct related electrocardiographic leads and who had received successful revascularization therapy. Exercise ECG was performed 10-14 days after infarct onset using Naughton protocol. Patients were divided into 2 groups according to presence (group I; n = 14) or not (group II; n = 16) of TWN in exercise ECG. Exercise parameters and coronary angiographic findings were compared between groups. Baseline and follow-up (mean 11 months) regional and global left ventricular function was analyzed by echocardiography. RESULTS: Exercise parameters were similar between groups. There was no difference in baseline ejection fraction and wall motion score between group I and II (EF; 56 +/- 12% vs 52 +/- 11%, p = ns. WMS; 21 +/- 3 vs 23 +/- 4, p = ns) and it was improved at the tenth month by similar magnitude (group I/group II, EF% change = 12 +/- 12% vs 7 +/- 6%, p = ns, WMS% change = 6 +/- 6% vs 7 +/- 5%, p = ns). The finding of no relation between TWN and functional recovery was observed also when the patients were analysed according to infarct location and presence or absence of Q-waves. CONCLUSION: As the exercise-induced TWN in patients with acute myocardial infarction was not related with better functional recovery of dysfunctional regional wall motion and ejection fraction, TWN does not appear to be an indicator of myocardial viability.

The reliability and validity of the junior temperament and character inventory.

The purpose of this study was to evaluate the reliability and validity of the Junior Temperament and Character Inventory (Junior TCI), a child and adolescent version of the Adult Temperament and Character Inventory (TCI), which is based on Cloninger's biosocial model of personality. The Junior TCI was translated into Korean and administered to 663 Korean middle school students (male/female, 360/303; age, 13.3 +/- 1.0 years) and their parents (mother/father, 444/84). Internal consistency was calculated by Cronbach alpha. A test-retest study of the Junior TCI was conducted across a 3-month interval with 97 subjects. Factor analyses for the temperament and character dimensions were performed using principal component analysis, rotating factors by Promax. Cronbach alpha values of the Junior TCI scales ranged from.48 to.80 for the temperament scales and from.64 to.68 for the character scales. Test-retest correlations ranged from.62 to.85 for the temperament scales and from.76 to.79 for the character scales. Principal component factor analyses showed three factors out of four temperament scales (Harm Avoidance 1 to 4; Novelty Seeking 1 to 4; Persistence; and Reward Dependence 1, 3, 4) and three factors out of three character scales (Self-Directedness 1 to 5; Cooperativeness 1 to 5; and Self-Transcendence 1 to 3) that were similar to factor structures of adult versions of the TCI. Explorative factor analysis with a condition of eigenvalue greater than 1 produced six factors, as compared to seven factors extracted in Cloninger's original report of the adult TCI. In conclusion, this was the first study to tests and report detailed psychometric properties of the Junior TCI. The current study confirms that the Junior TCI has satisfactory psychometric properties for the use in child and adolescent populations and reflects Cloninger's biosocial model of personality.
