Detection of fos oncogene products by monoclonal antibody FO-120 in lymphoproliferative disorders.

We investigated the expression of fos oncogene proteins in lymphoproliferative disorders, using a monoclonal antibody (FO-120) that was prepared against a synthetic oligopeptide of fos protein (amino acid sequence from 127 to 152). Although peripheral blood leukocytes were rarely positive for FO-120, they were transiently stained after lectin (PHA) stimulation. After culture with IL-2 for 1 or 2 weeks, less than 40% of the lymphocytes weakly reacted with FO-120, whereas strongly positive cells were detected in more than 70% of cells in half the T-cell lines established from preleukemic state of adult T-cell leukemia (pre-ATL) and all of ATL derived T-cell lines. All in vivo specimens of non-Hodgkin's malignant lymphomas, except for one case of T-cell lymphoma were also strongly positive. In addition, the extent of the antibody reactivity correlated with the histopathological grade of malignancy in B-cell lymphoma. The reactivity to most AILD-IBL lesions overlapped with that to T-lymphomas, and could be distinguished from that to reactive lesions. FO-120 appears to be a useful tool for detecting early neoplastic changes in lymphoproliferative disorders.

Characterization of variant gamma-glutamyl transpeptidase produced by pancreatocholangiocarcinoma cell lines in a protein-free, chemically defined medium.

Four pancreatocholangiocarcinoma cell lines (HPC-Y1, HPC-YT, MIA PaCa-2, and HChol-Y1) were established to propagate in a protein-free, chemically defined medium. High gamma-glutamyl transpeptidase (GGTP) activities were showed in their spent media (designated as the secreted (GGTP). Their GGTP activities in the spent media were 125, 85, 110, and 153 IU/L/mg of lyophilized spent media, whereas GGTP activities extracted from their cancer cell lines with bromelain were 105, 37, 86, and 112 IU/L/1 x 10(6) cells, respectively. The chemical characteristics of the GGTPs in the spent media from these cell lines resembled one of the GGTPs, sialic acid-rich GGTP, extracted from normal human pancreas with bromelain treatment as follows: the GGTPs secreted from the cancer cell lines bound to an anion exchange column moved fast on electrophoresis and then showed decreased electrophoretic mobility with neuraminidase treatment, showed a high affinity for concanavalin A and lentil lectin columns, and had an acidic isoelectric point. However, the elution patterns of erythroagglutinating phytohemagglutinin (E-PHA) column chromatography and thermostability tests demonstrated clear differences between the carcinoma GGTPs both in the spent media and cell lines and the sialic acid-rich GGTP of normal pancreas, namely the carcinoma GGTPs treated with neuraminidase showed affinity to E-PHA columns, and, in addition, the GGTPs in the spent media showed an apparent heat resistance at 56 degrees C. These findings indicate that the carcinoma GGTPs have a different oligosaccharide structure from that in normal pancreatic GGTPs.

[Quantification of myocardial infarct size by technetium-99m pyrophosphate single photon emission computed tomography].

Myocardial infarct size in 41 patients with the first attack of acute transmural myocardial infarction (MI) was assessed by technetium-99m pyrophosphate single photon emission computed tomography (99mTcPYP-SPECT). A ratio of the number of voxels of 99mTcPYP uptake into the infarct area to that into the thorax was calculated as a parameter of MI size. The ratio was positively correlated with both peak CPK activity (r = 0.53, p less than 0.005, n = 24) and extent score in 201TI-SPECT (r = 0.70, p less than 0.005, n = 14) significantly in patients with anterior MI but not in patients with inferior MI. There was also significant negative correlation between the ratio and the left ventricular ejection fraction (LVEF) measured by RI angiography in both acute (r = -0.67, p less than 0.005, n = 18) and chronic (r = -0.75, p less than 0.005, n = 25) phases in patients with anterior MI. Recovery in LVEF at chronic phase was noted in patients with small anterior MI but not with large anterior MI. 8 of 14 patients with inferior MI had right ventricular MI, that might have affected evaluation of MI size and resulted in no correlation between variables. It was suggested that 99mTcPYP-SPECT was a useful method to evaluate MI size and to predict prognosis of cardiac function in patients with anterior MI but not in patients with inferior MI.

[PTCA and coronary artery bypass operation prior to subsequent surgery of gastric cancer in patients with coronary artery disease].

Two patients with coronary artery disease required operation for gastric cancer. In one patient with single vessel disease, 37 days later after successful PTCA, gastric resection for early gastric cancer was performed without any cardiac complication. The other patient with severe three vessel disease initially underwent coronary artery bypass operation, and 13 days later gastric resection for advanced cancer was performed. This patient developed supraventricular arrhythmia as a sole complication which was easily controlled by medical treatment. In both cases, fentanyl was used as an anesthetic agent and nitroglycerin was continuously administered intravenously during the perioperative period. ECG, central venous pressure, and direct arterial pressure were also carefully monitored. We feel that noncardiac operation such as gastric resection can be performed without major cardiac complications even shortly after coronary artery bypass operation.

Purification of human pancreatic gamma-glutamyltranspeptidase by lectin affinity chromatography.

Two types of pancreatic gamma-glutamyltranspeptidase (GGTP) (EC 2.3.2.2), sialic acid poor and sialic acid rich, were purified by the following: anion-exchange chromatography, wheat germ agglutinin (WGA)-Sepharose chromatography, gel filtration chromatography, phenyl-Superose chromatography, and hydroxylapatite chromatography. Among these, WGA-Sepharose chromatography helped to increase the specific activity of the GGTPs by approximately 20-30-fold in one effort. On dodecyl sulfate polyacrylamide gel electrophoresis, the two pancreatic GGTPs had different molecular weights. Sialic acid-rich GGTP had two subunits of Mr 67,000 and 27,000; however, the sialic acid-poor type had two subunits of Mr 72,000 and 29,000. The pI value of the sialic acid-poor GGTP was 5.9, and that of the sialic acid-rich GGTP 3.6.

Persistent and enhanced expression of c-fos gene products in various kinds of human hematopoietic cell lines. Immunofluorescence study using prepared monoclonal antibodies.

Two kinds of monoclonal antibodies recognizing fos proto-oncogene (c-fos) products were prepared using a synthetic oligopeptide corresponding to amino acids 127-152 of the fos oncogene products. These monoclonal antibodies (FO-120 & FO-145) detected fos gene products induced in a human monocyte cell line (U-937) by phorbol acetate (TPA) and induced in both human and mouse fibroblast cell lines (284, BALB/c 3T3) by serum-stimulation. One of the monoclonal antibodies (FO-120) reacted with 50-kDa and 42-kDa proteins and the other antibody (FO-145) reacted with a 30-33-kDa protein. The expression of the fos gene in various human hematopoietic cell lines was investigated using these prepared monoclonal antibodies. While almost all hematopoietic cell lines tested reacted with these monoclonal antibodies to various degrees, the majority of normal peripheral blood lymphocytes cultured with lectin (PHA) and interleukin 2 (IL-2) did not, suggesting that cells of some permanent hematopoietic cell lines, irrespective of their lineage specificity and growth factor dependency, continuously express the fos oncogene. These monoclonal antibodies may be useful for detecting early neoplastic changes in hematopoietic cells.

Decreased hindquarters venous distensibility during high salt intake in stroke-prone spontaneously hypertensive rats.

This study was done to examine whether high salt intake decreases venous distensibility in stroke-prone spontaneously hypertensive rats (SHR-sp). Ten weeks old male SHR-sp and normotensive Wistar-Kyoto rats (WKY) were fed either high (8%) or pressure-volume curves were obtained by infusing Krebs-Henseleit solution retrograde into the inferior vena cava at a rate of 12 or 2.1 ml/min. After 3 weeks of dietary treatment, high salt intake shifted the venous pressure-volume curves at maximal venodilatation caused by nitroprusside toward the pressure axis in SHR-sp but not in WKY. The venous pressure-volume curve during interruption of infusion was also shifted toward the pressure axis in SHR-sp on high salt diet as compared with that in SHR-sp on normal salt diet. Water content and thickness of the smooth muscle layer of the venous wall were not different between the two groups of SHR-sp. These results suggest that high salt intake for 3 weeks decreased hindquarters venous distensibility at maximal venodilatation in SHR-sp but not in WKY. The salt-induced decrease in venous distensibility in SHR-sp might be related to changes in interstitial space compliance or the adventitia since water content or thickness of the smooth muscle layer of the venous wall was not altered.

[Microscopic changes in the human liver after intra-arterial infusion of anticancer agents suspended in lipiodol].

Microscopic injury to the human liver after an intraarterial infusion of anti-cancer agents suspended in lipiodol has been evidenced by Hematoxylin-Eosin staining and by Oil Red O staining. In cases involving primary hepatic tumors, the liver tumor tissue has been injured severely while normal tissue showed no injury. In cases involving metastatic liver tumors, the tumor tissue, and also the normal liver tissue was injured though the latter injury was only mild. However, when multi-anticancer agents that were suspended in lipiodol were infused intra-arterially, the normal liver tissue was injured, even from a single drug suspension.

[Cytofluorometric and histopathological studies on non-cancerous liver lesions after lipiodol-transcatheter arterial embolization].

We have investigated the effects of preoperative Lipiodol-Transcatheter arterial embolization (Lp-TAE) on both the nuclear DNA content of noncancerous liver cells by DNA-cytofluorometry (using NIKON SPM-RF1-D), and the histopathological findings of liver lesions. Lp-TAE through the hepatic artery was performed on ten primary hepatocellular carcinoma patients using Gelfoam in combination with adriamycin (40 mg) or mitomycin C (20 mg), emulsified in Lipiodol (10 ml) and 60% Urografin (2 ml). And hepatic resection was carried out after one or two months later in each case. Histologically, primary hepatocellular carcinoma lesions showed various patterns of necrosis, while the non-cancerous liver cells did not show hepatocellular infarction in the liver cirrhosis. The results of the nuclear DNA content showed an increase in the fractions of poliploid cells in the non-cancerous lesions treated with Lp-TAE compared with that in the normal age-matched controls. But we found no remarkable differences in the ploidy patterns between Lp-TAE treated patients and untreated patients with liver cirrhosis. In conclusion, preoperative Lp-TAE does not induce any remarkable histological hepatocellular damage of non-cancerous lesion and does not affect nuclear DNA content of non-cancerous liver cells.

Augmented vascular responses to ouabain in SHRSP during salt loading.

It is not known whether high salt diet alters the sarcolemmal Na-K pump activity of resistance vessels in stroke-prone spontaneously hypertensive rats (SHRSP). We examined hindquarters vascular responses to ouabain 10(-3) M in the isolated hindquarters perfused with the Krebs-Henseleit solution in SHRSP and in normotensive Wistar-Kyoto rats (WKY) after two weeks of either normal (0.3% NaCl) or high (8% NaCl) salt diet. Salt loading increased arterial pressure (P less than 0.05) and augmented ouabain-induced vasoconstriction (P less than 0.01) in SHRSP. In WKY, salt loading did not alter arterial pressure or vascular responses to ouabain. Neither hindquarters vascular resistance during maximal vasodilatation produced by nitroprusside nor hindquarters vascular responses to norepinephrine were different between the two groups of SHRSP, which suggested that augmented response to ouabain in SHRSP on high salt diet could not be explained by structural vascular changes. Vasodilatation by phentolamine during maximal vasoconstriction induced by ouabain was also not different between two groups of SHRSP, which suggested that ouabain-induced release of norepinephrine from nerve endings was not different between the two groups of SHRSP. These results suggest that salt loading might augment the sarcolemmal Na-K pump activity of hindquarters resistance vessels in SHRSP but not in WKY. The augmented sarcolemmal Na-K pump activity of resistance vessels may act against salt-induced vasoconstriction in SHRSP.

Discrimination of gamma-glutamyltranspeptidase from normal and carcinomatous pancreas.

Two types of gamma-glutamyltranspeptidase (gamma-GTP) were identified in normal human pancreas, but ductal cell carcinomas of the pancreas showed only one type on analysis with polyacrylamide gel electrophoresis, anion exchange column chromatography, concanavalin A affinity chromatography and isoelectric focusing. gamma-GTP from carcinoma tissue was almost identical to the sialic acid-rich type of normal pancreatic gamma-GTP, and our methods did not evidence a difference between the two. However, E-PHA lectin column chromatography demonstrated differences in their elution pattern. Compared with normal pancreatic gamma-GTPs, the elution pattern of the carcinoma gamma-GTP was slowly delayed by the E-PHA lectin column. This finding suggests the presence of a bisecting N-acetylglucosamine-rich sugar chain in pancreatic carcinoma gamma-GTP, as well as the possible presence of a different sugar structure in the carcinoma gamma-GTP.

[The relationship between cell kinetics and histological features of giant cell tumor of the bone].

This study was undertaken to clarify the relationship between the proliferative activity and histological findings of the giant cell tumor (GCT) of bone by means of an epi-illumination cytofluorometer (NIKON SPM-RF1-D). Fresh tissues of GCT were surgically obtained from two cases. In both cases, small pieces of tumor tissues were obtained from several different regions based on the macroscopic characteristics of the cut surface, and processed for single cell preparation using enzymatic method. These isolated cells were smeared and stained with acridine orange, and then analyzed cytofluorometrically to determine simultaneously DNA and RNA contents of the individual cells. The results showed that the proliferative activity of tumor cells was much higher in the regions composed of both many histiocytic stromal cells having polygonal or ovoid shape and many multinucleated giant cells, than either in the regions composed of fibrocytic stromal cells accompanying abundant collagen fibers or in the regions composed of foamy cells.

[Properties of the rectosigmoid mucosa--histopathological studies on the autopsied colorectum].

Twenty cases of the entire course of the large intestine were obtained from autopsy materials within four hours after death and examined histologically. The autolytic changes were found to be minimal and the mucosal architecture was fairly well preserved in our studies. The morphometric analysis showed that the glandular lengths in the rectosigmoid colon were greater than those of the other parts of the large intestine. Cystic dilatations of the glands were most frequently observed in the distal colon, and tended to occur in the vicinity of neoplastic changes. These results indicate that the hyperplastic changes of the glands, occasionally with cystic dilatations, are important as the background for the development of neoplastic diseases in the large intestine.

[Cell kinetics and histological types of gastric cancer as studied by DNA-RNA cytofluorometry after acridine orange fluorescence staining].

In order to clarify the relation between histological classification and biological behavior of individual tumor cells, we quantitatively analyzed the cell kinetics of 56 advanced gastric cancers using DNA-RNA cytofluorometry after acridine orange fluorescence staining and compared the results with histological findings. The gastric cancers examined could be classified into two main groups based on the ploidy patterns constituting the cell population kinetics. In group I, tumor cells were found to be chiefly composed of diploid cells with many S-phase cells even at their advanced growth stages, and their morphological characteristics were shown to correspond to undifferentiated carcinoma (according to Nakamura & Sugano). In group II, tumor cells were composed of various classes of polyploid cells as well as of diploid cells with an increased number of S-phase cells, and the extent of polyploidization appeared to progress with the growth of the tumor. The morphological characteristics of this group tended to correspond to differentiated adenocarcinoma. It is, therefore, concluded that both the histological structures and cell morphology have close relationships with cell kinetics in the human gastric cancers.
