Separation strategies of plant constituents--current status.

The paper summarizes the state of art of different separation methods which are used for the analysis and isolation of plant constituents. An overview about the extraction methods which are frequently used for the non-volatile constituents of plants is given. Special attention is paid to the identification possibilities of non-volatile and volatile compounds, since generally the role of identification of plant's constituents is undervalued. We would emphasize the facts that, for correct identification, the various chromatographic and spectroscopic methods have to be used in conjunction. The application of two different methods from each field is usually sufficient. For quantitative information, two independent methods are necessary and are acceptable if the results are within 3% of each other. If only one method is available for quantitative analysis, the results can only be accepted if, using the global optimum, the ratios of the components determined are identical to one decimal place with the ratios of three measurements (local optima) using different mobile phases with different selectivities. Based on our own 20-year experience and more than a hundred isolated compounds, we give an isolation strategy where the structures and properties of the compounds to be isolated do not have to be known. It is pointed out that without analytical monitoring, the results of preparative separations cannot be guaranteed.

Structure elucidation and antioxidant activity of (-)-isosilandrin isolated from Silybum marianum L.

A regioisomer of the known flavanolignan (-)-silandrin (3a), named (-)-isosilandrin (8a), was isolated from the fruits of a white-flowered variant of Silybum marianum L. populated in Hungary. Its structure was established both by spectroscopic methods and total synthesis, and its absolute configuration was determined by means of circular dichroism. This compound showed stronger inhibitory activity on the superoxide anion (O2*-) release by human polymorphonuclear leukocytes (PMNL) than (+)-silybin (1a,b).

[Comparative botanical and phytochemical investigations on the Gentiana species native to Hungary].

The aim of this work was to compare the three Gentiana species native to Hungary (Gentiana asclepiadea L., Gentiana pneumonanthe L. and Gentiana cruciata L.--all protected) based on their botanical and phytochemical properties. Furthermore we planned the exploration of the biodiversity of these species and the examination of the correlations of the variable elements observed to establish the importance of these factors. Morphological, histological and phytochemical investigations were made. A new analytical method was developed through the systematic optimization of sample preparation and chromatographic (HPLC-DAD) separation. The results of the morphological, histological and phytochemical investigations were analysed by multivariate exploratory techniques. Based on our results, the investigated species can be distinguished as they are strongly heterogenous regarding certain morphological, histological and phytochemical characteristics. We made the first study on the biodiversity of these Gentiana species in their whole habitat in Hungary. Based on the results of the investigations mentioned above, distinct interspecific variability was found, in addition some results showed significant intraspecific variability as well. Numerous correlations were revealed between the variable characteristics, for example significant relationship was found between the average gentiopicroside content of the underground parts of Gentiana cruciata on the biotopes and the slope of the biotope. The knowledge of this variability and these correlations can be a benefit during the introduction and the improvement of medicinal plants with high contents of agents.
