Novel surface polypeptides of Campylobacter jejuni as traveller's diarrhoea vaccine candidates discovered by proteomics.

Campylobacter jejuni is one of the most common causes of traveller's diarrhoea and food poisoning, therefore development of a vaccine is important. Using biochemical fractionation and mass spectrometry analysis, we identified more than 110 surface polypeptides. Eight C. jejuni identified surface proteins were expressed in Escherichia coli and purified. Mice were immunized with different doses of these purified proteins and challenged orally with C. jejuni strains ML1 and ML53. The degree of protection of mice was tested by intestinal colonization. At least two groups of mice vaccinated with purified proteins clear the infection faster than control mice. Here, we present the use of a proteomics based approach for the identification of novel protein based C. jejuni vaccines for the first time.

Sequential MCM/P1 subcomplex assembly is required to form a heterohexamer with replication licensing activity.

Replication licensing factor (RLF) is a multiprotein complex involved in ensuring that chromosomal DNA replicates only once in a single cell cycle. It comprises two components, termed RLF-M and RLF-B. Purified RLF-M consists of a mixture of complexes containing all six members of the MCM/P1 family of minichromosome maintenance proteins. The precise composition of these different complexes and their contribution to RLF-M activity has been unclear. Here we show that in Xenopus extracts, MCM/P1 proteins mainly form heterohexamers containing each of the six proteins. This heterohexamer is readily split into subcomplexes, whose interactions and subunit composition we characterize in detail. We show for the first time an ordered multistep assembly pathway by which the heterohexamer can be reformed from the subcomplexes. Importantly, this novel pathway is essential for DNA replication, since only the full heterohexamer can bind productively to chromatin and provide RLF-M activity.

[Protein phosphorylation in the normal nervous system and in Alzheimer's disease].

Protein phosphorylation induced by neurotransmitters, hormones, different growth factors, nerve impulse is involved in the regulation of a wide variety of physiological processes in the nervous system. The data on abnormal phosphorylation/dephosphorylation processes in the brain of patients with Alzheimer's disease are considered. In AD there are a few phosphoproteins with known function, protein kinases, phosphatases that are altered. The own data on the decrease in casein kinase 2 activity, content and altered spectrum of substrate proteins in Alzheimer's disease are presented. Drastic reductions in zinc- and magnesium-stimulated protein tyrosine activities are also recorded. The important role of abnormalities in the phosphorylation/dephosphorylation system in the stimulation of pathological processes in Alzheimer's disease is noted.

[Physiological changes in pyridinoline excretion in urine depending on age and sex].

Natural physiological alterations in excretion of pyridoline (total hydroxypyridoline and lysyl pyridoline) with urine were studied in children and adults depending on age (1-86 years old) and sex. Age-dependent alterations of pyridoline excretion with urine had apparently four phases: an increase, a decrease, stabilization and secondary increase. Distinct dissimilarity between men and women in the rate of pyridoline excretion was found only in the age group of 16-21 years old.

[The connection between the decomposition of mature collagen and the level of natural cytotoxicity in patients with focal scleroderma].

Increased rate of pyridinoline excretion with urine was detected in patients with focal sclerodermia: the ratio pyridinoline/creatinine was equal to 0.094 +/- 0.049 as compared with that of 0.073 +/- 0.049 in controls. Concentration of mature collagen cross-links involving pyridinoline structure in urine of patients with focal sclerodermia did not depend on the patients age and the disease duration. However, this pattern correlated distinctly with clinical form of the disease (0.073 +/- 0.049 in local form and 0.013 +/- 0.017 in generalized form of sclerodermia) as well as correlated reversely with the rate of activity of natural killer cells (r = -0.977).
