The fluoroquinolones.

At the Eastern Section Meeting of the Triological Society January 26, 1990, Levinson et al reported outstanding success with ciprofloxacin in the treatment of malignant otitis externa. Moreover, several of the individuals so managed had been refractory to conventional therapy with semisynthetic penicillin and aminoglycoside protocols. This new class of antibiotics may result in a profound change in our management of patients with otologic, neurotologic, and skull base infections.

Association of folic acid with rat liver microsomes.

Shin et al. (Biochim Biophys Acta 444: 794-801, 1976) described the subcellular location of [3H]folic acid after injection into rats. The microsomal fraction of the liver contained relatively large amounts of tracer initially but lower amounts at later times. Because of the heterogeneous nature of the microsomal fraction of the liver we re-examined the nature of the folate binding fraction. The location of injected [3H]folic acid resembled that of the microsomes derived from the plasma membrane, where ultracentrifugal analysis was conducted in the presence and absence of cesium ions. The location of the folate did not resemble that of microsomes derived from the endoplasmic reticulum (ER). One of the marker enzymes of the ER was the vitamin K-dependent carboxylase. A simple method for reducing vitamin K is described.

Nitrous oxide provokes changes in folylpenta- and hexaglutamates.

Liver folates isolated from rats fed a control diet consisted of H4PteGlu5 (44% of total) and 5-methyl-H4PteGlu5 (19%) with smaller amounts of 10-formyl-H4PteGlu5 (13%), H4PteGlu6 (13%), 10-formyl-H4PteGlu6 (4%) and 5-methyl-H4PteGlu6 (2%). Folates from methionine-deficient rats contained more nearly equal amounts, on a molar basis, of folylpenta- and hexaglutamates, and hence these were more suitable subjects for comparing the metabolism of these compounds in vivo. Livers from methionine-deficient rats contained H4PteGlu5 (30%), 5-methyl-H4PteGlu5 (13%), 10-formyl-H4PteGlu5 (10%), H4PteGlu6 (22%), 5-methyl-H4PteGlu6 (9%) and 10-formyl-H4PteGlu6 (11%). With exposure to N2O, 5-methyl-H4PteGlu5 and 6 increased to 27% and 13%, respectively, whereas H4PteGlu5 and 6 decreased to 20% and 17%, respectively. Nitrous oxide (N2O) perturbed both the penta- and hexaglutamates; the effect was somewhat more pronounced with the pentaglutamates. The partial depletion in tissue H4PteGlun with N2O treatment helps explain the concomitant inhibition of the oxidation of [ring-14C]histidine, an event dependent on tetrahydrofolates.

Subcellular localization of gamma-glutamyl carboxypeptidase and of folates.

The subcellular distributions of glutamyl carboxypeptidase, folate specific activities, and radioactive metabolites of injected [3H] folic acid were studied in rat liver. The specific activity of glutamyl carboxypeptidase in the lysosomal fraction was near or greater than four times that in the other subcellular fractions. The specific activity of folates was highest in the soluble fraction (102 ng folate/mg protein) and lowest in the microsomal fraction (22 ng folate/mg protein). Nuclear, mitochondrial, and lysosomal folates were 95% folate polyglutamates, and microsomal and soluble folates were 85--90% folate polyglutamates. Injected [3H] folic acid was initially concentrated in the microsomal fraction, as measured by 3h cpm per ng folate. Initially, injected [3H] folic acid was found converted to folate penta- and hexaglutamates in all fractions to a similar extent except in the microsomes where the percentage conversion was much less, as measured by the percentage of total 3H cpm determined to be [3H] folate penta- and hexaglutamates. At 24 h, the conversion of [3H] folates to penta- and hexaglutamates in each fraction was less than that found for the endogenous folates. Injected [3H] folic acid after 2h was found to consist of 94% reduced folates in the soluble fraction, 56% in the mitochondrial, 55% in the nuclear, 20% in the lysosomal, and 15% in the microsomal fraction.

Familial differences in antibody response of broiler chickens to vaccination with attenuated and inactivated Newcastle disease virus vaccine.

Genetic differences in immune response to Newcastle disease virus (NDV) were studied in 4-week-old broilers, vaccinated with attenuated (live) or inactivated NDV. The experiment included 370 chicks from two farms distributed among 22 sire families and 60 dam families. Results in chicks from both farms were similar. Survival after challenge was closely related to titer level. The genetic correlation between day-7 and day-12 titers (attenuated virus) was 1.0. Significant differences were found between sire families in both sorts of vaccinations. Heritabilities based on the sire variance components for attenuated and inactivated virus vaccinations were respectively 0.31 and 0.60. The genetic correlation between them was 0.49. Nevertheless, it is concluded that selection for response to NDV based on inactivated virus may be most effective in improving response to attenuated NDV vaccinations.
