A Walker 256 tumor-induced osteogenic small animal model for the evaluation of [99mTc] diphosphonate radiopharmaceuticals.

A mammalian model has been developed for the in vivo evaluation of bone imaging agents. The model is based upon the quantification of a discrete, initial secondary periosteal osteogenesis induced in cortical bone immediately adjacent to an intramuscularly implanted Walker 256 tumor in Fisher 344 rats. Evaluation of the model consists of a histopathological examination of the periosteal bone formation, biodistribution studies on 99mTc-MDP and 99mTc-HMDP commercial kit preparations, and biodistribution studies on two 99mTc-HEDP component fractions isolated after anion exchange chromatographic separations from an investigative "carrier added" preparation. Reversed phase HPLC separations of the 99mTc-MDP and 99mTc-HMDP commercial kit preparations illustrate distinct differences in chemical composition between the two bone agents.

Inhibition of T7 bacteriophage replication by a colicin Ib plasmid gene.

Clones containing fragments of the colicin Ib (ColIb) plasmid inserted into pBR322 have been found that inhibit the replication of T7 bacteriophage. Cells containing the whole ColIb plasmid grow T7 normally but cannot grow T7 protein kinase-negative mutants of T7. The cloned fragments inhibit not only the T7 protein kinaseless mutants but wild-type T7 as well. However, the whole plasmid can suppress the wild-type T7 inhibition caused by the cloned inhibiting genes. These results are consistent with a model in which a ColIb gene (pic) exists which can inhibit replication of T7 phage. A second gene (rpi) can repress the function of pic provided the rpi product is phosphorylated.

In vivo distributions of some component fractions of Tc(NaBH4)-HEDP mixtures separated by anion exchange high performance liquid chromatography.

Anion exchange high performance liquid chromatography has been utilized to separate the component Tc-HEDP complexes of "carrier added" as well as "no carrier added" 99mTc(NaBH4)-HEDP reaction mixtures. The chromatographically separated "carrier added" Tc-HEDP components exhibit distinctly different biodistributions which are directly related to the chromatographic characteristics of the individual components. The Tc-HEDP complexes with shortest retention times, and therefore the lowest negative "charge density" have the maximum uptake on the bone, minimum uptake in the kidney, and fastest blood clearance.

Analytical problems facing the development of electrochemical transducers for in vivo drug monitoring: an overview.

Even though significant progress has been made in the development of electrodes for the in vivo detection of catecholamines, oxygen, and various cations, there has been little research on the feasibility of continuously monitoring drugs in whole blood by electrochemical sensing devices. Electroanalytical problems associated with the development of such electrochemical transducers include the need for increased sensitivity and specificity, decreased biological matrix interferences, more rapid response times, improved miniaturization, and more reliable calibration procedures. We present a critical evaluation of potentiometric, conductometric, and amperometric techniques, with a brief review of basic principles and recent advances in ion-selective electrodes, chemical-sensing field-effect transistors, amperometric enzyme electrodes, and electrochemical immunosensors.
