A study on the traditional Chinese medicine Jinyebaidu for prevention and treatment of intrauterine infection with guinea pigs cytomegalovirus.

The purpose is to study the prophylactic and therapeutic effect of the traditional Chinese Medicine (TCM)-Jinyebaidu (JYBD) to guinea pig cytomegalovirus (GPCMV) intrauterine infection. The virus-free female and male guinea pigs were screened with nest-polymerase chain reaction (N-PCR). After inbred, pregnant guinea pigs were selected and divided into 3 groups randomly: 5 guniea pigs of the blank control group were not given either GPCMV or JYBD. 31 guniea pigs of the positive control group were inoculated 1 mL (10(7) TCID50) suspension of GPCMV intraperitoneal. 10 guniea pigs of the experimental group were inoculated GPCMV firstly and then perfused stomach with JYBD for 14 days (Dosage in accordance with the modulus of the weight ratio of human to guniea pig). The effects of JYBD on the intrauterine infection of GPCMV were observed. The results showed that JYBD could decrease the maternal infection rate from 100% (31/31) to 50% (5/10) (P < 0.001), the intrauterine infection rate from 100% (72/72) to 75% (21/28) (P < 0.001), and the rate of abnormal outcome of pregnancy from 64.4% (29/45) to 25.0% (7/28) (P < 0.001), the infective symptoms being relieved. It can be concluded that traditional Chinese medicine- JYBD can prevent and treat (GPCMV intrauterine infection, and can be expected a prophylactic drug for HCMV intrauterine infection.

Evaluation on clinical application of three testing methods for human cytomegalovirus infection in pregnancy.

The value of ELISA, N-PCR and RT-PCR in clinical practice for pregnant women with HCMV infection was investigated. 5581 pregnant women were screened by ELISA. Among them, 100 cases were positive for IgM (group 1), 69 for both IgM and serous DNA (group 2) and 69 for both IgM and mRNA (group 3). The infectious status, maternal-fetal transmission and pregnancy outcome were monitored. It was demonstrated that the accordance rate of group 3 and group 2 with group 1 was 56.25% and 43.75%, respectively. The maternal-fetal transmission rate in the group 1, 2 and 3 was 19.00%, 40.58% and 46.15%, respectively, with a significant difference found between group 2, 3 and group 1 (P < 0.01). Incidence of spontaneous abortion, fetal death, fetal abnormality and neonatal death in group 1, 2 and 3 was 10.00%, 15.94% and 30.77%, respectively, and that of group 3, 2 was 4 and 2 times as much as that of group 1, respectively (OR = 4.00, P < 0.001; OR = 2.343, P < 005, respectively). It was concluded that HCMV-IgM(+) can only be considered as an screening indicator for pregnant women with HCMV infection, while IgM(+) combined with serous DNA(+) or mRNA(+) indicates active infection and has a high incidence of maternal-fetal transmission and abnormal pregnancy outcome.

[Effects of combined pollution of heavy metals on root vitality of wheat seeds].

The combined effects of Cu, Cd, Pb and Zn on the growth and activity of the seminal roots of wheat (Triticum aestivum) were studied by the method of quadratic regression revolution design. The mathematical models describing the above relations were established. As for the toxicity to the root vitality, their single effects, marginal utility and combined effects between two heavy metals were analyzed. The results showed that Pb had the most important effect on the activity of root, and then, Cu, Cd and Zn in sequence. The interaction between Cu and Cd, Cu and Zn, Zn and Cd, Cd and Pb had great effects on the activity of seminal roots. Furthermore, among the concentration range of 0 to 20 mg.L-1, as for Cu and Zn, the lowest density had the biggest marginal utility, while Cd and Pb had the biggest ones at the highest density.

[Effects of different arbuscular mycorrhizal fungi on Tagetes erecta growth and diesel degradation].

The effects of G. mosseae, G. geospora, G. constrictum and bacteria on diesel tolerance of Tagetes erecta were investigated under greenhouse conditions. The results showed that AM fungi could still develop mycorrhizal assosiations with mum when the diesel concentration was 5,000 mg.kg-1. White mum was better than yellow mum in diesel tolerance, with 63.1% total biomass increased. The colonization rate of inoculating AM fungi treatment was 3.5%-29.9% higher than the control. G. mosseae and G. geospora were better strains, their biomass increasing 9.0% and 42.7% than the control, respectively, while the effect of inoculating mixed AM fungi was not obvious. Bacteria inhibited the colonization of arbuscular mycorrhizal fungi on white mum, but promoted the vegetative and reproductive growth of mycorrhizal mum. Among 5 inoculation treatments, treatments of inoculating G. geospora and inoculating mixed AM fungi and bacteria were better, with 16.51% and 14.05% more diesel degradation rate than that of the control, respectively.

[Interaction between microorganisms and heavy metals and its application].

Interactions between microorganisms and heavy metals were expatriated in many ways. Because of their resistance and detoxification to heavy metals, microorganisms are able to leach, absorb and transform heavy metals, which has been actively applied to extract heavy metals from low-rate ore in mine exploitation. Although heavy metals are toxic to microorganisms population and harmful to its processes, some special microorganisms have been applied to the treatment of industrial waste materials and the remediation of soils polluted by heavy metals. The application of microbiological biomass and activity to evaluating the pollution situation and ecological risk of heavy metals in environment media is also significant.

[Clinical value of antenatal diagnosis of intrauterine active human cytomegalovirus infection with reverse transcription polymerase chain reaction].

OBJECTIVE: To study the clinical value of antenatal diagnosis of intrauterine active human cytomegalovirus (HCMV) infection with reverse transcription polymerase chain reaction (RT-PCR). METHODS: The active HCMV infection of pregnant women was diagnosed with enzyme-linked immunosorbent assay (ELISA), serum Nest-PCR (N-PCR) and RT-PCR. The methods of antenatal diagnosis include villus sampling, amniocentesis and cordocentesis under ultrasound guidance. RT-PCR and N-PCR was used to screen fetal active infection. RESULTS: The infection rate of chorionic villi and cord blood whose mother was in active infection is 53.3% (16/30) and 47.8% (33/69) respectively, whereas that whose mother just had infectious history is 8.3% (4/48), 9.7% (6/62) respectively, the differences are statistically significant respectively (P < 0.001). The active infection rate of chorionic villi and cord blood in maternal active infection group is 30.0% (9/30) and 29.0% (20/69) respectively, whereas that with infectious history is 4.2% (2/48) and 4.8% (3/62), the differences are statistically significant respectively (P < 0.001). In the active intrauterine infection, the incidence of abortion and premature delivery was 41.2% (14/34), significantly higher than that of the latent infection 8.0% (2/25, P < 0.01). CONCLUSIONS: Antenatal diagnosis of intrauterine active HCMV infection with RT-PCR and the means of sampling described are of safety and reliability. It has a great clinical value in management of the fetus whose mother had been in active HCMV infection.
