In vitro study of magnesium-calcite biomineralization in the skeletal materials of the seastar Pisaster giganteus.

The mechanisms of formation of biogenic magnesium-rich calcite remain an enigma. Here we present ultrastructural and compositional details of ossicles from the seastar Pisaster giganteus (Echinodermata, Asteroidea). Powder X-ray diffraction, infrared spectroscopy and elemental analyses confirm that the ossicles are composed of magnesium-rich calcite, whilst also containing about 0.01 % (w/w) of soluble organic matrix (SOM) as an intracrystalline component. Amino acid analysis and N-terminal sequencing revealed that this mixture of intracrystalline macromolecules consists predominantly of glycine-rich polypeptides. In vitro calcium carbonate precipitation experiments indicate that the SOM accelerates the conversion of amorphous calcium carbonate (ACC) into its final crystalline product. From this observation and from the discovery of ACC in other closely related taxa, it is suggested that substitution of magnesium into the calcite lattice through a transient precursor phase may be a universal phenomenon prevalent across the phylum echinodermata.

Photophysical properties of hydroxylated amphiphilic poly(p-phenylene)s.

A homologous series of polyhydroxylated poly(p-phenylene)s with different alkoxy groups (C6PPPOH, C12PPPOH, and C18PPPOH) were synthesized with use of the Suzuki polycondensation reaction. Comparative studies of the structure correlation between their photophysical properties and film morphology is described. The absorption and emission spectra of polymers in solution and thin films showed similar features indicating that the electronic properties in solution were retained in the film state. Compared to the polymer with the short alkoxy chains (C6PPPOH), the polymers with long alkoxy groups (C12PPPOH and C18PPPOH) showed improved film forming properties with continuous and smooth film morphology. The absorption properties of the C12PPPOH showed an enhanced effective conjugation length and high quantum yield implying planarization of the backbone through alkoxy chain packing (C12H25O-) and potential hydrogen bonds. No overlap in the absorption and emission spectra was observed, which indicated minimized excimer formation or excitation energy transfer in the films. Time-resolved fluorescence measurements showed that the decay times increased from 43 ps (C6PPPOH) to 78 ps (C12PPPOH) and 99 ps (C18PPPOH). Electrochemical studies were performed for all polymers and the observed oxidation potential for C6PPPOH was higher than that of C12PPPOH and C18PPPOH. In addition, the C12PPPOH has the lowest band gap of DeltaE = 2.59 eV when compared to the 3.1 (C6PPPOH) and 2.61 eV (C18PPPOH) gaps. The optical band gaps estimated from the absorption onset of the polymers are significantly higher than those obtained from electrochemical data. C12PPPOH was chosen for investigating the charge carrier mobility by the time-of-flight (TOF) technique. The observed results also showed negative field dependent values of the drift mobility for the polymer C12PPPOH.

On-site polymer-coated hollow fiber membrane microextraction and gas chromatography--mass spectrometry of polychlorinated biphenyls and polybrominated diphenyl ethers.

Porous polypropylene hollow fiber membrane coated with a conjugated polymer was used as an on-site sampling device for the extraction of polychlorinated biphenyls and polybrominated biphenyl ethers from coastal sea water samples. The coated hollow fiber membrane was placed in a vial containing the sample, and the target compounds extracted via manual shaking of the vials at the site of sample collection. For each extraction, two fibers were used. After extraction, the fibers with the adsorbed analytes were brought back to the laboratory for further processing. Care was taken to preserve the integrity of the analytes and to avoid contamination during transport; after extraction, the fibers were carefully removed and placed in air-tight crimper vials which were stored in an ice-box. The analytes were desorbed by solvent in the laboratory and analyses were carried out using gas chromatography/mass spectrometry. This method was highly reproducible with relative standard deviations in the range of 1-9%. Recoveries from spiked water samples ranged from 83% to 98%. Low limits of detections between 0.04 and 0.21ngl(-1) were achieved. The extraction efficiency was compared with solid-phase microextraction.

Fabrication and characterization of multilayer films from amphiphilic poly(p-phenylene)s.

Over the past two decades, considerable efforts have been devoted to the development of conjugated polymeric materials for electronic applications due to the tunability of their properties through variation of their chemical structure. The LB technique is one of the most effective and precise methods for controlling the organization and thereby the properties of polymer films at the nanoscale for device fabrication. A detailed study was performed on the Langmuir-Schaefer (LS) monolayer and Langmuir-Blodgett-Kuhn (LBK) multilayer formation of newly designed conjugated poly(p-phenylene)s (C(n)PPPOH), incorporated with alkoxy groups with different chain lengths (C(6)H(13)O-, C(12)H(25)O-, and C(18)H(37)O-) and hydroxyl groups on the polymer backbone. The monolayer formed at the air-water interface was characterized using surface pressure-area isotherms, including hysteresis measurements. The films were then transferred to different hydrophilic solid substrates and analyzed using surface plasmon resonance spectroscopy, UV-vis spectroscopy, fluorescence spectroscopy, and AFM measurements. The results showed that the polymer with a short alkoxy chain (C(6)PPPOH) forms uniform monolayers at the air-water interface and can be transferred as multilayer films compared to C(12)PPPOH and C(18)PPPOH. The observed film thicknesses measured by SPR and AFM studies were similar to the theoretical value obtained in the case of C(6)PPPOH, whereas this was not the case with the other two polymers. The present study shows that the polymer C(n)PPPOH with short alkoxy chain can be transferred onto different solid substrates for device fabrication with molecular level control.

Size selective assembly of colloidal particles on a template by directed self-assembly technique.

We report a simple and effective approach to organize micron- and submicron-sized particles in a size selective manner. This approach utilizes the template assisted directed self-assembly technique. A topographically patterned photoresist surface is fabricated and used to create an ordered array of colloidal particles from their aqueous suspensions. Assembly of particles on this template is then achieved by using a conventional spin coating technique. Feasibility of this technique to form a large area of patterned particle assemblies has been investigated. To arrange the particles on the template, the physical confinement offered by the surface topography must overcome a joint effect of centrifugal force and the hydrophobic nature of the photoresist surface. This concept has been extended to the size selective sorting of colloidal particles. The capability of this technique for sorting and organizing colloidal particles of a particular diameter from a mixture of microspheres is demonstrated.

Polymer-coated hollow fiber microextraction combined with on-column stacking in capillary electrophoresis.

In this work, a novel microextraction method termed polymer-coated hollow fiber microextraction (PC-HFME) was developed in combination with capillary electrophoresis (CE). Polar dihydroxylated polymethylmethacrylate polymer was coated onto a porous propylene hollow fiber membrane and used as an adsorbent and that was placed in a stirred aqueous sample solution. Tumbling of the extraction device within the sample solution facilitated extraction. The amino alcohols (2-amino-1-phenylethanol, norephedrine, alprenolol and atenolol which are beta-blocker drugs), were used as model compounds to investigate the extraction performance. No organic solvent was used in this procedure. The extract was then further concentrated through on-column stacking (normal stacking mode) during CE analysis. The detection limits ranged from 0.9 to 7 ng ml(-1). Relative standard deviations (n=6) ranged from 4 to 6%. The extraction of the amino alcohols in spiked wastewater effluent (representing a complex matrix) was evaluated using the developed procedure.

Electrochemically nanopatterned conducting coronas of a conjugated polymer precursor: SPM parameters and polymer composition.

Here we describe the formation of precisely controlled corona-type nanopatterns on electroactive polymer precursor films using scanning probe microscopy (SPM) methods. The binary composition of electroactive groups in the polymer triggers the formation of corona-type nanopatterns at particular voltages and tip writing speeds through the electrooxidation of the polymer precursor film. Various parameters such as tip speed and applied bias were explored in the nanopatterning process, and the formation of a conductive nanopattern was investigated using conducting atomic force microscopy (C-AFM). The formation of the nanopattern was attributed to the flow of electrons from the AFM tip to the polymer film in a controlled electric field distribution. We also report a new method to distinguish the polymer composition and distribution of a polymer blend film by characterizing biasing differences in the patterning of a polymer film.

Direct electrochemical nanopatterning of polycarbazole monomer and precursor polymer films: ambient formation of thermally stable conducting nanopatterns.

The direct nanopatterning of polycarbazole on ultrathin films of a "precursor polymer" and monomer under ambient conditions is reported. In contrast to previous reports on electrochemical dip-pen nanolithography using monomer ink or electrolyte-saturated films in electrostatic nanolithography, these features were directly patterned on spin-cast films of carbazole monomer and poly(vinylcarbazole) (PVK) under room temperature and humidity conditions. Using a voltage-biased atomic force microscope (AFM) tip, electric-field-induced polymerization and cross-linking occurred with nanopatterning in these films. Different parameters, including writing speed and bias voltages, were studied to demonstrate line width and patterning geometry control. The conducting property (current-voltage (I-V) curves) of these nanopatterns was also investigated using a conducting-AFM (C-AFM) setup, and the thermal stability of the patterns was evaluated by annealing the polymer/monomer film above the glass transition (T(g)) temperature of the precursor polymer. To the best of our knowledge, this is the first report in which thermally stable conducting nanopatterns were drawn directly on monomer or polymer film substrates using an electrochemical nanolithography technique under ambient conditions.

Formation of transient amorphous calcium carbonate precursor in quail eggshell mineralization: an in vitro study.

To understand the mechanism of quail eggshell biomineralization, we have performed two CaCO(3) precipitation experiments. In the reprecipitation experiments, supersaturated Ca(HCO(3))(2) was prepared by bubbling CO(2) through a slurry of biogenic CaCO(3) obtained from bleach-treated eggshell followed by filtration to obtain a clear solution for crystallization experiments. The nucleated crystals were collected at various time intervals and analyzed. In the second experiment, the extracted SOM from the bleach-treated eggshell was added to the supersaturated clear solution of Ca(HCO(3))(2) solution obtained by bubbling CO(2) gas through a slurry of synthetic CaCO(3) followed by filtration. The crystals/precipitates collected at various time intervals were analyzed. Both experiments showed that amorphous CaCO(3) (ACC) was precipitated in the early stages, which then transformed to the most stable crystalline calcite phase. Amino acid analysis of the soluble organic matrixes (SOM) indicated the presence of high amounts of Glx and Asx amino acids. Ovomucoid--an acidic glycoprotein, and lysozyme--a basic protein, are the two major components along with a few low molecular weight peptides present in the SOM of quail eggshell matrix. Both ovomucoid and lysozyme did not induce precipitation of the ACC phase in in vitro conditions, while the fraction containing low molecular weight peptides induced the precipitation of ACC, suggesting that the latter play an important role in the eggshell biomineralization. Thus, organisms can produce inorganic minerals which assume nonequilibrium morphologies and intricate architecture by precipitating transient ACC, which then transformed into the crystalline phase. Altogether, these observations further demonstrate that this strategy may be common in both vertebrate and invertebrate mineralized structures.

Intramolecular hydrogen-bond-assisted planarization of asymmetrically functionalized alternating phenylene-pyridinylene copolymers.

We report on the synthesis and characterization of a series of asymmetrically functionalized amphiphilic polymers with alternating pi-donor units (e.g., substituted benzene) and pi-acceptor units (e.g., pyridine) along the polymer backbone. The purpose of our present work involves incorporation of functional groups along the main chain to form intrachain hydrogen bonds, which promote planarization of the polymer backbone, and to fine-tune the optical properties. The structure-property relationship of polymers P1-P6 was investigated by means of analytical methods, such as FTIR spectroscopy, 1H and 13C NMR spectroscopy, UV/Vis spectroscopy, fluorescence spectroscopy, gel permeation chromatography, thermogravimetric analysis, cyclic voltammetry, and X-ray powder diffraction. All polymers were soluble in common organic solvents, and the optical and fluorescence spectra of the polymers showed significant changes according to the formation (P4, P5) or absence (P6) of intramolecular hydrogen bonding along the polymer backbone. Moreover, the 2,6- or 3,5-linkage of the pyridine rings in P5 and P6, respectively, reduced the conjugation along the polymer backbone and this is reflected in their optical properties. The optical properties of the polymers were influenced by the addition of acid (P1-P6), base (P4-P6), and metal ions (e.g., Cu2+, Fe3+, Ag+, Ni2+, Pd2+, Mn2+, Zn2+, Mg2+, and Pr3+). Such polymers could be used in various applications, including sensors and stimuli-responsive displays.

Polymer-coated hollow-fiber microextraction of estrogens in water samples with analysis by gas chromatography-mass spectrometry.

A novel sorbent, dihydroxylated polymethylmethacrylate (DHPMM), coated on hollow-fiber membrane, is used for the polymer-coated hollow-fiber microextraction of trace amounts of natural and synthetic estrogens, such as diethylstilbestrol, estrone, 17beta-estradiol and 17alpha-ethynylestradiol, in aqueous samples. In this procedure, estrogens were extracted using the functionalized polar DHPMM polymer with derivatization using N-methyl-N-(trimethylsilyl)trifluoroacetamide followed by gas chromatography-mass spectrometric analysis. The detection limits for estrogens in aqueous sample were between 0.03 and 0.8 ng l(-1) and the calibration curves were linear over the concentration range 0.05-10 microgl(-1) and had correlation coefficients of >0.994. The relative standard deviations (RSDs) were <15% (n = 3). This simple, accurate, sensitive and selective analytical method is applicable to the determination of trace amounts of estrogens in reservoir and potable water samples.

Sol-gel-coated oligomers as novel stationary phases for solid-phase microextraction.

Amphiphilic and hydrophilic oligomers were synthesized and coated on fused silica capillaries using a sol-gel technique. Sol-gel-coated capillaries were evaluated for the solid-phase microextraction and preconcentration of a wide variety of non-polar and polar analytes. Both types of coatings were stable under high temperature (up to 280 degrees C). The extraction efficiency of the sol-gel coatings was evaluated for the extraction of both non-polar and polar analytes, including organochlorine pesticides, triazine herbicides, estrogens and alkylphenols (APs) and bisphenol-A (BPA). Compared with commercially available solid-phase microextraction (SPME) adsorbents such as poly(dimethylsiloxane)divenylbenzene and polyacrylate, the new materials showed comparable selectivity and sensitivity towards both non-polar and polar analytes. The new coatings gave good linearity and detection limits. For example with triazines, a detection limit of <0.005 microl l(-1), precision from 5.0 to 11.0% (n = 6) and linearity of the calibration plots (0.5 to 50 microl l(-1)) were obtained. The sol-gel coated SPME capillaries were used for the determination of triazine herbicides in reservoir water samples collected in Singapore.

Determination of alkylphenols and bisphenol-A. A comparative investigation of functional polymer-coated membrane microextraction and solid-phase microextraction techniques.

A functional polymer (hydroxylated polymethacrylate) coated on porous polysulfone hollow fiber membrane (PS-HFM) was used as an adsorbent for the extraction of alkylphenols and bisphenol-A from seawater samples. Analyses of the extracts were performed using gas chromatography-mass spectrometry (GC-MS) after injection-port derivatization using bis(trimethylsilyl)trifluoroacetamide (BSTFA). We term the procedure as polymer-coated hollow fiber microextraction (PC-HFME). Owing to high porosity PS-HFM coated with hydroxylated polymer showed high extraction efficiency. Compared with solid-phase microextraction (SPME), PC-HFME showed good selectivity and sensitivity. Detection limits of alkylphenols and bisphenol-A ranged between 0.07 and 2.34 ng l(-1). The linearity range was from 0.01 to 15 microg l(-1) and the correlation coefficient (r) up to 0.997. The sensitivity and selectivity of the coated HFM could be potentially tuned by changing the characteristics of the coated hydroxylated polymer. The PC-HFME procedure was applied to the detection of alkylphenols and bisphenol-A in the coastal waters of Singapore.

Developing macroporous bicontinuous materials as scaffolds for tissue engineering.

Calcareous skeletal elements (ossicles) isolated from the seastar, Pisaster giganteus, were characterized and tested as potential biocompatible substrates for cellular attachment. These ossicles have a remarkably robust open-framework architecture with an interconnected network of ca. 10 microm diameter pores. Scanning electron and confocal microscopy was used to characterize the cell-substrate interaction. Cell culturing experiments revealed that the cells firmly attach to the ossicle surface, forming cell aggregates of several layers thick. The anchored cells extended to form 'bridges' between the openings in the bicontinuous framework and the degree of coverage increased as culture time progressed. Osteoblasts grown on the ossicles were found to be viable up to 32 days after initial seeding, as proven by assaying with AlamarBlue and FDA/PI staining indicating the ossicle's potential as an alternative highly effective tissue scaffold. Given the limitation in availability of this natural material, the results presented here should be seen as offering guidelines for future development of synthetic materials with physical and chemical properties strongly conducive to bone repair and restoration.

Effect of heterocyclic based organoclays on the properties of polyimide-clay nanocomposites.

Polyimide-clay nanocomposites were prepared from their precursor, namely, polyamic acid, by the solution-casting method. Organomodified montmorillonite (MMT) clay was prepared by treating Na+MMT (Kunipia F) with three different intercalating agents, namely, piperazine dihydrochloride, 1,3-bis(4-piperidinylpropane) dihydrochloride and 4,4'-bipiperidine dihydrochloride at 80 degrees C. Polyamic acid solutions containing various weight percentages of organomodified MMT were prepared by reacting 4,4'-(1,1'-biphenyl-4,4'-diyldioxy)dianiline with bicyclo[2.2.2]oct-7-ene-2,3,5,6-tetracarboxylic dianhydride in N-methyl-2-pyrrolidinone containing dispersed particles of organomodified MMT at 20 degrees C. Nanocomposite films were prepared from these solutions by solution casting and heated subsequently at a programmed heating rate. These films were transparent and brown in color. The extent of layer separation in nanocomposite films depends upon the chemical structure of the organoclay. These films were characterized by inherent viscosity, FT-IR, DSC, TMA, WAXD, TEM, UV, and TGA. The tensile behavior and surface energy studies were also investigated. The nanocomposite films had superior tensile properties, thermal behavior, and solvent resistance. Among the three organoclays, piperazine dihydrochloride was the best modifier.

Development and application of a simple capillary-microreactor for oxidation of glucose with a porous gold catalyst.

An efficient oxidation of glucose to gluconic acid was performed using a porous gold(0) catalyst in a low-cost microreactor designed from Pyrex glass capillary tubing; compared with the conventional synthesis procedure this novel approach of using a capillary-microreactor offers a convenient and highly efficient means to optimise reaction conditions and catalytic activities.
