Synthesis and crystal structure of a novel germanate: (NH4)4[(GeO2)3(GeO1.5F3)2].0.67H2O.

The novel microporous germanate (NH4)4[(GeO2)3(GeO1.5F3)2].0.67H2O was prepared from an aqueous solution containing germanium dioxide, pyridine, hydrofluoric acid, and 2,6-diaminopyridine as a template. The solution was kept at 165 degrees C in a Teflon-lined autoclave for 4 days. Large crystals were produced and studied by X-ray powder diffraction, FTIR, thermal analysis, and elemental analysis. The structure was determined by single-crystal X-ray diffraction. The crystal is orthorhombic, space group Pbcn, with a = 7.0065(4) A, b = 11.7976(6) A, c = 19.5200(14) A, and Z = 4. The structure is a layered framework built up from GeO4 tetrahedral and GeO3F3 octahedral units. The polyhedral units are connected in such a way that they form a zeolite-like porous structure with three- and nine-membered rings. Half of the ammonium ions are located inside the nine-membered rings. The other half are above and below the three-membered rings. The connectivity of the germanium polyhedral units is interrupted along the c axis by ammonium ions and water molecules inserted between the layers.

A three-dimensional open-framework germanate containing four-, five- and six-coordinated germanium

A new three-dimensional open-framework germanate, namely ethylenediamine bis(ethylenediammonium) tetrahydroxooctadecaoxononagermanate, (C(2)H(8)N(2))(C(2)H(10)N(2))(2)[Ge(9)O(18)(OH)(4)], has been synthesized hydrothermally and its structure determined by single-crystal X-ray diffraction. The framework is built of [Ge(9)O(22)(OH)(4)] units formed by four-, five- and six-oxygen-coordinated germanium and templated by ethylenediamine. Three types of intersecting channels are formed in the framework, one by eight-membered rings running along the b axis and the other two by ten-membered rings running parallel to the a and c axes, respectively.

A role for adenosine in coronary vasoregulation in man. Effects of theophylline and enprofylline.

Adenosine has been suggested to have a role in regulation of the tone of the cardiac resistance vessels. To elucidate the coronary vasoregulatory role of endogenous adenosine in man, we studied the effects of adenosine receptor antagonism by theophylline on coronary blood flow at rest and during light exercise. However, theophylline may also exert pharmacological effects not related to adenosine antagonism. To clarify the contribution of endogenous adenosine in coronary hyperaemia, the effect of theophylline was compared to that of enprofylline, a xanthine which exerts similar pharmacological effects as theophylline while lacking antagonistic action at adenosine receptors. Twenty healthy subjects (10 males) aged 22-39 years were examined. Coronary sinus (CS) blood flow and blood oxygen content were determined at rest and during supine bicycle exercise, at a load of 50 watts, for 10 min. Thereafter, stepwise infusion of adenosine (30 to 60 micrograms/kg/min into the subclavian vein) was performed. Theophylline or enprofylline treatment was instituted randomly and double-blind (10 in each group), and the procedures (i.e. determinations at rest, during exercise and during infusion of adenosine) were repeated. In all 20 subjects, basal CS flow was 70 +/- 6 ml/min and the cardiac oxygen extraction ((A-CS)O2D) was 123 +/- 3 ml/l. During exercise, CS flow and (A-CS)O2D increased to 135 +/- 17 ml/min and 132 +/- 3 ml/l, respectively. Adenosine increased CS flow dose dependently to 161 +/- 27 ml/min, while (A-CS)O2D decreased to 66 +/- 7 ml/l. The vasodilatory effect of adenosine was readily counteracted by theophylline, the increase in CS flow being 33% vs. 133% in the control situation. Enprofylline, on the other hand, enhanced the response to exogenous adenosine. Theophylline, at a dose lacking effect on heart rate and blood pressure, decreased CS flow at rest by 14% (P < 0.05) and during exercise by 18% (P < 0.05). ((A-CS)O2D increased by 14% at rest and during exercise (P < 0.001). Enprofylline, on the other hand, was without effect. The differences in responses between theophylline and enprofylline with respect to coronary flow and oxygen extraction were significant both at rest and during exercise. It is concluded that theophylline increases coronary vascular resistance, while enprofylline, lacking adenosine antagonistic properties, was without such effect. This indicates a physiological role of adenosine in regulation of coronary flow.

A long-term follow-up of patients with essential hypertension treated with captopril.

Seventy-four patients from four short-term studies of captopril in mild-moderate essential hypertension continued in a cooperative long-term efficacy and tolerance program. The duration of observation is 2- greater than 4 years, the total treatment time being 2434 months. No development of resistance to therapy was observed. The total daily dose of captopril has been gradually decreased and in 20 patients changed from t.i.d. to b.i.d. regime. The drug has been well tolerated and only few and mild side-effects have been observed after the initial titration period. The drop-outs (n = 19) were mostly due to non-medical causes (n = 14). Except for one case of proteinuria, no laboratory abnormalities were detected and there were no signs of long-term toxicity.
