[A novel fungicide of the iturin group, obtained from a marine isolate of Bacillus subtilis. Isolation, physico-chemical and biochemical properties, identification].

Strain KMM 457 of Bacillus subtilis was isolated from a frozen sample of the soft coral Sarcophyton sp. The samples were collected in 1989 in the South China Sea near the Vietnam Shore during an expedition on the board of R/V "Akademik Oparin". Metabolites of the isolate were investigated and it was found to produce a number of physiologically active compounds. One of them designated as OGA showed fungicidal activity. By the physicochemical properties it was referred to the group of iturins. The comparison with the described antibiotics of the iturin group suggested that it was a new representative of the iturin group.

[Bioactive compounds from marine actinomycetes].

Studies of the origin of bioactive metabolites of marine actinomycetes are reviewed. Structures and properties of new metabolites from indigenous marine bacteria from Actinomycetales order, such as a benzanthraquinone antibiotic from a strain of the Chainia purpurogena, istamycins, aplasmomycins, altemicidin, new phenazine esters. C13-butanolide, marinone and debromomarinone, palmyromycin, urauchimicins and some others compounds are presented. Prospects of marine biotechnology and microbiology (with considerable emphasis on the development of the basis biology of marine microorganisms in cultures collection) are discussed.

[The serum and tissue concentrations of antibiotics used in pediatric pulmonology].

Measurements were made of the concentrations of different antibiotics in children with acute and chronic pneumonia. The data obtained do not only confirm the dependence of the peak concentration of the drug in blood serum and tissue fluid on the antibiotic single dose but also point to a direct relationship between the single dose and duration of a high drug concentration in the body. It has been established that parenteral drugs penetrate well the pleural cavity, making it inadvisable to administer antibiotics intrapleurally in the treatment of acute pneumonias complicated by exudative pleurisy. In chronic pneumonia, the maximal content of antibiotic in pulmonary tissue reaches 30% of its serum concentration 2.5-3 hours after a single intramuscular drug injection. Study of the pharmacokinetics of antibiotics administered per os in the treatment of acute pneumonia attests to their rapid absorption from the gastrointestinal tract; as a rule, the peak concentrations were recorded 1 hour after the drug intake and exceeded many times the minimum inhibitory concentration for pneumococci isolated from the patients placed under observation.

[The nonspecific changes in the immune system to the administration of the membrane-ribosomal fraction of the causative agent of pseudotuberculosis].

Nonspecific changes in different elements of the immune system of animals under the action of Y. pseudotuberculosis membrane-ribosomal fraction (MRF) with high protective potency have been studied for the purpose of the analysis of the preparation for immunological safety. MRF has been shown to produce no changes in humoral immune response to antigens of different nature or to enhance this response, to produce no essential effect on the intensity of the reaction of delayed hypersensitivity to sheep red blood cells and to stimulate phagocytic processes in peritoneal macrophages and polymorphonuclear leukocytes.

[Streptococcus pneumoniae serotypes in children with acute pneumonia and pleurisy].

The study of pneumococci of different serotypes, isolated from patients with acute pneumonia and pleuritis and from healthy children was carried out. Among the pneumococcal serotypes causing pneumonia and pleuritis in children serotypes 1, 6, 19, 14 and 3 were most widely spread and constituted 62.3% of all isolated pneumococci. In young children cases of acute pneumonia and pleuritis were more often induced by serotypes 6 and 14 and in older children, by serotypes 1 and 3. In patients with uncomplicated pneumonia and pleuritis differences in the detected serotypes of pneumococci were observed, and the disease course differed in severity. Serotypes 14, 3 and 3 induced destructive processes in the lungs more often than other serotypes. Monitoring of the sensitivity of pneumococci to antibiotics showed that most of the strains retained high sensitivity to penicillin and ampicillin. In most cases the detected resistant pneumococcal strains belonged to serogroup 19.

[Indices of endocrine activities of the digestive organs in patients with dysentery].

Some indices of pancreatic endocrine function were studied over time in patients with acute dysentery. The plasma levels of insulin, C-peptide, glucagon, and gastrin were determined by a radioimmunoassay. 35 patients and 30 healthy controls were examined. Clinical signs of the affection of the pancreas in the patients were undetectable. However, significant reduction of the level of C-peptide in the acute period of disease as compared to the normal level, an increase in the level of insulin during early convalescence, and the reduction of gastrin concentration during the entire period of disease were noted. These indices are indicative of disorder of pancreatic endocrine function in patients with acute dysentery.

[Effect of lipopolysaccharide of the pseudotuberculosis bacterium on the functional activity of macrophages].

Study of the effect of lipopolysaccharide (LPS) of Y. pseudotuberculosis on the culture of peritoneal macrophages of guinea pigs has established that in concentrations of 1-10 micrograms/ml LPS stimulated the absorption and digestive activity of the macrophages. In concentrations of 50-100 micrograms/ml it had a cytotoxic effect on the cell elements and inhibited their phagocytic function. The study of the LPS effect on absorption and liberation of 14C-labeled bacteria of Y. pseudotuberculosis showed that the biopolymer increased 2-3 times the uptake of the labeled antigen from the macrophages and lowered its elimination. In the experiments on mice infected with a labeled culture of S. aureus no increase in the uptake under the effect of the pseudotuberculosis LPS was observed, whereas an increase in the rate of the label elimination was stated. The difference was statistically significant. The results of the experiments are indicative of the specific effect of LPS. The study demonstrated a significant role of LPS of Y. pseudotuberculosis in the pathogenesis and protection of the host from this infection.

[Role of sterols in the membranotropic activity of triterpene glycosides].

Interaction of stichoposide A, a triterpene glycoside and trepang sterols in solution was studied. With the use of liposomes the binding of the glycoside and its effect on the ion permeability of the lipid membranes of different sterol composition were also studied. It was shown that the membrane activity of the glycoside was defined by the level of its affinity to the sterol receptor. It decreased in the following order: cholesterol, total fraction of delta 5-sterols, total fraction of delta 7-sterols, delta 7-sterol xylosides and delta 5-sterol sulfates. Possible involvement of glycosylated and sulfated forms of sterols in development of resistance in the cell membranes and tissues of trepangs to their own glycosides is discussed.

A comparative study in vitro of physiological activity of triterpene glycosides of marine invertebrates of echinoderm type.

1. The cytostatic and antimicrobial activity of triterpene glycosides of 19 holothurian species of the Pacific tropical zone has been studied. 2. It has been demonstrated that yeast and tumor cells display a comparable sensibility to the action of triterpene oligosides of sea cucumbers. 3. Gram-positive and Gram-negative bacteria are not sensible to the action of glycosides in doses to 500 mkg/ml. 4. Triterpene glycosides-stichoposides, thelothurins and oligosides of Holothuria of genus Bohadschia are the most active in relation to fungal, yeast microflora and tumor cells.

[Spectrophotometric determination of the stichoposide A from the holothurian Stichopus japonicus S].

The absorption spectrum of stichoposid A with sulfuric acid, vanillin and alkaline was studied for the purpose of possibly using the reactions with these substances for quantitative determination of stichoposid A in biological materials. The reaction with 1 N NaOH was shown to be the most useful since it is highly specific with respect to the glycoside aglycone and makes it possible to investigate stichoposid A interaction with the cells of the yeast microflora.
