Hematological changes produced in mice by ochratoxin A and citrinin.

The effect of 6 weekly injections of ochratoxin A (5 mg/kg) and citrinin (20 mg/kg) on the following hematological indices of mice was studied: erythrocyte sedimentation rate, hematocrit percentage, mean corpuscular values, red cell indices, platelet count, total count of bone marrow cells, differential count of bone marrow cells, total count of splenic cells, spleen weight, and serum calcium. These mycotoxins resulted in a significant decrease in platelet count and hematocrit values. On the other hand, they increased erythrocyte sedimentation rate, mean corpuscular values and red cell indices. Weight of the spleen and the splenic cell count decreased significantly in ochratoxin A and citrinin treated mice. Total count of bone marrow cells, erythrocyte precursors, leucocyte precursors and megakaryocyte precursors of femoral bone marrow decreased significantly in toxin treated mice. Calcium concentration in the serum of toxin treated mice also was decreased significantly.

Hematological changes produced in mice by Nuvacron or Furadan.

The effects of multiple intraperitoneal doses of Nuvacron (0.8 mg/kg) or Furadan (0.25 mg/kg) on hematological values in mice were studied. The following were measured: clotting time, hemoglobin content, total count of red blood cells and white blood cells, differential count of white blood cells, platelet count, erythrocyte sedimentation rate, hematocrit value, mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration, bone marrow and splenic measurements. These pesticides caused a significant decrease in hemoglobin content, total count of red blood cells, platelet count, erythrocyte sedimentation rate and hematocrit value. The administration of Nuvacron or Furadan prolonged the clotting time and caused an increase in total count of white blood cells. Among white blood cells, there was an increase in neutrophils and basophils and a fall in the count of lymphocytes in treated animals. Bone marrow depression and splenic hyperplasia were observed in mice after administration of the pesticides.

Mouse (C57BL/KsJ) liver phosphofructokinase. Allosteric kinetics and age-related changes in the genetically diabetic state.

The regulatory kinetic properties of phosphofructokinase partially purified from the livers of C57BL/KsJ mice were studied. The fructose 6-phosphate saturation curves were highly pH dependent. At a fixed MgATP concentration (1 mM), allosteric kinetics was observed in the range of pH studied (7.3 to 8.3) and the S0.5 values for fructose 6-phosphate decreased by about 0.2 to 0.3 mM for each 0.1-unit increment in pH. Allosteric effects on the sigmoidal response to fructose 6-phosphate: activation by AMP, NH4+, and glucose 1,6-bisphosphate, inhibition by MgATP2-, and synergistic inhibition between ATP and citrate, were all present at pH 8.0 to 8.2. Comparative kinetic studies with liver phosphofructokinase isolated from both the normal (C57BL/KsJ) and the genetically diabetic (C57BL/KsJ-db) mice of 9 to 10 and 15 to 16 weeks of age showed that the enzyme from the livers of diabetic mice exhibited decreased activity at subsaturating concentrations of fructose 6-phosphate. However, phosphofructokinase isolated from the livers of normal and genetically diabetic mice of 4 to 5 weeks of age showed no difference in kinetic properties. Thus, there appears to be a correlation between the change in properties of liver phosphofructokinase and the expression of hyperglycemia and obesity in the genetically diabetic mice. The decreased activity of liver phosphofructokinase in the older diabetic animals may well be one of the causes of the increased blood glucose levels. The results are also discussed in a general context with regard to the possible role of phosphofructokinase in the regulation of hepatic gluconeogenesis.

Increased ATP inhibition of liver phosphofructokinase from genetically diabetic mice.

Phosphofructokinase (ATP:D-fructose-6-phosphate 1-phosphotransferase, EC 2.7.1.11) was partially purified from the livers of genetically diabetic mice (C57BL/KsJ-db) and their lean littermates (C57BL/KsJ). These genetically diabetic mice have been shown to be hyperglucagonemic and to exhibit symptoms resembling those of maturity-onset diabetes in humans. Two isoenzymes of phosphofructokinase were obtained after DEAE-Sephadex chromatography of extracts of livers from either normal or diabetic animals. One of these isozymes, peak II, from the genetically diabetic mice was shown to be more sensitive to ATP inhibition at physiological pH than the peak II isozyme from the normal animals. In addition, the peak II isozyme from the diabetic mice exhibited decreased affinity for fructose 6-phosphate. The altered kinetic properties of phosphofructokinase from diabetic animals are markedly similar to those recently reported for liver phosphofructokinase isolated from normal animals after glucagon treatment. Our results suggest that increased glucagon levels in diabetes may lead to altered regulation of phosphofructokinase in this disease.

Studies on plasma fibrinogen level in pre-eclampsia and eclampsia.

The plasma fibrinogen level of maternal blood has been estimated in 30 cases of pre-eclampsia, 60 cases of eclampsia and 35 cases of normal pregnancy of 3rd trimester. The plasma fibrinogen value increased by about 70% and 145% in pre-eclampsia and eclampsia, respectively. In essential hypertension, the fibrinogen level remains more or less the same as in normal pregnancy.

Proximal coronary artery stenosis: three-dimensional MRI with fat saturation and navigator echo.

The purpose of this study was to compare the diagnostic value of MR angiography (MRA) with conventional contrast angiography in coronary artery disease. Thirty-five patients underwent MRA and coronary angiography within 4 hours. Of these, three patients were investigated twice: once before and once after balloon angioplasty. The pulse sequence was a cardiac-triggered, single-slab, three-dimensional gradient-echo sequence, employing a spin-echo navigator echo measurement to track the variation of the diaphragm during the scan. The following segments of the coronary arteries were included in this prospective study: left main coronary artery, proximal and middle left anterior descending, proximal and middle left circumflex, proximal and middle right coronary artery, and intermediate branch, if present. In total, 176 segments were classified as normal or having a stenosis of less than 50% and as having a stenosis of more than 50%. Five patients were excluded because of lack of cooperation. Over all, 45 of 54 stenoses were detected and interpretable by MRA. Sensitivity, specificity, and positive and negative predictive values of MRA for detecting significant stenoses were 83%, 94%, 87%, and 93%, respectively. MRA identified significant stenoses within the major coronary arteries with a high degree of accuracy. Sensitivity and specificity are higher compared with exercise tests or scintigraphy or top of the precise localization.

Nutritional status of pavement dweller children of Calcutta City.

Pavement dwelling is likely to aggravate malnutrition among its residents due to extreme poverty, lack of dwelling and access to food and their exposure to polluted environment. Paucity of information about nutritional status of street children compared to that among urban slum dwellers, squatters or rural/tribal population is quite evident. The present study revealed the magnitude of Protein Energy Malnutrition (PEM) and few associated factors among a sample of 435 underfives belonging to pavement dweller families and selected randomly from clusters of such families, from each of the five geographical sectors of Calcutta city. Overall prevalence of PEM was found almost similar (about 70%) to that among other 'urban poor' children viz. slum dwellers etc., but about 16% of them were found severely undernourished (Grade III & V of IAP classification of PEM). About 35% and 70% of street dweller children had wasting and stunting respectively. Severe PEM (Grade III & IV) was more prevalent among 12-23 months old, girl child, those belonged to illiterate parents and housewife mothers rather than wage earners. It also did increase with increase of birth rate of decrease of birth interval.
