Hadamard transform ion mobility spectrometry.

A detection scheme that makes use of the Hadamard transform has been employed with an atmospheric-pressure ion mobility spectrometer fitted with an electrospray ionization source. The Hadamard transform was implemented through the use of a linear-feedback shift register to produce a pseudorandom sequence of 1023 points. This pseudorandom sequence was applied to the ion gate of the spectrometer, and deconvolution of the ion signal was accomplished by the Hadamard transform to reconstruct the mobility spectrum. Ion mobility spectra were collected in both a conventional and Hadamard mode, with comparisons made between the two approaches. Initial results exhibited low spectral definition, so an oversampling technique was applied to increase the number of data points across each analyte spectral peak. The use of the Hadamard transform increases the duty cycle of the instrument to 50% and results in a roughly 5-fold enhancement of the signal-to-noise ratio with a negligible loss of instrument resolution. It is also shown that any potential multiplex disadvantage, which limits the attractiveness of some high-throughput techniques, is not a limiting factor in this new implementation.

Plasma-source mass spectrometry for speciation analysis: state-of-the-art.

Current and emerging capabilities of plasma-source mass spectrometry (PS-MS) as it is employed for elemental speciation analysis are reviewed. Fundamental concepts and their advantageous aspects, experimental conditions, and analytical performance are described and illustrated by recent examples from the literature. Novel instrumentation, techniques, and strategies for inductively-coupled plasma mass spectrometry (ICP-MS), microwave-induced plasma (MIP) mass spectrometry, glow-discharge (GD) mass spectrometry, and electrospray ionization (ESI), among others, are described. The use of ionization sources that provide tunable ionization, others that can be modulated between different sets of operating conditions, and others used in parallel is also examined.

The collagen binding alpha1beta1 integrin VLA-1 regulates CD8 T cell-mediated immune protection against heterologous influenza infection.

A common feature of many infections is that many pathogen-specific memory T cells become established in diverse nonlymphoid tissues. A mechanism that promotes the retention and survival of the memory T cells in diverse tissues has not been described. Our studies show that the collagen binding alpha1beta1 integrin, VLA-1, is expressed by the majority of influenza-specific CD8 T cells recovered from nonlymphoid tissues during both the acute and memory phases of the response. Antibody treatment or genetic deficiency of VLA-1 decreased virus-specific CTL in the lung and other nonlymphoid tissues, and increased them in the spleen. In spite of the increase in the spleen, secondary heterosubtypic immunity against flu was compromised. This suggests that VLA-1 is responsible for retaining protective memory CD8 T cells in the lung and other tissues via attachment to the extracellular matrix.
