[TNF sensitivity of the established cell lines derived from esophageal squamous cell carcinomas and combined effect of TNF and IFN gamma].

Tumor necrosis factor (TNF) has been reported to have a high selective cytotoxicity to various kinds of tumor cells, but its effect on head and neck cancer is uncertain. To evaluate the effect of TNF on squamous cell carcinoma, we measured TNF sensitivity of six established cell lines derived from esophageal squamous cell carcinomas, TE-1, 2, 3, 4, 5 and 7 as well as L-M, FL, WISH, HEp-2 and THP-1 by means of 3H-thymidine incorporation method. HEp-2 derived from human laryngeal cancer, L-M, TE-3 and TE-4 were sensitive to TNF while THP-1, TE-1, 2, 5 and 7 were not. IFN gamma is known to be an inducer of TNF receptor of some kinds of tumor cells. TE-2 and TE-3 were sensitive to IFN gamma. Combined use of these two cytokines resulted in additive inhibitory effect on TE-2 and 3, but no remarkable synergistic effect on any of TE-1, 2, 3 or 4.

[Evaluation of the efficacy of ceftriaxone in acute suppurative otitis media and acute exacerbation of chronic suppurative otitis media. A comparative study with cefotiam as the control].

In order to objectively evaluate the efficacy and the safety of ceftriaxone (CTRX) using once daily administration of 1 g to cases of acute suppurative otitis media and acute exacerbation of chronic suppurative otitis media, a group comparison study by the envelope method was conducted using cefotiam (CTM) as the control drug (2 g twice daily). The results obtained are summarized as follows. 1. Clinical efficacies evaluated by the committee were 71% in the CTRX group and 86% in the CTM group for acute suppurative otitis media, and 63% and 60%, respectively, for chronic suppurative otitis media. When all cases were considered both groups evidenced a clinical efficacy of 64%, and no significant difference was observed between the 2 groups. 2. Clinical efficacies evaluated by the physician in charge were 65% in the CTRX group and 86% in the CTM group for acute suppurative otitis media, and 72% and 60%, respectively, for chronic suppurative otitis media. When all cases were considered efficacies were, respectively, 70% and 64%, showing no significant difference between the 2 groups. 3. Bacteriological efficacies were 88% in the CTRX group and 86% in the CTM group for acute suppurative otitis media, and 74% and 62%, respectively, for chronic suppurative otitis media. With all cases bacterial eradication rates were, respectively, 76% and 67%. Bacterial eradication rates were always higher for the CTRX group than for the CTM group, but the difference was not significant between the 2 groups. 4. Against infections caused by Staphylococcus aureus alone, CTRX showed equal clinical and bacteriological efficacies to CTM. 5. As side effects, dermatitis, vomiting, and malaise were observed in 5 cases (4%) of the CTRX group and 3 cases (3%) of the CTM group. As clinical testing abnormalities, elevations of GOT, GPT, and Al-P, and thrombocytopenia were noted only in 3 cases (5%) of the CTRX group. Furthermore, all of these abnormalities were temporary and of moderate degree or mild, thus the safety of either drug was considered high. 6. Clinical utilities were 71% in the CTRX group and 86% in the CTM group for acute suppurative otitis media, and 72% and 62%, respectively, for chronic suppurative otitis media. When all cases were included, they were 72% and 66%, respectively, and there was no significant difference between the 2 groups. It is concluded from the above results that CTRX is a highly useful drug with once daily administration of 1 g in the treatment of suppurative otitis media.

Correlation between morbid adenoid and atelectatic ear.

We observed the nasopharynx of 30 patients with atelectatic ears (AE), and 10 with healthy ear drums and mouth breathing by CT scan, to examine the relation between adenoid and otitis media. The adenoids of those with healthy ear drums were of the large posterior type with free space in the pharyngeal fossa, while those with AE were of the large anterior type in 18 patients and of the pendulous projection type over the choana in the other 12. The anatomical location of the adenoids should be considered when one discusses the relation between the adenoid and otitis media. The "Toynbee phenomenon", in the pendulous projection type and in the large anterior type of adenoid, was considered to be one of the etiological factors in AE. To understand the etiology of otitis media, it seems important to investigate the reason why the anterior projection of the adenoids occurs.

Atelectatic ear and pathologic nasopharynx.

Pathologic relation between atelectatic ears and their nasopharynx were investigated in 50 patients with various degrees of atelectatic ears by transnasal fibrescope and C.T. And their morbid adenoids were compared with the adenoids of 20 patients with mouth breathing but free from ear problems. The process of drum bulging after catheterization, retraction after inflation, and inward movement during swallowing, proved that intratympanic pressure was influenced by nasopharyngeal pressure produced by swallowing, and that Eustachian tube itself was functioning to the pressure. Intratympanic negative pressure was proved by the movement of blue solution after the puncture of 5 atelectatic ear with effusion. And more dynamic negative pressure produced by swallowing was demonstrated in 5 other cases. The morbid adenoid with atelectatic eardrum was differentiated from the large posterior type of adenoid of the healthy eardrum, by the pendulous projection over the choana especially in swallowing. A dynamic vacuum caused by "Toynbee phenomenon" is discussed.

A morphological study of the vestibular sensory epithelia in the rabbit.

Surface preparations were made of the vestibular sensory end-organs of the rabbit. The total sensory cell counts and surface area measurements were made from the composite photographic reconstructions. The general topographical and cytoarchitectural arrangements were found to be similar to those previously described for guinea pigs, squirrel monkeys and humans. The sensory cell counts of the maculae are about 50% larger in the rabbit than in the guinea pig, while those of the cristae remained almost unchanged in both species of animals despite a large difference in both size and weight. The body size of the species does not seem to play a decisive role in determining the sensory cell population of the vestibular sensory end-organs. The growth of the maculae is compared with that of the cristae or vice versa seems to vary from one species to another, probably depending upon a physiological need of the species.

A case of chronic lymphocytic leukemia with properties characteristic of natural killer cells.

A case of chronic lymphocytic leukemia that consisted of a homogeneous population of cells that had properties similar to those described for natural killer (NK) cells is presented. These leukemic cells had a morphology of large granular lymphocytes (LGL) and receptors for sheep erythrocytes (ER) and for the Fc portion of IgG (Fc gamma-R). They expressed pan-T antigens OKT3 and Leu-4, but neither helper/inducer T-cell differentiation antigens OKT4 and Leu-3a nor cytotoxic/suppressor T-antigens OKT8 and Leu-2a. HNK 1 antigen, which can be expressed on human NK cells, could be detected on almost all leukemic cells (LGL), whereas a myeloid differentiation antigen, OKM1, which can be expressed on macrophages, granulocytes, and NK cells, was not detected. Thus, it was concluded that the leukemia cells had a characteristic profile of the surface markers: ER+, Fc gamma-R+, HNK-1+, OKT3+, Leu-4+, OKT4-, OKT8-, Leu-3a, Leu-2a, and OKM1-. Although freshly isolated leukemic cells showed no cytotoxicity on NK targets, after incubation at 37 degrees C, the cells did show a potent cytotoxicity on targets of erythroleukemic cell, T cell, and monocyte (but not B cell) origins. When the cells were incubated at 37 degrees C, interferon (IFN gamma) was spontaneously produced in the culture fluids. Treatment with anti-HNK-1 and complement completely abrogated expression of NK activity and interferon production of the patient's lymphocytes in culture. These characteristic features of surface markers and functions strongly suggest the possibility that the leukemia cells of this case are of NK cell origin. The relationship between this case and chronic lymphocytic leukemia of T-cell origin is discussed.
