[Analysis and characterization of fingerprint infrared spectra for ecology-planted Coptis chinensis].

The fingerprint features of infrared spectra for Coptis chinensis in different shading conditions (shelf, firry wood, corn wood) and with different ages (one year to five years) were studied. According to the ratio of characteristic peaks, the rules of the influence of shadings and ages on Coptis chinensis growth were illustrated, which confirms the feasibility of ecological planting technique for Coptis chinensis. Fourier transform infrared (FTIR) spectroscopy was used to analyze the influence of growing conditions on the growth of Coptis chinensis. The shadings and ages have no obvious influence on the quality of Coptis chinensis root, which is consistent with the results of HPLC. The quality of Coptis chinensis with ecological planting is identical with those in shelves. Infrared technique provides a meaningful guide to standardized planting of medicinal plants.

[Study on the identification of standard and false Gancao by Fourier transform infrared spectroscopy].

Standard Gancao (Glycyrrhiza uralensis Fisch) and false Ciguogancao (Glycyrrhiza pallidiflata Batal) were identified fast, nondestructively by Fourier transform infrared spectroscopy (FTIR) combined with derivative spectra and two-dimensional correlation spectroscopy (2D) in the present article. The result shows that although the two kinds of Gancao belong to one genus, there are some certain differences in their chemical components that are reflected in the IR spectra, but with some similarity and dissimilarity in the IR spectra. The two kinds of Gancao are quite different from each other in second derivative spectra and 2D spectra. Based on the differences reflected in the IR and 2D IR, the standard gancao can be identified from the false easily and clearly. The result also proved that there is a relationship between the IR spectra and the chemical components of the herbs. This method is fast, accurate and nondestructive, and the wastage of sample is less. The fast, accurate property of 2D spectroscopy makes it a powerful and new approach to evaluating medicinal herbs impersonally.

[Analysis and discrimination of Collocalia esculenta L. via FTIR spectroscopy].

Collocalia esculenta L. is called Yanwo in China. Thirty kinds of dry yanwo were identified by Fourier transform infrared spectroscopy (FTIR) . The results indicated that there were some obvious differences in FTIR between the dry yanwo and the natural yanwo, mainly represented in some absorption peaks of protein, amino acid (1 634, 1 535 cm(-1)) and amylase (1 034 cm9-1)). Meanwhile, the shape, position and intensity of the peaks from methylene (2 935 cm(-1)), protein and amino acid (1 647, 1 533, 1 447 cm(-1)) in dry yanwo were similar to those of the glutin. Therefore, different amounts of glutin were added in dry yanwo samples. The content of candy in some dry candy yanwo products was as high as 99%, but the content of yanwo was only 1%. The characteristic peaks of yanwo in these products even could not be found. So the quality of yanwo in the market could not be guaranteed from these results. FTIR is fast and direct to analyse and evaluate the quality of yanwo product.

[Quality analysis and evaluation of Rhizoma Coptidis under different cultivation conditions].

AIM: To develop methods for the fingerprint analysis of Rhizoma Coptidis and the determination of berberine, palmatine and jatrorrhizine in Rhizoma Coptidis, and analyze the contents of these three alkaloids in Rhizoma Coptidis under different cultivation conditions, from different areas and processed with different methods. METHODS: Two methods (HPLC-UV and HPLC-MS) have been developed and used in fingerprint analysis of Rhizoma Coptidis. An HPLC method was used to determine the contents of three alkaloids. RESULTS: With HPLC-MS techniques, seven major chromatographic peaks in the fingerprint analysis of Rhizoma Coptidis were identified by their MS spectra and compared with the reference standards. In different cultivation conditions, shading conditions and growing ages have obvious influence on the contents of three alkaloids in Rhizoma Coptidis, while planting density was not the major factor that influenced the contents of three alkaloids. The contents of three alkaloids of Coptidis samples were almost higher than those of Coptidis reference material. For Coptidis samples from different cultivation area, the contents of these three alkaloids were different greatly. For Coptidis samples processed with different methods, the contents of three alkaloids were not influenced obviously by processing methods. CONCLUSION: The results showed that the ecology cultivation method to replace the traditional shading method was feasible and provided the theoretical foundation for scientifically processing Rhizoma Coptidis.

Study of traditional Chinese animal drugs using FT-IR and 2D-IR correlation spectroscopy.

Using FT-IR and 2D-IR correlation spectroscopy, the intensity changes and their sequence of amide I and II bands of four traditional Chinese animal drugs (Cornu Cervi Pantotrichum, Cornu Saigae Tataricae, Scorpio and Hirud) under thermal perturbation are studied, and component of Ca(3)(PO(4))(2) in the drug of Cornu Cervi Pantotrichum and sulfates components in the drug of scorpion are identified. The drug of Cornu Cervi Pantotrichum contains inorganic salt Ca(3)(PO(4))(2) and the drug of Scorpio contains sulfates. It is assigned that the bands of 604 and 561 cm(-1) belong to the component of Ca(3)(PO(4))(2), and the bands of 637 and 615 cm(-1) belong to sulfates. Organic components of these drugs respond to the thermal perturbation far stronger than that of the inorganic components. The intensities of amide I and II bands in the drugs, except for amide II band in drug Scorpio, change strongly. For the drugs of Cornu Cervi Pantotrichum, Cornu Saigae Tataricae and Hirudo, the intensity changes of amide II band occurs prior to that of amide I band. The C-N bond in the product operator(3)(4) conjugative system converts from double bond to single bond, but the C=O bond remains double bond during the heating process. On the other hand, amide II vibrations, which may involve much more of the hydrogen bonded local structures of amide groups in the system compared to the C=O dominated amide I vibration, may undergo thermally induced changes at a temperature much lower than the other mode. The traditional Chinese animal drugs can be identified rapidly and non-separately by using FT-IR and 2D-IR correlation spectroscopy.

Research on processing medicinal herbs with multi-steps infrared macro-fingerprint method.

How to apply rapid and effective method to research medicinal herbs, the representative of complicated mixture system, is the current study focus for analysts. The functions of non-processed and processed medicinal herbs are greatly different, so controlling the processing procedure is highly important for guarantee of the curative effect. Almost, the conventional criteria of processing are based on personal sensory experience. There is no scientific and impersonal benchmark. In this article, we take Rehmannia for example, conducting a systematic study on the process of braising Rehmannia with yellow wine by using the multi-steps infrared (IR) macro-fingerprint method. The method combines three steps: conventional Fourier transform infrared spectroscopy (FT-IR), second derivative spectroscopy, and two-dimensional infrared (2D-IR) correlation spectroscopy. Based on the changes in different types of IR spectra during the process, we can infer the optimal end-point of processing Rehmannia and the main transformations during the process. The result provides a scientific explanation to the traditional sensory experience based recipe: the end-point product is "dark as night and sweet as malt sugar". In conclusion, the multi-steps IR macro-fingerprint method, which is rapid and reasonable, can play an important role in controlling the processing of medicinal herbs.

[Monitoring all-trans-retinoic acid-induced differentiation of HL-60 cells by Fourier-transform infrared spectroscopy].

This study reports a new in vitro analytical method, based on Fourier-transform infrared (FTIR) spectroscopy, tomonitor themyeloid differentiation process in human myeloblast leukemia HL-60 cells induced by all-trans-retinoic acid (ATRA). The alteration of characteristic bands was identified in the differentiated cells, arising from proteins, lipids, carbohydrates and nucleic acids. Besides the changes in lipid content and plasma membrane fluidity, the most striking changes were observed in the region of nucleic acids and carbohydrates. The authors speculate that the glycosylation and phosphorylation of proteins and the hydrogen-bonding of nucleic acids were involved in differentiation. The spectral parameters were correlated with the differentiation index, as determined by NBT reduction assay. These results suggest that FTIR spectroscopy can be used to monitor the differentiation process of HL-60 cells.

[Quality analysis of powdered milk via FTIR spectroscopy].

Eleven kinds of powdered milk were identified by Fourier transform infrared spectroscopy (FTIR). The main nutritious components had distinct fingerprint characteristics in the IR spectra of milk powder. For samples from the same factory but of different brands, there was some dissimilarities in the relative intensities of infrared absorption peaks due to the difference in fat, protein and carbohydrate as well. For the samples of the same sort, but from different factories, there were certain similarities in the IR spectra. The spectra of milk powders with high level of fat, protein, and carbohydrates have their own characteristic absorption prak in the FTIR. Three kinds of commonly used excipients: maltodextrin, sucrose and lactose, all have specific fingerprint features, which change as their amounts alter. The present research provides a fast and direct method for evaluating the quality of powdered milk.

[Analysis and assessment of Coptis chinensis for different parts, ages, and heights using Fourier transform infrared spectroscopy].

OBJECTIVE: To study the infrared (IR) fingerprint spectra of Coptis chinensis for different parts, ages, and heights, and to analyze the integrate rules about the content of berberine component in Coptis chinensis for different parts, ages, and heights. METHOD: The Fourier transform infrared (FTIR) spectroscopy was applied to detect the infrared spectra of Coptis chinensis samples rapidly and non-separately. RESULTS: The IR spectra of the root, leafstalk, and fibre of Coptis chinesis have their unique features. The content of berberine component in Coptis chinensis was different for different parts and the sequence: root > leafstalk > fibre. When Coptis chinensis grows, the content of berberine component in leafstalk also increases. The content of berberine component in leafstalk which planting in 1 200 m was less than that in 1 300, 1 400, and 1 500 m. The ages and heights provide no obvious influences on the content of berberine in the root of Coptis chinensis. CONCLUSION: Using FTIR spectroscopy, the quality of Coptis chinensis can be controlled, which provides a useful method for the standardized planting of Coptis chinensis.

[Analysis of contents of berberine in Coptis chinensis of Lichuan].

OBJECTIVE: To determine the contents of berberine in Coptis chinensis of Lichuan and establish its best cultivation scheme. METHOD: We used high performance liquid chromatography (HPLC) to measure the contents of berberine at different altitude, growth age, and leaves. RESULTS: Analytic data showed that the growth age and firry woods shading did not affect the contents of berberine in rhizome of Coptis chinensis. Low altitude was more suitable for Coptis chinensis to synthesize berberine. The contents of berberine in rhizome of Coptis chinensis with floral leaf were higher than those with lusterless and lustrous leaves, but no significant difference. CONCLUSION: The synthesis of berberine is closely correlated with shading conditions. It is recommended that the shading ratio should be reduced or the sheds removed in the middle growth age (2-3 years), and then the herb should be reshaped at the last year to enhance the synthesis of berberine, so as to obtain high-quality Coptis chinensis in the harvest.

Discrimination of Fritillary according to geographical origin with Fourier transform infrared spectroscopy and two-dimensional correlation IR spectroscopy.

Fritillaria is a traditional Chinese herbal medicine for eliminating phlegm and relieving a cough with a long history in China and some other Asian countries. The objective of this study is to develop a nondestructive and accurate method to discriminate Fritillaria of different geographical origins, which is a troublesome work by existing analytical methods. We conducted a systematic study on five kinds of Fritillaria by Fourier transform infrared spectroscopy, second derivative infrared spectroscopy, and two-dimensional (2D) correlation infrared spectroscopy under thermal perturbation. Because Fritillaria consist of a large amount of starch, the conventional IR spectra of different Fritillaria only have very limited spectral feature differences. Based on these differences, we can separate different Fritillaria to a limited extent, but this method was deemed not very practical. The second derivative IR spectra of Fritillaria could enhance spectrum resolution, amplify the differences between the IR spectra of different Fritillaria, and provide some dissimilarity in their starch content, when compared with the spectrum of pure starch. Finally, we applied thermal perturbation to Fritillaria and analyzed the resulting spectra by the 2D correlation method to distinguish different Fritillaria easily and clearly. The distinction of very similar Fritillaria was possible because the spectral resolution was greatly enhanced by the 2D correlation spectroscopy. In addition, with the dynamic information of molecular structure provided by 2D correlation IR spectra, we studied the differences in the stability of active components of Fritillaria. The differences embodied mainly on the intensity ratio of the auto-peak at 985 cm(-1) and other auto-peaks. The 2D correlation IR spectroscopy (2D IR) of Fritillaria can be a new and powerful method to discriminate Fritillaria.

Real-time monitoring of Bacillus subtilis endospore components by attenuated total reflection Fourier-transform infrared spectroscopy during germination.

Chemical changes of particular Bacillus subtilis spore components were monitored by attenuated total reflection Fourier-transform infrared spectroscopy (ATR/FTIR) during spore germination on a ZnSe internal reflection element. Within minutes of the initiation of spore germination, significant changes in the amount of calcium dipicolinate (DPA-Ca) and proteins were noted in the wild-type strain. The changes in a germination mutant (strain 1G9, gerD) were similar to those in the wild-type strain, but the rates of change were slower. The changes in another germination mutant (strain 1G7, gerA) were very different from those in the first two strains: germination was slow and incomplete, and proteins and DPA-Ca remained unaltered throughout the course of the germination study. This technique thus offers a sensitive and non-destructive method for real-time monitoring of various cellular components during spore germination.
