The neutrophil and lymphocyte nonspecific esterase in patients with malignancies.

In 52 patients with malignancies such as lung cancer, carcinoma of the stomach, and cancer of the colon (25, 18, and 9 cases, respectively) activity and intracellular localization of alpha-naphthyl acetate esterase has been evaluated by means of semiquantitative cytochemical method. Patients with advanced metastatic process occurring during clinical evolution of those malignancies showed decreased activity of above enzyme within the peripheral blood neutrophils and decreased numbers of enzyme-positive lymphocytes having intact enzyme-positive lysosomal granules. The authors discuss the significance of that observation in relation to the enzymatic alterations of cells during antitumor response of immune system.

Output of peritoneal cells into peritoneal dialysate. Cytochemical and functional studies.

The quantitative and qualitative characteristics of cells in the peritoneal dialysate from 12 patients were examined. The number of cells in each subsequent fraction of dialysate decreased, while the differential cell count remained relatively constant for each individual patient. Macrophages, lymphocytes, granulocytes and occasionally mesothelial cells were observed. In 1 patient, plasmocytes were also found. Evident differences in cellularity and cell composition were noticed in dialysate obtained from different patients, especially in 2 patients with bacterial peritonitis there was a rise in cellularity with neutrophilia. Cytochemical (peroxidase, nonspecific esterase activity) and functional (phagocytosis, receptor expression) tests revealed that macrophages form a heterogeneous population of cells.

Dialysis for psoriasis - preliminary remarks concerning mode of action.

In one patient treated by peritoneal dialysis for psoriasis the skin lesions cleared completely. In two persons hemodialysis gave unsatisfactory results. In treated and another three untreated patients cellular immunity was evidently suppressed. This phenomenon is similar to the immunologic changes in persons with uremia. Solutes in middle molecular weight range (SMMWR) suppress cellular immunity. These compounds were therefore investigated in psoriatics. The level of SMMWR was lower before than after each dialysis, although their concentration increased slightly during the dialytic treatment. This suggests that solutes in middle molecular weight are fixed in the epidermis and/or in the walls of the skin capillaries and that dialysis liberates them from these places. Peritoneal dialysis seems to be a more effective method of treating psoriasis than hemodialysis. Solutes in middle molecular weight range might play an important role in the pathomechanisms of this multifactorial disease.

Influence of middle-molecular-weight solutes from dialysate on the migration rate of leukocytes.

To investigate further the biologic activity of solutes in the middle-molecular-weight range, we studied the influence of these compounds on the migration of unseparated white blood cells and separated granulocytes and lymphocytes. Middle molecules (MM) inhibit the migration of unseparated leukocytes, but this effect on lymphocytes was seldom observed. With scanning electron microscopy, unseparated leukocytes were shown to adhere to one another, forming cellular clumps. This phenomenon could not be seen when separated cells were used. These results give some insight into the mechanism of inhibition exerted by MM and into the changes, observed in earlier experiments, of cellular composition of skin exudate obtained from untreated patients with uremia.

The microscopical structure and quantitative changes of pituitary cell populations in frogs (Rana esculenta L.) irradiated by ultraviolet light.

The percentage of cell populations of hypophysis pars distalis, their ultrastructure, degranulation phenomena and volume of cell nuclei easured by karyometry was examined in frogs, Rana esculenta L. irradiated by UV light. The observed changes, reflecting alterations of the secretory activity were most prominent in B2 cells producing gonadotropic hormones and in B3 cells probably responsible for the secretion of ACTH. A concomitant increase of the number of chromophobic cells was observed. The obtained results are discussed in relation to data from the existing literature.

Effect of the new theophylline derivatives on degranulation of mast cells and phosphodiesterase activity in smooth muscles.

Effect of the new theophylline derivative (beta-hydroxy-benzylopiperazinopropyl-theophylline), designed as R6 on degranulation of the mast cells from rat mesenteries, was tested in vitro and in vivo. In vitro it was found that R6 significantly inhibited mast cell degranulation induced both by the histamine liberator C 48/80 and by rabbit globulin directed against rat serum proteins. The effect was comparable to that revealed by mepyramine, but much stronger than the effects of DSCG and aminophylline. The influence of R6 on mast cells degranulation was less pronounced in the in vivo systems. It was demonstrated by histochemical methods that R6 did not change activity of lactic and succinic dehydrogenases, neither activity of lysosomal hydrolases. In separate system including small intestine from rat it was shown that R6 inhibited activity of the phosphodeisterase in smooth muscles. It was suggested that the effect of R6 on mast cells degranulation is mediated by an increase in cAMP as well as by indirect influence on Ca++ influx.

Some aspects of cellular immunity in untreated and maintenance hemodialysis patients.

The cellular pattern of skin exudate in untreated uremic patients differed from that in healthy persons. The skin exudate composition in patients on maintenance hemodialysis approached normal values. Such normalization was, however, static: after sensitization with dinitrochlorobenzene--contrary to the control--both treated and untreated patients exhibited no changes in the cellular pattern of exudate. The activity of acid phosphatase in the cells of exudate was highly positive in control, almost normal in dialyzed, but weak in untreated uremic patients. Our investigations confirmed that cellular immunity in untreated and in maintenance hemodialysis patients is suppressed.

The enzymatic activities and NBT reduction test of granulocytes in untreated and dialysed uraemic patients.

It was found that neutrophils in untreated uraemic patients as well as in subjects on regular dialysis treatment displayed higher activity of acid phosphatase, alkaline phosphatase and peroxidase. Spontaneous reduction of nitro blue tetrazolium (NBT) by granulocytes was also higher in both groups in comparison to controls. Stimulation with latex particles gave similar results of NBT reduction in investigated patients and controls. Lymphocytes also showed an increase in acid phosphatase activity if compared to healthy persons. It seems possible that granulocytes which take part in unspecific defense mechanisms are more active in uraemic patients due perhaps to subclinical infections.

Modification of the activity and surface topography of peritoneal macrophages by cytophilic antibody.

Cytophilic antibody (Cab) attached to the macrophage surface substantially changes the cell surface relief as tested in the scanning electron microscope. Diffuse loss of surface structure is accompanied by diminished ability to interiorize opsonized sheep erythrocytes and to damage non-specifically bystanding target cells (chicken erythrocytes). Cross-linking of Cab on the macrophage surface by antigen or antibody partially restores macrophage membrane activity.

Cytoenzymatic studies on the lymphocytes of peripheral blood and lymphatic nodes of rats in an experimental magnesium deficiency.

Wistar rats kept on a magnesium deficient diet show several changes in the lymphatic organs as well as some disorders in the function of the immunological system, which appear as an impairment of cellular immunity and also as hypogammaglobulinemia. In the present experiment the level of leuco- and lymphocytosis has been studied. Furthermore, the activity of some lysosomal enzymes in blood lymphocytes, as well as the ability to incorporate labelled leucine shown by lymph nodes lymphocytes of deficient rats have been investigated. The rise in leuco- and lymphocytosis is similar to that reported by other authors. A significant rise in the activity of beta-glucuronidase as well as a considerable drop in the percentage of enzyme-negative lymphocytes have been observed as early as the first week of experiment. Tissue cultures of the lymph nodes, lymphocytes of deficient rats showed significantly lower values of labelled leucine incorporation with respect to the controls; in contrast, after phytohemagglutinin M stimulation the increase of incorporation in the lymphocytes of deficient and control rats was similar. Our findings may be indicative of some disorders in the redistribution of T and B lymphocytes in the blood and tissues of deficient rats. The observed cytochemical changes may be due to the intensification of autophagic processes in the lymphocytes that manifest a diminished ability to synthetize proteins.
